Proteomic analysis of glutamate‐induced toxicity in HT22 cells

Abstract In the present study, we have investigated the proteome changes associated with glutamate‐induced HT22 cell death, a model system to study oxidative stress‐mediated toxicity. Among a number of HT22 proteins exhibiting altered expression, several molecular chaperones demonstrated substantial changes. For example, the levels of Hsp90 and Hsp70 decreased as cell death progressed whereas that of Hsp60 increased dramatically. Interestingly, cytosolic Hsp60 increased more prominently than mitochondrial Hsp60. Concomitantly, the accumulation of poly‐ubiquitylated proteins and differential regulation of the peptidase activities and the subunits of 26S proteasomes were observed in glutamate‐treated HT22 cells. Our findings that the molecular chaperones and the ubiquitin‐proteasome system undergo changes during glutamate‐induced HT22 cell death may suggest the importance of a protein quality control system in oxidative damage‐mediated toxicity.

Compact method of SARS-CoV-2 PCR detection enhances overall process of diagnosing Delta and Omicron variants

Abstract Nations across the world are currently suffering from various mutations of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). The ability to infect via respiratory routes such as coughs and sneezes and high mutation rate due to its single strand positive-sense RNA nature makes it very challenging to effectively contain, diagnose, and vaccinate. Moreover, current diagnostic guidelines insist PCR detection for SARS-CoV-2 be done as individual trials for sorting out one mutated variant at a time. To improve the overall diagnosis process and further enhance the efficiency of PCR tests, 4 distinct color emitting probes were used to simultaneously match Delta variant, Omicron variant, positive and negative controls. Amplified signals of individual and merged primers sets were able to successfully detect both Delta and Omicron variants. Additional validation of PCR trials and RNA extracts of SARS-CoV-2 patients further showed the effectiveness of the implemented method. The improved method will further compress the current detection protocol and further benefit diagnosing the ongoing pandemic.

Protein profiling and identification of modulators regulated by the E7 oncogene in the C33A cell line by proteomics and genomics

Human papillomaviruses (HPVs) have been recognized as the primary cause of cervical cancer. HPV 16 E7 binds to tumor suppressor retinoblastoma protein, and interferes with its function, causing release of the transcription factor E2F, which influences expression of cell cycle-related genes. This study was performed to identify the genes and proteins modulated by the HPV E7 oncogene. An HPV-negative cervical cancer cell line (C33A) was prepared to establish a stable cell line expressing E7. In order to analyze the target molecules modulated by E7 expression, we used two approaches: matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) and DNA microarrays. Forty-seven spots were identified in C33A/E7 by two-dimensional electrophoresis and MALDI/TOF MS. Protein disulfide isomerase A3, integrase interactor 1 protein, growth inhibitory protein, glutathione S-transferase P, and vav proto-oncogene were down-regulated, whereas heat shock 60 kDa protein 1, Ku70 binding protein, alpha enolase, 26S proteasome subunit were up-regulated. A genomic approach using a microarray kit showed that IL-12R beta 1, cytochrome c, and tumor necrosis factor receptor II were induced by the E7 oncogene. These results suggest that E7 can evade immune surveillance by suppressing or inducing these cell signaling factors, cell cycle regulators, and chaperones.

A Thermostable Xylose Isomerase from Thermus thermophilus: Biochemical Characterization, Crystallization, and Preliminary X-ray Analyses

A highly thermostable xylose isomerase from Thermus thermophilus has been expressed in Escherichia coli and crystallized. The purified enzyme shows its optimum temperature at . It has been crystallized at room temperature using polyethylene glycol 4000 as the precipitant. The crystal belongs to the orthorhombic space group , with unit cell parameters of a = 73.34 , b = 144.05 , c = 155.07 . The presence of one molecule of tetrameric xylose isomerase in the asymmetric unit gives a crystal volume per protein mass () of 2.32 and the solvent content of 47.0% by volume. The diffraction pattern extends to 1.9 Bragg spacing with synchrotron radiation and a set of native data has been collected to 2.3 .

Peltier Heating-Assisted Low Temperature Plasma Ionization for Ambient Mass Spectrometry

Low temperature plasma (LTP) ionization mass spectrometry (MS) is one of the widely used ambient analysis methods which allows soft-ionization and rapid analysis of samples in ambient condition with minimal or no sample preparation. One of the major advantages of LTP MS is selective analysis of low-molecular weight, volatile and low- to medium-polarity analytes in a sam- ple. On the contrary, the selectivity for particular class of compound also implies its limitation in general analysis. One of the critical factors limiting LTP ionization efficiency is poor desorption of analytes with low volatility. In this study, a home-built LTP ioniza- tion source with Peltier heating sample stage was constructed to enhance desorption and ionization efficiencies of analytes in a sam- ple and its performance was evaluated using standard mixture containing fatty acid ethyl esters (FAEEs). It was also used to reproduce the previous bacterial identification experiment using pattern-recognition for FAEEs. Our result indicates, however, that the bacterial differentiation from FAEE pattern recognition using LTP ionization MS still has many limitations.
