Spiroplasma leptinotarsae sp. nov., a Mollicute Uniquely Adapted to Its Host, the Colorado Potato Beetle, Leptinotarsa decemlineata (Coleoptera: Chrysomelidae)

Spiroplasma strain LD-1T (T = type strain), which was isolated from the gut of a Colorado potato beetle (Leptinotarsa decemlineata) larva collected in Maryland, was serologically distinct from other spiroplasmas. Similar isolates were obtained from other L. decemlineata specimens collected in various parts of North America, in Poland, and in other eastern European countries and from Leptinotarsa texana specimens collected in Texas. Cells of strain LD-1T, which in early passages were spiral, exhibited exceptionally rapid translational motility. This rapid motility and the spiral shape were lost after extended passage in culture. The organism required serum for growth. Originally isolated in coculture with insect cells in DCCM medium, strain LD-1T adapted to several media in the absence of cocultured cells. Use of anaerobic conditions allowed primary isolation in a variety of media. The organism did not grow in serum-free media containing 2% serum fraction. Optimal growth in M1D medium occurred at 30 to 37°C (doubling time, 7.2 h). On solid M1D medium containing 2.0% Noble agar (pH 6.25) at 30°C, strain LD-1T produced discrete colonies with numerous satellites. Strain LD-1T hydrolyzed arginine, but did not utilize urea; there was evidence of weak fermentation of glucose. The guanine-plus-cytosine content of the DNA was determined to be 25 ± 1 mol%, and the genome size was 1,085 kb. The results of extensive studies of the ecology of this spiroplasma suggest that it is host specific for Leptinotarsa beetles. Strain LD-1 (= ATCC 43213) is designated the type strain of a new species, Spiroplasma leptinotarsae.

Unclassified Viruses of Insects

Some spherical virus-like particles (VLPs) possess similarities to viruses in recognized groups but differ in one or more biochemical or morphological characteristics. There are many reports of unclassified viruses and VLPs in insects. The most abundant particles, termed VLP1 were present as filamentous and oval forms within vesicles in the corpus allatum, midgut, muscle, tracheal matrix, and oenocytes near fat body, and in the glial cells of corpus cardiacum and thoracic ganglion. The VLPs and copia are, therefore, probably the viral particle and proviral forms, respectively, of the copia element. The VLPs associated with the Mexican bean beetle appear to be a latent virus which also infects the host reproductive tissues and is transmitted vertically in infected beetle populations. Morphogenesis of an unusual spherical VLP has been observed in the nuclei of calyx cells in the ichneumonid parasitoid Nemeritis canescens. Infection with these VLPs also appears to significantly reduce the life span of adult crickets.

Birnaviridae is a new family established by the International Committee on Taxonomy of Viruses. The major difference between birnaviruses of insects and birnaviruses of invertebrates other than insects consists in the method of virus detection. Cytopathological studies to determine morphogenesis and viral structure have been conducted in fish cell lines since these viruses have never been observed in naturally infected host tissues. The Drosophila X virus (DXV) genome contains two segments of dsRNA which are composed of different nucleotide sequences. DXV was found contaminating Drosophila and Ceratitis capitata established cell lines. The contamination was eliminated by the use of virus-free Drosophila stocks to start the cell lines and serum screened on Drosophila flies for cell line maintenance. DXV has been isolated from natural populations of Culicoides sp. but no reports of histopathologic examinations have been presented. Cell cultures of Drosophila and Ceratitis capitata are susceptible to DXV.

Fine Structure of the Integument of the Two-Spotted Spider Mite, Tetranychus telarius (Acarina: Tetranychidae)

Electron micrographs of transverse and saggital sections of the opisthosoma of the two-spotted spider mite, Tetranychus telarius (L.), showed a layered cuticle and a hypodermal layer with a basement membrane. The entire cuticle measured 0.70 μ–0.91 μ thick, the epicuticle being 0.01 μ, exocuticle 0.09 μ–0.11 μ, and endocuticle 0.16 μ–0.78 μ. Cuticular extensions, lobed at the apices, were 1.37 μ–1.74 μ high and about 0.09 μ thick at the narrowest point. The hypodermal layer was about 1.19 μ–1.71 μ and delineated by the basement membrane.

Determination of the Active Nuclear Polyhedrosis Virus Content of Untreated Cabbages 1

A new method was used for determining the active nuclear polyhedrosis virus content of untreated marketable cabbages. The activity of the virus was determined by inter-polating the dose-mortality data obtained from bioassays of homogenized wrapper leaves, heads, and whole cabbages. An average of 4.61 × 10 3 active polyhedra/g of cabbage was obtained from untreated edible cabbage heads assayed in 1972.

Testing Penaeid Shrimp for Susceptibility to an Insect Nuclear Polyhedrosis Virus 1

Postlarval, early, and late juvenile stages of two species of penaeid shrimp, Penaeus aztecus Ives and P. setiferus (L.), were tested for susceptibility to a nuclear polyhedrosis virus from Autographa californica (Speyer). Shrimp were exposed to the virus by intramuscular inoculation of polyhedral protein-free virus and by feeding a diet containing virus polyhedra. Mortality attributable to viral infection did not occur dunng the 30-day test period, nor was there histological evidence of viral activity in shrimp hepatopancreas, gut, gill, striated muscle, ventral nerve, and segment nerve ganglia, or hypodermis.
