Evaluating Wood Pulp as Feedstuff for Ruminants and Substrate for Aspergillus Fumigatus

TEN chemical pulps and two mechanical pulps were analyzed for Klason lignin, total carbohydrate, ash, in vitro rumen dry matter digestibility and single-cell protein growth by Aspergillus fumigatus. The pulp fines from each pulp that passed through a 100-mesh wet screening were also analyzed. In vitro digestibility ranged from 67% to 98% for the chemical pulps and pulp fines and from 0% to 7% for the mechanical pulps and pulp fines. Aspergillus fumigatus produced fungal growth with 14.5% to 30% crude protein in 4 days at 35 C when grown on chemical wood pulps from hardwood species with lignin content ranging from 25% to less than 0.1%. The fungus grew only on softwood pulps that had low-lignin content; it did not grow on the mechanical pulps.

Elevated-Temperature Technique for the Isolation of Salmonella from Streams

With a modified Moore swab for sampling bacteria, Salmonella organisms were recovered consistently from surface waters when the enrichment broths of SBG-sulfa and tetrathionate and Brilliant Green Agar plates were incubated at 41.5 C. When an equal portion of the same swab sample was incubated at 37.0 C, no salmonellae were detected. By use of the elevated-temperature technique, salmonellae were recovered from stream sites having relatively low total coliform densities of 2,200 per 100 ml and fecal coliform densities of 220 per 100 ml. These pathogens were isolated in water sampled as far as 73 river miles and 4 days time of travel downstream from the source of pollution under an ice cover that averaged 2.5 ft (76.2 cm) in thickness.

Acinetobacter spp.: distinct morphology on eosin methylene blue agar as an aid to identification in drinking water

Acinetobacter calcoaceticus, frequently found in drinking waters and implicated in nosocomial infections, was presumptively identified by its tiny, blue colonial appearance on Levine eosin methylene blue agar. All of the 33 isolates from drinking water showing this distinctive colonial appearance were identified as A. calcoaceticus.

Characterization of dysgonic, heterotrophic bacteria from drinking water

Only a small percentage of the heterotrophic bacteria encountered in water distribution systems are identifiable, because of these organisms fail to grow on the conventional media used for biochemical characterization. Organisms that would not subculture from the same standard plate count agar used for initial isolation were successfully subcultured on a low-nutrient medium, R3A. These cultures were then inoculated to a modified O/F base medium containing specific substrates. This, combined with a lower incubation temperature (30 degrees C), increased the enzymatic activity of many of the organisms. These reactions established a groundwork for tentative taxonomy.
