Dental Structure and Element Analysis of Patients with Cleidocranial Dysplasia

Objective To investigate the structure and elements of deciduous teeth in a patient with cleidocranial dysplasia (CCD). Methods The deciduous teeth of a CCD patient were collected,the histological structure and elements of which were analyzed by scanning electron microscope and energy dispersive X-ray spectrometer. Results The patients have typically clinical characteristics of CCD. Spectrum analysis of enamel of CCD deciduous teeth showed that the proportion of Ca and P was significantly lower than that from normal,however,this did not appear to be the case for the dentin. In light microscope,the enamal of CCD teeth was shown no less than that of the corresponding normal teeth,howe-ver,more enamel lamella and enamel tuft were found;in scanning electron microscope,dentin of CCD teeth appeared irregular,size and distribution appeared un-uniformed;cellular cementum was virtually lacking. Conclusion The mutation of RUNX2 gene has an effect on histological structure and elements of deciduous teeth of patients with CCD.

[Association of vitamin D receptor gene polymorphisms with the susceptibility to chronic periodontitis of Han nationality].

To investigate association of vitamin D receptor (VDR) gene polymorphisms with the susceptibility to chronic periodontitis (CP) of Han Nationality.Buccal swabs from 166 patients with severe, moderate and mild CP respectively and 80 matched control individuals were collected. DNA was extracted from these buccal swabs using Chelex-100 method. VDR BsmI, ApaI, TaqI were tested with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The distribution of the genotypes and allele frequencies in the patient and control groups were analyzed.The frequency of VDR ApaI allele A was significantly higher among patients with CP than controls. Frequencies of VDR ApaI allele A were significantly higher in severe CP patients than in moderate CP and mild CP respectively. There was no significant difference in the genotype distribution or the allele frequencies of VDR BsmI and TaqI between the controls and CP patients.These data indicate that VDR ApaI allele A may be related to the susceptibility to CP in Han Nationality.

Research on Pathologic Changes of Periodontal Tissue in Diabetic Rats

Objective Established the model of diabetic rats,study the pathologic changes of diabetic rats and CD4,CD8,RANKL,OPG of periodontal tissue in diabetic rats.Methods Thirty-eight SD rats were randomly divided into following groups:diabetic group (n=30) and normal group (n=8).The diabetic rat models were established by injecting streptozotocin into its veins,the tissue sections were stained for histological and immunohistochemistry of CD4,CD8,RANKL,OPG,and LSCM was used to test CD4,CD8,RANKL,OPG expression between diabetic group and normal group.Results The results showed that CD4,CD8,RANKL were significantly higher than the normal group (P0.05),comparing with normal group,OPG was lower in the diabetic group,but have no significant difference (P0.05).Conclusion Diabetets mellitus may through changing immune adjustment and RANKL/OPG way to promote thedevelopment of periodontitis.

Device modeling analysis and simulation of SiC P-i-N diode under pulsed power conditions

Silicon carbide (SiC) material properties make it well suited for pulsed power applications. SiC has a higher electric field breakdown, allowing for thinner, more compact high voltage devices. Silicon carbide also has a much shorter minority carrier lifetime which equates to much faster recovery time; it also has good thermal conductivity equating to minimum cooling requirements for a pulsed system and a high Young's modulus enabling SiC to withstand high current and voltage pulse stresses [1].

A Semi-Analytical Solution for Estimating the Fracture Width in Wellbore Strengthening Applications

Abstract Lost circulation is a major challenge in well construction operations, especially where drilling margins are narrow or in pressure-depleted reservoirs. Wellbore strengthening techniques (e.g., StressCage) have successfully been used to increase formation fracture resistance and reduce mud losses during drilling operations. The increase of fracture resistance in sands has been used to improve wellbore stability (through the use of higher mud weights), reduce casing requirements, and access resources that may have been undrillable using conventional drilling methods. The StressCage technique requires the estimation of the width of an induced fracture at a target fracture length for a given wellbore pressure. This estimation involves either a finite element calculation or a closed form line crack analytical solution. Populating and running the finite element solution requires specialized software and expertise, which has limited its use to larger operators and service companies that are staffed with geomechanics experts. The closed form line crack analytical solution is both simple to implement and easy to use, but it assumes transverse isotropic in-situ stress conditions relative to the borehole axis, which is almost never the case in the presence of a deviated well. This assumption results in either the underestimation of the calculation of the fracture width in the presence of deviation or abnormal in-situ stresses, which can result in a failed implementation of a StressCage formulation. We have developed a new semi-analytical line crack solution that accounts for stress anisotropy from either borehole inclination or abnormal in-situ stresses. This new solution is simple to implement. The calculation of the fracture width has been verified against finite element calculations through a range of stress anisotropies, borehole sizes, Young’s moduli, Poisson’s ratios, and target increases in wellbore strength.

ITRW: Formulating a Roadmap for WBG and UWBG Materials and Devices

The purpose of the International Technology Roadmap for Wide-Bandgap Power Semiconductors (ITRW) Materials and Devices Working Group, which considers the materials science of Wide-and Ultra-Wide-Band-Gap (WBG and UWBG) semiconductors, in addition to device design, fabrication, and evaluation, is to formulate a long-term, international roadmap for WBG and UWBG materials and devices, consistent with the packaging and applications working groups of ITRW. The working group is co-chaired by Victor Veliadis (primarily representing silicon carbide (SiC) and related materials) and Robert Kaplar (primarily representing gallium nitride (GaN) and related materials, as well as emerging ultra-WBGs) and is split into four sub-working-groups, which are: 1) SiC materials and devices (co-chairs Jon Zhang and Mietek Bakowski). 2) Lateral GaN materials and devices (co-chairs Sameh Khalil and Peter Moens). 3) Vertical GaN materials and devices (co-chairs TBD). 4) Emerging UWBG materials and devices (co-chairs Mark Hollis). The first two subgroups represent technology that is far more mature than that of the latter two, and devices are available as commercial products in power applications. The primary focus of this article will be on developments in subgroups 1 and 2, with only brief descriptions of the latter two sub-groups, including future activities as they mature technologically.

Expression and purification of human amelogenin in Escherichia coli

OBJECTIVE To establish the expression and purification route for the gene encoding human amelongenin (AMG) mature peptide in Escherichia coli (E. coli). METHODS Recombined plasmid pGEX-4T-1/AMG was identified by double endonuclease digestion electrophoretogram and DNA sequence analysis. The recombined plasmid was transformed to E. coli BL21. The inducing time, isopropyl-beta-D-thiogalactopyranoside (IPTG) concentration and inducing temperature were optimized for the express system. Under the optimized condition, the target fusing protein in superatant, periplasm, plasm and inclusion body was analyzed separately. A great amount of target fusing protein was found in the dissoluble protein. AMG fusing protein was purified by the GSTrapFF affinity column. RESULTS Double endonuclease digestion electrophoretogram and DNA sequence analysis were done to identify the recombined vector pGEX-4T-1/AMG. The results were consistent with the anticipation. The optimum inducing time was 14.5 hours. The optimum IPTG concentration was 1.0 mmol/L. The optimum inducing temperature was 20 degrees C. Under this condition, the target protein was expressed to a maximum. Plentiful target protein was expressed in plasm and inclusion body under the optimized condition. A mount of plasm protein was obtained and purified by the GSTrapFF affinity column. The purified liquid was collected and analyzed by SDS-polyacrylamide gel electrophoresis (SDS-PACE). The protein electrophoresis map showed that AMG fusing protein was purified successfully. After twice elution, high pure fusing protein was obtained. CONCLUSION pGEX-4T-1/AMG system is used successfully to express human AMG fusing protein.

An Experimental Investigation on the Impact of Diffusion Osmosis, Chemical Osmosis, and Capillary Suction on Shale Alteration

Well bore instability in shales can be the most challenging and costly issue in drilling operations. Well bore instability in shales can be attributed to many factors, some of which have been well studied and documented. However, the physicochemical and mechanical property alterations in shales, which eventually lead to well bore failure, have been largely ignored. The movement of water and ions in and out of shales plays a major role in the alteration of the physicochemical and mechanical properties of shales. Water and ions can move in and out of shales by many mechanisms. The mostly agreed on mechanisms that are responsible for water and ion movement in and out of shales are diffusion osmosis, chemical osmosis, convection (e.g., hydraulic flow), and capillary suction. This article presents experiments used to analyze the effects of chemical osmosis, diffusive osmosis, and capillary suction on water and ion movement when shale interacts with drilling fluids. Results show that water movement is not only controlled by chemical osmosis (water activity), but is also influenced by diffusive osmosis and capillary suction. The immersion of shale into fluids changes the chemical composition of the shale due to ion movement, and thus its physicochemical and mechanical properties are altered. The gravimetric-swelling test is used in the laboratory to acquire a better understanding of the relationship between water flow, ion flow, and shale swelling. Insights gained from this study provide information used to optimize drilling fluids to effectively control or mitigate well bore instability when drilling through shale.
