MiR-539-3p impairs osteogenesis by suppressing Wnt interaction with LRP-6 co-receptor and subsequent inhibition of Akap-3 signaling pathway

X-linked hypophosphatemia (XLH), an inheritable form of rickets is caused due to mutation in Phex gene. Several factors are linked to the disease’s aetiology, including non-coding RNA molecules (miRNAs), which are key post-transcriptional regulators of gene expression and play a significant role in osteoblast functions. MicroRNAs sequence analysis showed differentially regulated miRNAs in phex silenced osteoblast cells. In this article, we report miR-539-3p, an unidentified novel miRNA, in the functional regulation of osteoblast. MiR-539-3p overexpression impaired osteoblast differentiation. Target prediction algorithm and experimental confirmation by luciferase 3’ UTR reporter assay identified LRP-6 as a direct target of miR-539-3p. Over expression of miR-539-3p in osteoblasts down regulated Wnt/beta catenin signaling components and deteriorated trabecular microarchitecture leading to decreased bone formation in ovariectomized (Ovx) mice. Additionally, biochemical bone resorption markers like CTx and Trap-5b were elevated in serum samples of mimic treated group, while, reverse effect was observed in anti-miR treated animals along with increased bone formation marker P1NP. Moreover, transcriptome analysis with miR-539-3p identified a novel uncharacterized Akap-3 gene in osteoblast cells, knock down of which resulted in downregulation of osteoblast differentiation markers at both transcriptional and translational level. Overall, our study for the first time reported the role of miR-539-3p in osteoblast functions and its downstream Akap-3 signalling in regulation of osteoblastogenesis.

Lucas sequences and Fibonacci triads of graphs in PMO calculation on the charge‐transfer bands of a series of EDA complexes: Correlation with experimental and AM1 results

The use of Lucas sequences and Fibonacci triads of graphs in a complete PMO calculation on a series of phenols is shown. The results were correlated with charge-transfer absorption maxima of a series of molecular EDA complexes of p-chloranil with these phenols as donors. The correlation brings out the PMO parameters in a straightforward way. The graph theoretical results are also shown to correlate well with the donor ionization potentials calculated by the AM1 method, and this correlation yields a value of the sp2-C Coulomb integral, α, which agrees well with the one obtained from photoelectron spectroscopic results. © 1997 John Wiley & Sons, Inc. Cytotoxic clerodane diterpenoids from the leaves ofPolyalthia longifolia

The ethanolic extract of Polyalthia longifolia var. pendula was fractionated to get a hexane-soluble residue, which led to the isolation of four clerodane diterpenes: (-)-3α,16α-dihydroxycleroda-4(18),13(14)Z-dien-15,16-olide (1), (-)-3β,16α-dihydroxycleroda-4(18), 13(14)Z-dien-15,16-olide (2), (-)-16α-hydroxycleroda-3,13(14)Z-dien-15,16-olide (3) and (-)-16-oxocleroda-3,13(14)E-dien-15-oic acid (4). Diterpene 1 is a new compound, while 2 is reported for first time from this plant. Both 1 and 2 were tested for their growth inhibitory activity on four cancer cell lines in vitro. IC(50) values suggest that they are effective as cytotoxic agents.

Assessing Radiometric Parameters at a Continental Global Atmosphere Watch (GAW) Station, Nagpur, in India

Several radiometric parameters viz. direct solar irradiance, long wave radiation, Angstrom turbidity and transmission coefficient for Nagpur have been analyzed and discussed based on data for the period of 1989–1998. Solar irradiance at Nagpur is the highest in winter and amongst the winter months in January due to cloudless sky and good transparency of the atmosphere. It is the least during the monsoons due to increased moisture content of the atmosphere and extended cloudiness. Diurnal variation in direct solar irradiance shows that for all the months the peak is reached between optical air masses 1.5 FN and 1.5 AN with a maximum value of about 700 W/m $^2$ in January. Turbidity values show fluctuations on yearly basis. During the study period, the lowest value was observed to be 0.082 in 1990 and the highest was 0.108 in 1996. In a year, winter season experiences lowest turbidity while the post-monsoon season witnesses the highest. Diurnal variation of turbidity indicates that optical air mass 3.0 FN is less turbid for all the months in a year while the peak is reached in between air masses 1.5 FN and 1.5 AN. It is seen that transparency of the atmosphere over Nagpur decreases from winter till May. This is to be attributed to the increasing dust content of the atmosphere over Nagpur. The atmosphere transmits only 65% of the incident energy. At early morning and late afternoon hours, transparency is found to be higher when compared to higher solar elevations. Rather, the transparency decreases from morning till noon and later increases. This could be attributed to convective activity during the maximum temperature epoch of the day. Net long wave radiation is found to be the highest in pre-monsoon (64–65 W/m $^2$ ). With the monsoon onset, it goes on decreasing, with the lowest value of 42.6 W/m $^2$ in August. This is mainly due to the extensive cloud cover and high humidity conditions. After the monsoon withdrawal again it starts rising.

Fatou theorem and its converse for positive eigenfunctions of the Laplace-Beltrami operator on Harmonic $NA$ groups

We prove a Fatou-type theorem and its converse for certain positive eigenfunctions of the Laplace-Beltrami operator $\mathcal{L}$ on a Harmonic $NA$ group. We show that a positive eigenfunction $u$ of $\mathcal{L}$ with eigenvalue $β^2-ρ^2$, $β\in (0,\infty)$, has an admissible limit in the sense of Korányi, precisely at those boundary points where the strong derivative of the boundary measure of $u$ exists. Moreover, the admissible limit and the strong derivative are the same. This extends a result of Ramey and Ullrich regarding nontangential convergence of positive harmonic functions on the Euclidean upper half space.

Evaluation of the precipitation-in-gel reaction in the diagnosis of smallpox.

Specimens of vesicular or pustular fluids and of scabs from patients with smallpox as well as emulsions of variola-infected chorioallantoic membrane (CAM) were tested for virus titres and by the precipitation-in-gel (PIG) reaction. They were also tested after exposing them directly to sunlight and after keeping them at temperatures of -20 degrees C, 4 degrees C, and 25 degrees C. It was found that when extracts of fresh specimens were diluted to the point where the PIG reaction became negative there was still a titre of 10(4)-10(5) infectivity in the swab extracts and 10(3)-10(4) infectivity in the scab extracts. It was also found that the PIG reactions were all negative on specimens that were kept for 14 days at 25 degrees C, and that several were negative after only 7 days; the loss in infectivity titre, however, was only slight in all the specimens tested. It is concluded that the laboratory diagnosis of smallpox by virus inoculation of CAM is more reliable than by the PIG test.
