Hydrolysis of poly(l‐lactide)/ZnO nanocomposites with antimicrobial activity

ABSTRACT This work investigates the effect of the incorporation of zinc oxide (ZnO) nanoparticles within a poly( l ‐lactic acid) (PLLA) matrix as an approach to speed up the hydrolysis of PLLA film surfaces. Hydrolysis was done by immersing nanocomposite films having 1 wt % of ZnO in 0.25 M sodium hydroxide at 58 °C. This concentration has been selected as it provides the maximum changes of physicochemical properties of hosting PLLA matrix. The evolution of the thermal properties, ultraviolet–visible transparency, wettability, and morphology were monitored at different time points. The amount of carboxylic groups onto PLLA/ZnO surfaces was quantified according to Toludine Blue‐O assay. Hydrolysis was mainly limited to film surfaces, which were grafted by carboxylic groups as a result of the random scission of PLLA ester linkages. The presence of such functional groups decreases the inherent surface hydrophobicity of PLLA at short hydrolysis times. On the contrary, long hydrolyses increase the hydrophobicity as a result of surface nanostructuring induced by the degradation of PLLA to water‐soluble oligomers. Overall, ZnO nanoparticles enable shorter surface modification times and provide a quick approach for the modification on the polarity of polylactide surfaces. The potential of hydrolyzed films as antimicrobial materials was explored using Gram‐negative Escherichia coli as a model. © 2019 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2019 , 136 , 47786.

[Kerion Celsi. A diagnostic problem? Experience with 6 cases].

We report six patients with Kerion Celsi due to Trichophyton verrucosum. Five of the patients were hospitalized with the diagnosis of Staphylococcal abscess. This confusion is due to that highly suppurative and inflammatory nature of the infection. Griseofulvin is the antimicrobial of choice for treatment, associated with imidazolics and corticosteroids to prevent alopecia. The authors suggest that an adequate use of simple microbiological diagnostic tests in the diagnosis of pyodermitis in rural children, may prevent unnecessary hospitalizations and permanent hair loss.

Genomic Variability of Haemophilus influenzae Isolated from Mexican Children Determined by Using Enterobacterial Repetitive Intergenic Consensus Sequences and PCR

ABSTRACT Genomic fingerprints from 92 capsulated and noncapsulated strains of Haemophilus influenzae from Mexican children with different diseases and healthy carriers were generated by PCR using the enterobacterial repetitive intergenic consensus (ERIC) sequences. A cluster analysis by the unweighted pair-group method with arithmetic averages based on the overall similarity as estimated from the characteristics of the genomic fingerprints, was conducted to group the strains. A total of 69 fingerprint patterns were detected in the H. influenzae strains. Isolates from patients with different diseases were represented by a variety of patterns, which clustered into two major groups. Of the 37 strains isolated from cases of meningitis, 24 shared patterns and were clustered into five groups within a similarity level of 1.0. One fragment of 1.25 kb was common to all meningitis strains. H. influenzae strains from healthy carriers presented fingerprint patterns different from those found in strains from sick children. Isolates from healthy individuals were more variable and were distributed differently from those from patients. The results show that ERIC-PCR provides a powerful tool for the determination of the distinctive pathogenicity potentials of H. influenzae strains and encourage its use for molecular epidemiology investigations.

Shape‐memory Polymers for Tissue Engineering Applications

Growing interest on tissue engineering has driven the fast advancement of shape-memory polymers. These materials are able to recover a programmed shape when an external stimulus is applied. This unique functionality has been used to promote tissue defect filling and stimulate cell differentiation and new tissue generation. In this review, we describe the structural specifications that explain the shape recovery in the diverse range of shape-memory polymers, as well as the critical requirements that must been considered in their design to their successful integration and in vivo performance. Moreover, this chapter reviews recent investigations related to tissue engineering applications of shape-memory homopolymers/copolymers, composites, and hydrogels. We reviewed the physicochemical properties of SMPs with a huge variety of structures and trigger stimuli, and their contributions to the tissue regeneration and formation in cellular and in vivo experiments devoted to the repair of different tissue types. New perspectives in the field that allow the effective translation of these materials to the clinic are also highlighted.

An Efficient, PIFA Mediated Approach to Benzo-, Naphtho-, and Heterocycle-Fused Pyrrolo[2,1-c][1,4]diazepines. An Advantageous Access to the Antitumor Antibiotic DC-81

The synthesis of a series of optically pure benzo-, naphtho-, and heterocycle-fused pyrrolo[2,1-c][1,4]-diazepin-5,11-dione derivatives starting from l-proline methyl ester is presented. The synthetic plan includes an aroylation step at the proline nitrogen followed by transformation of the ester residue into a N-methoxyamide group. The subsequent key cyclization step embraces the PIFA mediated formation of a N-acylnitrenium intermediate and its succeeding intramolecular trapping by the aromatic ring. The presented general approach solves the need of starting from not very accessible amino (or a related functionality) aromatic starting materials, and its effectiveness is demonstrated in the synthesis of the antitumor antibiotic DC-81.

A concentration-dependent endocytic trap and sink mechanism converts Bmper from an activator to an inhibitor of Bmp signaling

Bmper, which is orthologous to Drosophila melanogaster crossveinless 2, is a secreted factor that regulates Bmp activity in a tissue- and stage-dependent manner. Both pro- and anti-Bmp activities have been postulated for Bmper, although the molecular mechanisms through which Bmper affects Bmp signaling are unclear. In this paper, we demonstrate that as molar concentrations of Bmper exceed Bmp4, Bmper dynamically switches from an activator to an inhibitor of Bmp4 signaling. Inhibition of Bmp4 through a novel endocytic trap-and-sink mechanism leads to the efficient degradation of Bmper and Bmp4 by the lysosome. Bmper-mediated internalization of Bmp4 reduces the duration and magnitude of Bmp4-dependent Smad signaling. We also determined that Noggin and Gremlin, but not Chordin, trigger endocytosis of Bmps. This endocytic transport pathway expands the extracellular roles of selective Bmp modulators to include intracellular regulation. This dosage-dependent molecular switch resolves discordances among studies that examine how Bmper regulates Bmp activity and has broad implications for Bmp signal regulation by secreted mediators.

Characterization and Optimization of the Alkaline Hydrolysis of Polyacrylonitrile Membranes

There is currently an increasing interest in the development of polyacrylonitrile (PAN)-based membranes with new and enhanced properties which are of special importance in the processes of pervaporation, purification, and water treatment. Thus, the optimization of the functionalization of PAN membranes and its effect on their morphology, hydrophilicity, and mechanical properties plays an essential role in a wide range of applications. In this paper, the alkaline hydrolysis of asymmetric PAN membranes was investigated in order to get carboxyl-enriched surfaces that are of a great interest for more demanding subsequent modifications. The process was monitored using –C=NH intermediate bonds, which could be observed during the hydrolysis reaction by X-Ray photoelectron spectroscopy (XPS) and Fourier-transform infrared spectroscopy (FTIR) before the formation of carboxyl and amide groups. The amount of introduced carboxylic acid groups could be determined by thermogravimetric analysis (TGA) and by the interaction with toluidine blue O (TBO) dye. Hydrolysis was revealed as a simple way to modulate hydrophilicity (decreasing contact angle from 60 to 0° for reaction times from 0–3 h) and the mechanical properties of PAN membranes.
