In the ethylene signalling pathway, EIN2 is an essential signal transducer linking ethylene perception on endoplasmic reticulum to transcriptional regulation in the nucleus, EIN3/EIL1 transcription factors initiate various responses leading to an ethylene response, and EBFs function in ethylene perception by regulating EIN3/EIL1 turnover. In this work, we isolated and characterised three EIN3 homologs (CpEIN3a, CpEIN3b, and CpEIL1), and two EBF homologs (CpEBF1 and CpEBF2) from papaya fruits. During the fruit development, the transcripts of all these genes were accumulated at a low level before reaching a maximum in the third month, then showed a decrease. Exogenous ethylene application and 1-MCP treatment had different effects on the expression of these genes. The correlations between various physiological characteristics with the gene expressions showed that CpEIN3b, CpEIL1, and CpEBF1/2 were negative regulators in terms of papaya fruit ripening under exogenous ethylene treatment while CpEIN3a was a positive one, CpEIN3a was negatively correlated with the respiration rate and ethylene production of papaya fruits under 1-MCP treatment. It is suggested that CpEIN2, CpEIN3a/b, CpEIL1, and CpEBF1/2 are development- and ripening-related genes in papaya fruits. These results provide new insights into the understanding of the ethylene signalling cascade in papaya fruit development and ripening.
Papaya (Carica papaya L.) is sensitive to low temperature and easy to be subjected to chilling injury, which causes fruit ripening disorder. This study aimed to investigate the relationship between the expression of genes related to ethylene and fruit ripening disorder caused by chilling injury. Papaya fruits were firstly stored at 7°C and 12°C for 25 and 30 days, respectively, then treated with exogenous ethylene and followed by ripening at 25°C for 5 days. Chilling injury symptoms such as pulp water soaking were observed in fruit stored at 7°C on 20 days, whereas the coloration and softening were completely blocked after 25 days, Large differences in the changes in the expression levels of twenty two genes involved in ethylene were seen during 7°C-storage with chilling injury. Those genes with altered expression could be divided into three groups: the group of genes that were up-regulated, including ACS1/2/3, EIN2, EIN3s/EIL1, CTR1/2/3, and ERF1/3/4; the group of genes that were down-regulated, including ACO3, ETR1, CTR4, EBF2, and ERF2; and the group of genes that were un-regulated, including ACO1/2, ERS, and EBF1. The results also showed that pulp firmness had a significantly positive correlation with the expression of ACS2, ACO1, CTR1/4, EIN3a/b, and EBF1/2 in fruit without chilling injury. This positive correlation was changed to negative one in fruit after storage at 7°C for 25 days with chilling injury. The coloring index displayed significantly negative correlations with the expression levels of ACS2, ACO1/2, CTR4, EIN3a/b, ERF3 in fruit without chilling injury, but these correlations were changed into the positive ones in fruit after storage at 7°C for 25 days with chilling injury. All together, these results indicate that these genes may play important roles in the abnormal softening and coloration with chilling injury in papaya.

Human activities, such as mining, industrialization, industrial waste emissions, and agricultural practices, have caused heavy metals to become widespread and excessively accumulated in soil. The high concentrations of heavy metals in soil can be toxic to plants, severely affecting crop yield and quality. Moreover, these heavy metals can also enter the food chain, affecting animals and humans and leading to various serious illnesses. Selenium (Se) is not only an essential element for animals and humans but is also beneficial for plants, as it promotes their ability to respond actively to biotic and abiotic stresses. The global issue of Se deficiency in diets has made plants the primary source for human Se supplementation. This paper comprehensively reviews the effects of heavy metal stress on plant growth and development, physiological responses of plants to such stress, and the intracellular transport processes of heavy metals within plants. It particularly focuses on the mechanisms by which Se alleviates heavy metal stress in plants. Additionally, the study delves into how Se significantly enhances plant tolerance mechanisms against typical heavy metals, such as cadmium (Cd), lead (Pb), and mercury (Hg). This integrative research not only expands the boundaries of research in the field of plant heavy metal stress and Se application but also provides new perspectives and solutions for understanding and addressing complex environmental heavy metal pollution issues. By integrating these aspects, this paper not only fills existing gaps in the literature but also offers comprehensive scientific basis and strategic recommendations for environmental protection and sustainable agriculture development.
Ginkgolide is a unique terpenoid natural compound in Ginkgo biloba, and it has an important medicinal value. Proper selenium has been reported to promote plant growth and development, and improve plant quality, stress resistance, and disease resistance. In order to study the effects of exogenous selenium (Se) on the physiological growth and the content of terpene triolactones (TTLs) in G. biloba seedlings, the seedlings in this work were treated with Na2SeO3. Then, the physiological indexes, the content of the TTLs, and the expression of the related genes were determined. The results showed that a low dose of Na2SeO3 was beneficial to plant photosynthesis as it promoted the growth of ginkgo seedlings and increased the root to shoot ratio. Foliar Se application significantly increased the content of soluble sugar and protein and promoted the content of TTLs in ginkgo leaves; indeed, it reached the maximum value of 7.95 mg/g in the ninth week, whereas the application of Se to the roots inhibited the synthesis of TTLs. Transcriptome analysis showed that foliar Se application promoted the expression levels of GbMECPs, GbMECT, GbHMGR, and GbMVD genes, whereas its application to the roots promoted the expression of GbDXS and GbDXR genes. The combined analysis results of metabolome and transcriptome showed that genes such as GbDXS, GbDXR, GbHMGR, GbMECPs, and GbCYP450 were significantly positively correlated with transcription factors (TFs) GbWRKY and GbAP2/ERF, and they were also positively correlated with the contents of terpene lactones (ginkgolide A, ginkgolide B, ginkgolide M, and bilobalide). Endogenous hormones (MeJA-ILE, ETH, and GA7) were also involved in this process. The results suggested that Na2SeO3 treatment affected the transcription factors related to the regulation of endogenous hormones in G. biloba, and further regulated the expression of genes related to the terpene synthesis structure, thus promoting the synthesis of ginkgo TTLs.
Plant growth is affected by various abiotic stresses, including water, temperature, light, salt, and heavy metals. Selenium (Se) is not an essential nutrient for plants but plays important roles in alleviating the abiotic stresses suffered by plants. This article summarizes the Se uptake and metabolic processes in plants and the functions of Se in response to water, temperature, light, salt, and heavy metal stresses in plants. Se promotes the uptake of beneficial substances, maintains the stability of plasma membranes, and enhances the activity of various antioxidant enzymes, thus alleviating adverse effects in plants under abiotic stresses. Future research directions on the relationship between Se and abiotic stresses in plants are proposed. This article will further deepen our understanding of the relationship between Se and plants.
Selenium (Se) is essential for human health, however, Se is deficient in soil in many places all around the world, resulting in human diseases, such as notorious Keshan disease and Keshin–Beck disease. Therefore, Se biofortification is a popular approach to improve Se uptake and maintain human health. Beneficial microorganisms, including mycorrhizal and root endophytic fungi, dark septate fungi, and plant growth-promoting rhizobacteria (PGPRs), show multiple functions, especially increased plant nutrition uptake, growth and yield, and resistance to abiotic stresses. Such functions can be used for Se biofortification and increased growth and yield under drought and salt stress. The present review summarizes the use of mycorrhizal fungi and PGPRs in Se biofortification, aiming to improving their practical use.
