Tracing Local Disorder in Near-Infrared-Upconverting Crystals of Li⁺-Doped Gd₂O₃ through the Gd(III)-O Bond Distance

Using atomic pair distribution function (PDF) analysis of high-energy synchrotron total scattering, the current study addresses how an imparted local disorder in near-infrared-upconverting (UC) crystals dictates the concerned bulk properties. Aiming to perturb the local crystal field symmetry around the activator ions, Li+ was incorporated in varying amounts within the UC crystals of Gd2O3/Yb3+/Ho3+. The respective photoluminescence (upconversion luminescence, UCL) intensities of the crystals got enhanced first and then diminished after a certain Li+ concentration. The phase of the crystal remained constant as the cubic Ia-3 space group in all the samples with no significant changes in average structures as obtained from the Rietveld refinement of synchrotron experiments. Results from extended X-ray absorption fine structure indicated some changes at the atomic level. A comparative analysis among the Li+-incorporated crystals was considered. Systematic PDF analysis revealed the appearance of local disorder, which was connected with the Gd(III)-O bond distances in some way. The extent of local disorder followed the variation trend of UCL intensities. When Gd1-O and Gd2-O approximately approached their respective shortest and longest limits among the Li+-incorporated crystals, the corresponding local disorder appeared to be maximum with the corresponding crystal showing the highest UCL intensity. The relative variation of Gd–O bond distances and hence the extent of local disorder was found to get manifested in the respective tensile lattice strains so generated. Through rationale and earlier published evidence, the study attempted to hypothesize that local disorder is fundamentally connected with the lattice strain in UC crystals at a significantly higher concentration of a symmetry perturbing agent.

The highly intense synchrotron-radiation-based X-ray beams offer an undisputed advantage in assessing the structural parameters of crystalline solids. Herein, through extensive synchrotron-based probing, an attempt was made to understand the structural attributes that dictate the variation of upconversion luminescence intensity (UCL) resulting from the distortion of local crystal field symmetry. Four different sets of UC crystals (NaYF4/Yb3+/Ln3+: Ln = Ho, Er, Tm) have been designed with varying Li+ concentrations. The analysis indicated that, out of many possible structural factors, the compressive lattice strains generated in those systems exhibited an unexpected correlation with the respective UCL intensities near their maximum values, irrespective of the activator species. Interestingly, the single-activator-based samples showed maximum UCL intensities when their respective compressive lattice strains reached close to a particular value. Through experimental evidence, these findings tend to extend and complement the existing hypothesis of symmetry distortion of a UC lattice by chemical manipulation.

Osteoblast‐Derived Giant Plasma Membrane Vesicles Induce Osteogenic Differentiation of Human Mesenchymal Stem Cells

Abstract Exosome‐mediated differentiation of human mesenchymal stem cell (hMSC) has opened up a new possibility for the directed osteogenic differentiation of hMSC by the osteoblast‐derived vesicles. Here, it is hypothesized that giant plasma membrane vesicles (GPMVs) generated from osteoblast, can direct the osteogenic differentiation of hMSC. GPMVs having different membrane characteristics are generated from osteoblast cells (MG‐63) using chemical vesiculants. It is deciphered that there is a treatment‐dependent variation in the size and complexity in the GPMVs. It is also showed a variation in lipid composition among the GPMVs. It is demonstrated that GPMVs can be fused with hMSC in vitro and such fusion has no detrimental effect on the viability of hMSC. Further, it is showed that there is a significant increase in the expression of osteogenic differentiation markers in GPMV‐fused hMSC at protein and mRNA level. A close correlation between the extent of fusion and the degree of osteogenic differentiation is observed. The GPMV reported here may be explored for future application in bone tissue engineering.
