Renal cell carcinoma (RCC) is one of the most common malignant tumors in urogenital system with an incidence accounting for about 3% of the whole body malignant tumor. NudC domain containing 1 (NudCD1), a new member of NudC family distributed in nucleus, is found to be upregulated in multiple tumors. However, its expression and role in RCC tissue has not been elucidated. NudCD1 expression in RCC tissue was measured by western blot and immunohistochemistry (IHC). NudCD1 level was elevated by overexpression vector to investigate its regulatory role on LIS1/Dynein signaling pathway. Cell morphology, intracellular localization, and cell division were observed by immunofluorescence together with delayed microscope photograph. The impact of NudCD1 overexpression on cell migration was assessed by Transwell assay. NudCD1 expression was significantly increased in RCC tissue compared with that in adjacent normal control. NudCD1/LIS1/Dynein signaling pathway was obviously upregulated in RCC tissue. Overexpression of NudCD1 level in A498 cell line markedly elevated NudCD1/LIS1/Dynein signaling pathway, suggesting they might be involved in RCC process. NudCD1 upregulation also caused abnormal microtubule fasciculus structure with multinuclear morphology, and promoted cell migration. NudCD1 expression was obviously increased in RCC and affected RCC cell division and migration possibly through activating NudCD1/LIS1/Dynein signaling pathway, indicating therapeutic targeting NudCD1 might be a new approach to inhibit RCC cell migration.
To report our experience and evaluate the efficacy and safety of real-time ultrasonography-guided percutaneous nephrolithotomy (PCNL) using SonixGPS navigation in patients with kidney stones.From December 2012 to May 2013 in our hospital, the SonixGPS needle tracking system was used for real-time ultrasonography-guided puncture during PCNL in 25 patients with kidney stones. All the patients were under general anesthesia, in prone position; holmium laser lithotripsy was used in all patients after renal access was established. The time needed to perform puncture and the success rate of PCNL were recorded as two most important outcomes of this study.All 25 patients underwent PCNL successfully with the help of SonixGPS navigation. The mean puncture time was 5.5 minutes (range 3-9 min), and the mean operative time was 74 minutes (range 30-100 min), respectively. The primary stone clearance rate of PCNL was 92% (23/25); no major intraoperative or postoperative complications occurred.The results of this study indicate that puncture under SonixGPS navigation is safe and efficacious in PCNL, which may provides a new choice for PCNL.
Objective To evaluate the symptoms of lower urinary tract dysfunction with chronic prostatitis(CP) by analyzing urodynamic results.Methods The retrospective study was based on 30 patients with type Ш of chronic prostatitis in urology clinics.The clinical data included the measurement of maximum urofow rate(Qmax),prostatic pressure,filling cystometry,pressure-flow(P/Q),and voiding and electromyography of the external urethral sphincter.Results Data showed low maximum uroflow rate in 18 patients(60.0%),had increased maximum prostatic pressure in 5 patients(16.7%),detrusor hyperreflexia in 11 patients(36.7%),bladder outflow obstruction in 9 patients(30%),hypersensitive bladder in 5 patients(16.7%),low compliance bladder in 4 patients(13.3%),detrusor contraction in 4 patients(13.3%),detrusor-sphincter dyssynergia in 14 patients(46.7%).Conclusions The lower urinary tract symptoms may be associated with bladder hyperesthesia,bladder outflow obstruction,detrusor and sphincter coordinated disturbances induced by prostate or pelvis factors.We believe that recognizing and eliminating these relative factors will be helpful to increase therapy efficacy on chronic prostatitis.
Objective: To inverstigate the best condition in chemosensitivity of gastric cancer cell line-SGC in vitro.Methods: Design orthogonal test to screen the best condition of drug sensitivity test.Results: The best condition was that the number of seeded cell was(2~3)×10~4/ml,the drug concentration was 10PPC and the killing time was 72 h.Conclusion: The best condition can improve the sensitivity for drug sensitivity test,and maybe useful for individual chemotherapy.
Using Darth,Virginia,Fengxiang three varieties of strawberries as test materials,strawberry stem tip regeneration in vitro was researched to produce high-yielding and high-quality strawberries and to improve efficiency.With MS as the basic culture medium,different concentration gradient plant growth regulators were added to conduct buds induction and tube seedlings proliferation.Vaccination in MS,1/2 MS culture to rooting induction and picked out the best rooting medium.The results showed that:the best bud culture medium for proliferation respectively:Darth,Virginia MS + 2.0 mg·L-16-BA + 0.1 mg·L-1 NAA;Fengxiang MS + 0.5 mg·L-16-BA + 0.05 mg·L-1NAA.1/2 MS culture was the best rooting basic culture medium;Which in a 1/2 MS + 0.5 mg·L-1IBA,rooting rate was 100% and grew strong,stout and more.Three materials vitro-preservation callus induction were higher and provided the basis for the high yield and good quality strawberry production
OBJECTIVE:To study the promotion and protective effects of ginkgolide B(GBE) on the growth and differentiation of embryonic rat midbrain neurons.METHODS:The primary cell culture was made in vitro by using midbrain neuronic cell suspension from 14 days old Wistar rats.Rats were divided into normal control group,GBE control(50 mg·L-1) group,KA(100 μmol·L-1) ,NGF(50 mg·L-1+KA100 μmol·L-1) ,GB high-dose,medium-dose and low-dose(100,50,25 mg·L-1+KA100 μmol·L-1) group.The embryonic rat midbrain neurons were cultured for 10 days respectively.Neurons was indentificated by NSE.Subsequently,the contents of malondialdehyde(MDA) and nitric oxide(NO) in the cultured cells were measured by fluorometric assay and nitric acid reduction methods.Superoxide dismutase(SOD) activities were detected by xanthinoxidase method.RESULTS:Cell growth and differentiation in GB high-dose,medium-dose and low-dose groups were significantly better than in KA group.Compared with normal control group,the activities of SOD in KA group was weakened significantly,while the content of MDA,NO were increased significantly.Compared with KA group,the activities of SOD in GB high-dose,medium-dose and low-dose groups was enhanced significantly,while the content of MDA,NO were decreased significantly.CONCLUSION:GBE can promote the growth and differentiation of embryonic rat midbrain neuronic cells and prevent the nerves from damage made by KA,which may be related with relief the lipid peroxidation in the cultured embryonic rat midbrain neurons.

Purpose: To prospectively evaluate 5α-reductase inhibitors (5αRIs) for benign prostatic hyperplasia (BPH) patients with a large prostate (>80 mL) after transurethral resection of the prostate (TURP).Materials and methods: Eighty-seven patients were recruited from January 2007 to October 2014. Patients were randomized into a trial and a control group. The trial group was treated with 5αRIs for 3 years after TURP, while the control group received a placebo. We evaluated the indicators before, peri and after TURP.Results: There were no significant differences in the indicators before and peri-TURP. Six months later, there were significant differences in PSA and hematuria (HU). Three years after TURP, there were significant differences in prostate volume (PV), level of prostate-specific antigen (PSA), the maximum flow rate (Qm), and HU between the trial and control groups. Additionally, there were significant differences in the PV, PSA, international prostate symptom score (IPSS), patient quality of life (QoL) in the trial group alone between those treated with finasteride and those treated with dutasteride.Conclusions: After TURP for large BPH, administration of 5αRIs for 3 years improved PV, PSA, Qm and HU. Additionally, dutasteride produced superior improvements in PV, PSA, IPSS and QoL compared with finasteride.
