Abstract. Descriptions of the developmental stages of Sarcophaga (Liopygia) argyrostoma (R.-D.) are given. Scanning electron microscope images of most of its immature stages are presented for the first time. The sequence of mitochondrial cytochrome c oxidase subunit I (COI) gene fulfilling DNA barcoding standards is presented for the first time.

Cytochrome b gene (cytb) sequence diversity in a Microtus oeconomus population from Bialowieza Primeval Forest

Based on published information about the glacial, postglacial, and recent distribution of the root vole, Microtus oeconomus, we hypothesized that a population inhabiting the pristine wetland in eastern Poland (Bialowieza Primeval Forest) might comprise a high diversity of haplotypes. The support for this hypothesis was provided by an analysis of partial cytb gene sequences from 149 voles sampled within a two-hectare plot during a nine-year study. In this population, we identified eight haplotypes (PLB1-PLB8), four of which were new to the root vole. These haplotypes were characterized by low nucleotide diversity (π = 0.0054, SE = 0.0019), the absence of transversional differences between sequences, and no changes in the encoded amino acid sequence: features suggesting a lack of immigration from the distant populations. The haplotype number and their frequency distribution in males and females did not differ significantly. An assessment of the persistence of matrilines in the local population throughout the study period revealed that the haplotype composition was relatively stable for only about 3 years. A more complete haplotype network for root voles in Europe was constructed by combining the newly identified haplotypes with the 45 previously described haplotypes. Two of the haplotypes detected in this study occupy key positions in this network: PLB5, as the closest link to the North European group, and PLB8, as an ancestor to many other Central European haplotypes.

First microsatellite markers for the European Robin (Erithacus rubecula) and their application in analysis of parentage and genetic diversity

The European Robin is a small passerine bird associated with woodlands of Eurasia and North Africa. Despite being relatively widespread and common, little is known of the species' breeding biology and genetic diversity. We used Next Generation Sequencing (NGS) to develop and characterize microsatellite markers for the European Robin, designing three multiplex panels to amplify 14 microsatellite loci. The level of polymorphism and its value for assessing parentage and genetic structure was estimated based on 119 individuals, including seven full families and 69 unrelated individuals form Poland's Białowieża Primaeval Forest and an additional location in Portugal. All markers appeared to be highly variable. Analysis at the family level confirmed a Mendelian manner of inheritance in the investigated loci. Genetic data also revealed evidence for extra-pair paternity in one family. The set of markers that we developed are proven to be valuable for analysis of the breeding biology and population genetics of the European Robin.

Heat shock proteins expression during thermal risk exposure in the temperate xerothermic ant Formica cinerea

The abiotic conditions of the desert habitat fluctuate in a circadian rhythm of hot days and cold nights. Species living in desert habitats evolved many adaptations to increase their chances of survival. However, abiotic conditions in xerothermic habitats of a temperate climate are much different. Diurnal fluctuations are not as strong, but animals have to cope with seasonal changes and hibernate during the winter, which may potentially influence their adaptations to critical temperature conditions. We attempted to assess heat resistance adaptations using the example of a widely distributed xerothermic ant Formica cinerea. Using Real-Time PCR, we measured the expression of three heat shock protein genes (Hsp60, Hsp75, Hsp90) and assessed the adaptations of F. cinerea to enable foraging in risk prone conditions. The analysis of gene expression using the Generalized Linear Model surprisingly indicated that there was no significant effect of temperature when comparing workers from the control (23ºC) with workers foraging on the surface of hot sand (47-54ºC). As a next step we tried to estimate the threshold of a thermal resistance with the use of thermal chambers. Expression of all Hsps genes increase compare to the control group, expression of Hsp60 and Hsp90 continued up to 45ºC.

Sequencing and Analysis of the Complete Organellar Genomes of Prototheca wickerhamii

Of the Prototheca genus, Prototheca wickerhamii has the highest clinical significance in humans. However, neither nuclear nor organellar genomes of this species were sequenced until now. The hitherto determined and analyzed mitochondrial and plastid genomes of the alleged P. wickerhamii species belong in fact to another species, recently named Prototheca xanthoriae. This study provides a first insight into the organellar genomes of a true P. wickerhamii (type strain ATCC 16529). The P. wickerhamii mitochondrion had a 53.8-kb genome, which was considerably larger than that of Prototheca ciferrii (formerly Prototheca zopfii gen. 1) and Prototheca bovis (formerly Prototheca zopfii gen. 2), yet similarly functional, with the differences in size attributable to a higher number of introns and the presence of extra unique putative genes. The 48-kb plastid genome of P. wickerhamii, compared to autotrophic Trebouxiophyceae, was highly reduced due to the elimination of the photosynthesis-related genes. The gene content of the plastid genome of P. wickerhamii was, however, very similar to other colourless Prototheca species. Plastid genome-based phylogeny reinforced the polyphyly of the genus Prototheca, with Helicosporidium and Auxenochlorella branching within clades of Prototheca species. Phylogenetic reconstruction also confirmed the close relationship of P. wickerhamii and P. xanthoriae, which is reflected in the synteny of their organellar genomes. Interestingly, the entire set of atp genes was lost in P. wickerhamii plastid genome, while being preserved in P. xanthoriae.
