Investigating the Anti-inflammatory Activity of Wheatgrass Juice in Albino Rats

To study the anti-inflammatory effect of wheatgrass juice in albino rats. Anti-inflammatory activity of wheatgrass juice was evaluated by using carrageenin-induced rat hind paw edema model for acute study, rexin pellet-induced granuloma in rat model and formalin-induced rat hind paw edema model for chronic study. The results obtained were compared with diclofenac Sodium, standard drug and control. Wheatgrass produced significant anti-inflammatory effect in chronic models. In rexin pellet model, the effect of wheatgrass was more than diclofenac sodium. In formalin-induced rat hind paw edema model, the effect of wheat grass was less than diclofenac sodium. In carrageenin-induced rat hind paw edema model wheatgrass juice showed very minimal activity. However, the wheatgrass juice showed no significant anti-inflammatory activity in acute models. Wheatgrass juice exhibits anti-inflammatory activity in chronic inflammation.

Antimicrobial peptides (AMP) showed a broad spectrum of cytotoxic activity against cancer cells, which could provide a new class of anticancer drugs.In this study, we have evaluated the cytotoxic and apoptotic activity of magainin II, an AMP compared with 5-Fluorouracil (5-FU) in colo320 DM cancer cells, and magainin II cytotoxicity was assessed on vero cell line by MTT.The cells cultured in the presence or absence of magainin II and 5-FU.The magainin II inhibited cell viability in dose dependent manner was quantified by MTT assay.The IC 50 of magainin II was found to be 89.3nM and at this concentration the cytotoxicity was not observed in vero cell line.The cytotoxicity of the magainin II was further confirmed by lactate dehydrogenase (LDH) assay and neutral red uptake assay (NRU).Apoptosis induction was verified by DNA fragmentation analysis.The morphological changes in colo 320 DM cells was measured by DAPI and acridine orange/ethidium bromide staining, showed nuclear condensation, and this substantiates the apoptotic action of magainin II.The cell cycle analysis by FACS revealed that there was a accumulation of cells at G0/G1 phase in magainin II treated cells.Overall, these results revealed that magainin II has selective cytotoxicity in cancer cells and has the potential to induce apoptosis.Magainin II may offer a novel and effective therapeutic candidate in the treatment of colon cancer with potentially low cytotoxic effects on normal cells.These findings highlight the importance of magainin II as a drug capable of exerting an in vitro antitumoral activity by triggering apoptosis. INTRODUCTION:Antimicrobial peptides (AMP) gain increasing attention as potential candidates in cancer therapy and have various advantages such as selective cytotoxicity against cancer cells, active against multidrug-resistance mechanism as well as effects in combination therapy 1 .

The aim of the present study was to compare the lysozyme concentration and candidal count in saliva between HIV-seropositive and HIV-negative individuals, and to correlate the relationship between lysozyme concentrations, candidal count, and CD4 count in HIV patients.A study was conducted in 90 HIV-seropositive patients (subgroups: 1 [CD4 ≥ 500 cells/μL], 2 [CD4 200-499 cells/μL], and 3 [CD4 ≤ 200 cells/μL] and 30 HIV-negative individuals. A total of 6 mL unstimulated saliva was collected and stored at -80°C. Samples were centrifuged and divided into two portions of 600 μL each. One portion was used for the candidal assay and the other for the lysozyme assay using ready-made kits. Student's independent t-test and Karl Pearson correlation coefficient were used for the statistical analysis.There was a significant increase (P < 0.001) in lysozyme levels and the candidal count in the saliva of HIV-positive individuals compared with the HIV-negative individuals. A significant increase (P < 0.004) in the salivary candidal count was observed in the HIV subgroups 1-3. There was a significant negative correlation (P < 0.01) between the CD4 and candidal counts in subgroup 1 (P < 0.02) and between the lysozyme concentration and CD4 count in subgroup 3. There was no correlation between the lysozyme concentration and oral candidal carriage.An association exists between the lysozyme concentration and specific immunity. Yeast colonization serves as a marker of immunodeficiency in HIV disease progression.

Oxidative stress in alcoholic liver disease.

We report on the prooxidant (lipid peroxides) and antioxidant levels (ascorbic acid, reduced glutathione, superoxide dismutate activity) in healthy individuals (30) and patients with cirrhosis (37; 22 alcoholic cirrhosis and 15 non alcoholic cirrhosis). A significant increase in plasma lipid peroxide (P < 0.05) and ascorbic acid (P < 0.01) and a significant decrease in reduced glutathione (P < 0.001) and superoxide dismutase activity (P < 0.05) in haemolysate was observed in cirrhosis patients compared to the control group. A significant decrease in reduced glutathione (P < 0.01) and superoxide dismutase (P < 0.05) activity was also observed when the alcoholic cirrhosis group was compared to non alcoholic group. A significant increase in aspartate transaminase (P < 0.05), gamma glutamyl transaminase (P < 0.01) and aspartate transaminase/alanine transaminase (P < 0.05) ratio was seen in alcoholic cirrhosis group. A significant positive correlation between gamma glutamyl transferase and lipid peroxides (r = 0.48, P < 0.05) was observed in alcoholic cirrhosis.

Antifibrotic effect of Cleome viscosa Linn on carbon tetra chloride (CCl4) induced liver fibrosis.

Liver fibrosis is the excessive accumulation of extracellular matrix proteins including collagen that occurs in most types of chronic liver diseases. This study was carried out to evaluate the antifibrotic effect of ethanolic extract of whole plant of Cleome viscosa Linn. Liver fibrosis was induced by carbon tetra chloride (CCl4) administration in rats. The extent of liver fibrosis was assessed by measuring the level of liver hydroxyproline, thiobarbituric acid and serum enzymes levels. Following CCl4 administration, hydroxyproline, thiobarbituric acid levels were significantly increased and total platelet was decreased and serum enzymes levels were elevated. Treatment with two different doses of the ethanolic extract of Cleome viscosa Linn reduces hydroxy proline, thiobarbituric acid and also the serum enzyme levels. The liver weight that increased following CCl4 administration due to deposition of collagen was reduced by the ethanolic extract Cleome viscosa Linn. Thus the results showed that the ethanolic extract of whole plant of Cleome viscosa Linn found to possesses Antifibrotic effect and that evidenced by the biochemical parameters.

EVALUATION OF SERUM-FREE IRON AND GLYCATED HEMOGLOBIN IN UNCONTROLLED TYPE-II DIABETIC PATIENTS

ABSTRACTObjective: Diabetes mellitus (DM) is a common health problem in the world. Free iron may contribute to the pathogenesis and progress of thisdisease and its complication. Iron causes hyperinsulinemia by decreasing the insulin uptake and metabolism by hepatocytes. Elevated iron stores arecommonly found in insulin resistance. Iron in its free form is known to induce oxidation of biomolecules by producing harmful hydroxyl radicals. Inthis study, we aimed to estimate and compare the serum levels of free iron in diabetes and healthy individuals.Methods: This study included 244 subjects in two groups. Group-I comprised 204 subjects with DM and Group-II comprised 40 healthy subjects. Bloodsugar, free iron, and glycated hemoglobin were analyzed in blood samples using standard kits. The results of all the parameters were expressed asmean±standard deviation. Student t-test was done to assess the statistical significance between two groups. The association between the parameterswas studied by Pearson correlation.Results: In this study, we found a significant increase in serum free iron in Group-I (p<0.01) when compared with Group-II. A significant correlationbetween the serum free iron and glycated hemoglobin (r=0.59; p<0.001) and fasting blood sugar (r=0.43; p<0.001).Conclusion: The elevated serum free iron in uncontrolled diabetes may contribute to oxidative stress which may be associated with complicationsof diabetes.Keywords: Free iron, Glycated hemoglobin, Hyperinsulinemia, Diabetes mellitus.

Association between CYP17A1, CYP19A1, and HSD17B1 Gene Polymorphisms in Hormone Synthesis pathway with Ovarian Cancer Risk

Objective: To investigate the polymorphisms of genes in the steroidogenesis pathway to understand the etiological mechanisms to OC risk in the South Indian population Design: Case-Control Study Setting and Sample: Ovarian cancer cases (200) and healthy individuals (200) from the South Indian population. Methods: All the cases and controls were genotyped for SNPs by using allelic discrimination assay. Main outcome measures: Genetic distribution of SNPs of Steroidogenesis pathway genes in the South Indian population. Results: The observed results for rs743752, the homozygous CC genotype revealed significant association (OR; 1.68; 95%CI, 1.25-2.26; p =<0.05) and the dominant model, recessive model and additive model showed a significant association with an OR of 1.62; 95%CI, 1.09 – 2.42; p = 0.015, OR of 0.29, 95%CI, 0.14 – 0.60; p = <0.001 and OR of 1.68, 95%CI, 1.25 – 2.26); p = <0.001 respectively in cases and controls for OC risk. In rs10046, the heterozygous CT genotype (OR; 1.61; 95%CI 1.06 – 2.43; p = 0.023), the dominant (OR; 1.65; 95%CI, 1.11 – 2.45; p = 0.012) and the additive (OR; 1.46; 95%CI, 1.07 - 1.98; p = 0.015) models were found to be statistically significant. There was no significant association between rs605059 genotypes with ovarian cancer risk. Conclusions: To conclude, results indicated that the polymorphisms of CYP17A1 (rs743572) and CYP19A1 (rs10046) genes are associated with increased risk of ovarian cancer risk in South Indian population.
