Carbon-dot-based fluorescent turn-on sensor for selectively detecting sulfide anions in totally aqueous media and imaging inside live cells

Sulfide anions are generated not only as a byproduct from industrial processes but also in biosystems. Hence, robust fluorescent sensors for detecting sulfide anions which are fast-responding, water soluble and biocompatible are highly desirable. Herein, we report a carbon-dot-based fluorescent sensor, which features excellent water solubility, low cytotoxicity and a short response time. This sensor is based on the ligand/Cu(II) approach so as to achieve fast sensing of sulfide anions. The carbon dot (CD) serves as the fluorophore as well as the anchoring site for the ligands which bind with copper ions. For this CD-based system, as copper ions bind with the ligands which reside on the surface of the CD, the paramagnetic copper ions efficiently quench the fluorescence of the CD, affording the system a turn-off sensor for copper ions. More importantly, the subsequently added sulfide anions can extract Cu(2+) from the system and form very stable CuS with Cu(2+), resulting in fluorescence enhancement and affording the system a turn-on sensor for sulfide anions. This fast-responding and selective sensor can operate in totally aqueous solution or in physiological milieu with a low detection limit of 0.78 μM. It displays good biocompatibility, and excellent cell membrane permeability, and can be used to monitor S(2-) levels in running water and living cells.

Spherulite Formation of Lipophilic Surfactant Induced by Noncrystalline Amphiphilic Diblock Copolymer

As the lipophilic surfactant tetraoctylammonium bromide (TOAB) was cast from its organic solutions, needle-shape crystals formed. However, with the addition of an amphiphilic noncrystalline diblock copolymer [(poly(acrylic acid-b-styrene)] into the TOAB solutions, spherulites formed in the solid films cast from the solutions. Under preferable conditions, millimeter-sized spherulites could be obtained. It has been found that some factors such as the type of solvent, the film-forming temperature, and the ratio of polymer to surfactant can affect the spherulite formation. Small angle X-ray scattering and wide-angle X-ray diffraction investigation suggests that the formation of the spherulites and the pure TOAB crystals are organized by closely packed lamellar structure, while the addition of diblock copolymer decreased the degree of order of TOAB crystals. Dynamic light scattering study reveals that, in organic solvents such as tetrahydrofuran, TOAB formed molecular-disperse solution, while the amphiphilic copolymer chains formed micelles with or without the presence of TOAB molecules in the solution. We suppose that the morphology change of TOAB crystal is induced by the diblock polymer chains: During the solvent-evaporating film formation, the hydrophobic PS blocks of the amphiphilic copolymer resided between some of the TOAB lamellar crystallites, which might cause the splaying and branching of the surfactant crystallites during the crystal growth and eventually lead to spherulite formation. This result could provide a useful way for spherulite formation, and open interesting opportunities for adjusting the crystal morphology and/or properties of lipophilic surfactant.

A fluorescent ratiometric nanosensor for detecting NO in aqueous media and imaging exogenous and endogenous NO in live cells

As an endogenously generated signal-inducing agent in the immune, cardiovascular and nervous systems, nitric oxide (NO) is involved in various biological processes and a large number of pathologies. Hence, sensitive and reliable detection of NO in biological samples and inside living cells has been of great interest. Herein, we reported a carbon-dot-based fluorescent ratiometric nanosensor for NO detection in water and imaging in living cells. Ratiometric sensing is realized through the fluorescence resonance energy transfer (FRET) process, and the carbon dots (CDs) serve as both the energy donor and the anchoring site for the NO probe moiety, which turns into the energy acceptor in the presence of NO. This nanosensor is highly selective for NO, and is operable in totally aqueous media with a very low detection limit of 3 nM. Due to the biocompatible nature and small size of the carbon dots, the nanosensor exhibits very low cytotoxicity and can easily permeate through the cell membrane. The nanosensor is capable of tracking the exogenous NO levels in several cell lines such as Hela, L929 and Raw 264.7 cells, and it can be also used to visualize the endogenously produced NO in the Raw 264.7 macrophage cell line.

The fluorescence spectra of a novel monomer, dimethyl aminophenyl acrylamide solution, and its polymer solution have been studied in this work. Results have shown that, there are two parts, electron donor and acceptor, in a single molecule. The electron transfer between molecules was occured by light excitation. The fluorescence intensity of this monomer is weaker, than that o'f its corresponding polymer. This monomer could be polymerized by light excitation

Light-Induced Anisotropy in Azobenzene Doped Copolymer Films

In this study, an azobenzene dye, Dispersion Red 1 (DR1) was doped into the copolymers of methyl methacrylate(MMA) and butyl acrylate(BA) to obtain five bulk composites with varied ratios of methyl methacrylate to butyl acrylate. An experimental setup, in which the He-Ne laser produced signal beams and Ar+ laser, the pump beams was employed to investigate the photoinduced anisotropic properties of these samples. The results show that, the lower rigidity of the copolymers chains caused by the increased BA content would lead to a lower extent of birefringence for the samples. With the increased pump beam power, the extent of birefringence first slightly increased, reached a maximum value, and then decreased. These increases and then decreases would be caused by the co-effects of both orientation and saturation mechanism. On the other hand, the optical dichroism properties can be detected in the bulk samples with photoinduced anisotropic property. The birefringence and dichroism properties exhibited by the dye-doped bulk composites have great potential in optical devices and optical communication systems; in particular, these bulk polymeric materials are very important for three-dimensional optical applications.

A Gold Nanocage/Cluster Hybrid Structure for Whole‐Body Multispectral Optoacoustic Tomography Imaging, EGFR Inhibitor Delivery, and Photothermal Therapy

Gold nanocages (AuNCs) and gold nanoclusters (AuClusters) are two classes of advantageous nanostructures with special optical properties, and many other attractive properties. Integrating them into one nanosystem may achieve greater and smarter performance. Herein, a hybrid gold nanostructure for fluorescent and optoacoustic tomography imaging, controlled release of drugs, and photothermal therapy (PTT) is demonstrated. For this nanodrug (EA-AB), an epidermal growth factor receptor (EGFR) inhibitor erlotinib (EB) is loaded into AuNCs, which are then capped and functionalized by biocompatible AuCluster@BSA (BSA = bovine serum albumin) conjugates via electrostatic interaction. Upon cell internalization, the lysosomal proteases and low pH cause the release of EB from EA-AB, and also induce fluorescence restoration of the AuCluster for imaging. Irradiation with near-infrared light further promotes the drug release and affords a PTT effect as well. The AuNC-based nanodrug is optoacoustically active, and its biodistribution and metabolic process have been successfully monitored by whole-body and 3D multispectral optoacoustic tomography imaging. Owing to the combined actions of PTT and EGFR pathway blockage, EA-AB exhibits marked tumor inhibition efficacy in vivo.

A Fluorescence Turn‐on Sensor for Iodide Based on a Thymine–Hg^II–Thymine Complex

Abstract Iodide plays a vital role in many biological processes, including neurological activity and thyroid function. Due to its physiological relevance, a method for the rapid, sensitive, and selective detection of iodide in food, pharmaceutical products, and biological samples such as urine is of great importance. Herein, we demonstrate a novel and facile strategy for constructing a fluorescence turn‐on sensor for iodide based on a T–Hg II –T complex (T=thymine). A fluorescent anthracene–thymine dyad (An‐T) was synthesized, the binding of which to a mercury(II) ion lead to the formation of a An‐T–Hg II –T‐An complex, thereby quenching the fluorescent emission of this dyad. In this respect, the dyad An‐T constituted a fluorescence turn‐off sensor for mercury(II) ions in aqueous media. More importantly, it was found that upon addition of iodide, the mercury(II) ion was extracted from the complex due to the even stronger binding between mercury(II) ions and iodide, leading to the release of the free dyad and restoration of the fluorescence. By virtue of this fluorescence quenching and recovery process, the An‐T–Hg II –T‐An complex constitutes a fluorescence turn‐on sensor for iodide with a detection limit of 126 n M . Moreover, this sensor is highly selective for iodide over other common anions, and can be used in the determination of iodide in drinking water and biological samples such as urine. This strategy may provide a new approach for sensing some other anions.
