Effect of nano-curcumin supplementation on cardiometabolic risk factors, physical and psychological quality of life, and depression in patients with coronary slow flow phenomenon: a randomized double-blind clinical trial

Extensive evidence has suggested the cardio-protective properties of the polyphenol curcumin. The aim of this study was to investigate the effects of a highly bioavailable curcumin supplement on cardiometabolic risk factors, health-related quality of life, and depression in patients with coronary slow flow phenomenon (CSFP). This randomized double-blind placebo-controlled clinical trial was conducted in 42 patients with CSFP (age 35–70 years, 25 ≤ body mass index < 40 kg/m2). Patients received either 80 mg/day nano-curcumin or placebo for 12 weeks. Serum levels of visfatin, high-sensitivity C-reactive protein (hs-CRP), and glycemic indices were measured before and after the intervention. The short form 36-item quality of life (SF-36) and Beck's Depression Inventory-II (BDI-II) questionnaires were assessed, as well. No significant improvements were observed in circulating hs-CRP and visfatin following the intervention. A significant increase was observed in pre- to post-fasting blood glucose (− 0.9 ± 12.2 vs. 7.7 ± 12.4 mg/dl, p = 0.02) and hemoglobin A1C (− 0.1 ± 0.8 vs. 0.5 ± 0.8%, p = 0.04) levels, in the placebo compared with the intervention group. Physical (8.2 ± 8.1 vs. − 1.2 ± 6.5, p < 0.001) and mental (6.8 ± 11.8 vs. − 1.1 ± 10.4, p = 0.02) component summary scores were significantly improved in the nano-curcumin than the placebo group. Additionally, the number of patients with lower degrees of depression was significantly better in the intervention than the placebo group following the supplementation (p = 0.046). Curcumin supplementation prevented deterioration of glycemic control and improved physical and psychological quality of life and depression in patients with CSFP. Effect of glatiramer acetate on short‐term memory impairment induced by lipopolysaccharide in male mice

Abstract Glatiramer acetate ( GA ) demonstrates neuroprotective, neurogenesis, and anti‐inflammatory properties. This study examines the probable protective effect of acute GA on lipopolysaccharide ( LPS )‐induced memory impairment in male mice and further explores which routes of administration [subcutaneous (s.c.) or intracerebroventricular (i.c.v.)] exert optimum effect. Memory performance was evaluated in two‐trial recognition Y‐maze and passive‐avoidance tasks evaluating special recognition memory and fear memory, respectively. Memory impairment was induced by LPS [100 μg/kg, intraperitoneally (i.p.)], 4 h before training. In Y‐maze, GA (10, 2.5, 0.625, 0.153, and 0.03 mg/kg, s.c.; 250 μg/mouse; i.c.v.) was administered 10 min following LPS , and special memory was assayed in Y‐maze apparatus. In passive avoidance, LPS (100, 250 μg/kg; i.p.) was injected 4 h before receiving foot shock, and GA (10, 2.5; s.c.) or (250 μg/mouse; i.c.v.) was administered 4 h before the shock. Following 24 h, the fear memory was evaluated. Memory impaired significantly following LPS (100, 250 μg/kg; i.p.) in Y‐maze and passive‐avoidance tasks, P < 0.001 and P < 0.05, respectively. The data revealed that GA (250 μg/mouse, i.c.v.) and GA (10, 2.5 mg/kg; s.c.) in Y‐maze reversed memory impairment ( LPS 100 μg/kg, i.p.) ( P < 0.01). In passive‐avoidance task, GA (2.5, 10 mg/kg; s.c.) reversed LPS ‐induced impairment and the mice showed significantly longer latency times during the retention trial ( P < 0.01). GA improved memory impairment both centrally and systemically. It improved spatial recognition memory increasing the average time in the novel arm and improved fear memory increasing latency time. GA administration improved memory impairment profoundly through both systemic and central routs.

Preparation of All-Trans-Retinoic Acid-Loaded mPEG-PLGA Nanoparticles Using Microfluidic Flow-Focusing Device for Controlled Drug Delivery

In recent years, microfluidic devices present unique advantages for the development of a new generation of nanoparticle synthesis method compared to bulk methods. In this study, we report a microfluidic flow-focusing method for the production of all trans retinoic acid (ATRA)-loaded methoxy poly(ethylene glycol)-poly(lactide-coglycolide) (mPEG-PLGA) nanoparticles (NPs). Box–Behnken experimental design (BBD) was applied to optimize of formulation ingredients and process conditions with minimum particle size, maximum drug loading% (DL%) and encapsulation efficiency% (EE%). Polymer concentration, drug concentration and flow rates of solvent (S) and antisolvent (AS) were selected as independent variables. Based on optimization strategy, minimum particle size achieved shows average (SD) particle size of [Formula: see text][Formula: see text]nm with DL of [Formula: see text][Formula: see text]wt.% and EE of [Formula: see text][Formula: see text]wt.%, respectively. While maximum DL has been reported to be [Formula: see text][Formula: see text]wt.% with particle size of [Formula: see text][Formula: see text]nm and EE of [Formula: see text][Formula: see text]wt.%, respectively. Moreover, the results have shown that the AS/S ratio represents the most significant effect on particle size. Indeed, increasing the AS flow rate directly results in generating smaller particles. The AS/S ratio represents the least significant effect on DL%, such that, at fixed flow rates, higher DL was observed at high concentration of drug and lower concentration of polymer. In conclusion, optimization of the ATRA-loaded mPEG-PLGA NPs by BBD yielded in a favorable drug carrier for ATRA that could provide a new treatment modality for different malignancies.

Toxicity assessment of nanosilver wound dressing in Wistar rat.

Antibiotic resistance to microorganisms is one of the major problems faced in the field of wound care in burns patients. Silver nanoparticles have come up as potent antimicrobial agent and are being evaluated in diverse medical applications ranging from silver based dressings to silver coated medical devices. We aimed in present study to test the release of nanosilver from nanosilver wound dressing and compare the dermal and systemic toxicity of nanosilver dressings in a repeated dose (21 days) model. Under general anesthesia, a limited standard 2nd degree burns were provided on the back of each rat in all treatment, negative control (simple dressing) and 5% silver nitrate groups, each contained 5 male wistar rats. According to the analysis made by atomic absorption spectrometry, the wound dressings released 0.599 ± 0.083 ppm of nanosilver during first 24 hrs of study. Daily observations were recoded and wounds were covered with new dressings each 24 hrs. Burn healing was observed in nanosilver wound dressing group in shorter time periods than the control groups. In toxicity assessment, this dressing didn't cause any hematological and histopathological abnormalities in treatment group but biochemical studies showed significant rise of plasma transaminase (ALT) at the endpoint (21 days) of the study (P=0.027). Portal mononuclear lymphoid and polymorphonuclear leukocyte infiltrations in three to four adjacent foci were recognized around the central hepatic vein in treatment group. Mild hepatotoxic effects of nanosilver wound dressing in wistar rat emphasize the necessity of more studies on toxicity potentials of low dose nanosilver by dermal applications.

Effect of laminated hydroxyapatite/gelatin nanocomposite scaffold structure on osteogenesis using unrestricted somatic stem cells in rat

Bone matrix consists of two major phases at the nanoscale: organic and hydroxyapatite. Nanotechnology as a diverse and interdisciplinary area of research has the capacity to revolutionise many areas of applications such as bone tissue engineering. Nanohydroxyapatite/gelatin composite has higher osteoblast attachment and proliferation than micro-sized ones, and shorter culturing period and lower cell seeding density compared to pure gelatin. A nanostructured scaffold was fabricated by three methods for bone repair using nanohydroxyapatite and gelatin as the main components. Its biocompatibility, alizarin red test on the 14th and 21st days, gene expression on the 21st day in in vitro using and histomorphometry after 4 and 8 weeks post-implantation in the rat were investigated. Cultured unrestricted somatic stem cells used for in vitro study showed an excellent level of cell attachment to the scaffold. Cells induced more osteoblast differentiation on the scaffold than in 2D cell culture. Osteoblast differentiation and bone regeneration results of in vitro and in vivo investigation on scaffold were extremely significant, better than control and treatment groups. These effects could be attributed to the shape and size of nanoHA particles and good architecture of the scaffold. The results confirm the feasibility of bone regeneration using synthesised scaffold as a temporary bone substitute.

A Survey on Aflatoxin M1 in Raw Milk of Fariman City, Khorasan Province, Iran

Background: Although milk is the valorous source of nutrition, consumption of aflatoxin M1 (AFM1) contaminated milk induces problems to human health. AFM1 is found in milk when dairy animals are fed with AFB1-contaminated feedstuff. AFM1 remains stable during milk pasteurization and storage as well as during the preparation of various dairy products. Objectives: The aim of the current study was to evaluate AFM1 contamination in raw milk of Fariman city. Materials and Methods: During the summer (July to September, 2012), 45 samples of raw milk were collected randomly from Fariman, Iran. The samples were analyzed with a commercial competitive enzyme-linked immunosorbent assay (ELISA) kit. Results: AFM1 was detected in all milk samples. The mean concentration of AFM1 was 11.61 ± 0.72 ng/L with a range of 6.3-23.3 ng/L. None of the samples had levels exceeding the maximum tolerance limit (50 ng/L) accepted by the European Union. Conclusions: AFM1 contamination of milk is not a concern in this city. However, dairy cattle feed samples of various livestocks must be regularly checked for aflatoxin and the storage conditions of feeds should be strictly controlled.

Cloning, Expression and Purification of Penicillin Binding Protein2a (PBP2a) from Methicillin Resistant Staphylococcus aureus: A Study on Immunoreactivity in Balb/C Mouse.

Staphylococcus aureus (S. aureus) is a major nosocomial pathogen and the infection with this organism in human is increasing due to the spread of antibiotic resistant strains. One of the resistance mechanisms of S. aureus comprises modification in binding proteins to penicillin. Vaccine strategy may be useful in controlling the infections induced by this organism. This study aimed at developing and producing the recombinant protein PBP2a as a vaccine candidate and evaluating the related humoral immune response in a murine model.A 242 bp fragment of mecA gene was amplified by PCR from S. aureus COL strain and then cloned into prokaryotic expression vector pET-24a. For expression of recombinant protein, pET24a-mec plasmid was transformed into competent E. coli BL21 (DE3) cells. Recombinant protein was over expressed with 1 mM isopropythio-β-D-galctoside (IPTG) and purified using Ni-NTA agarose. SDS-PAGE and western blotting were carried out to confirm protein expression. For immunization of experimental groups, Balb/c mice were injected subcutaneously with 20 µg of recombinant PBP2a three times with three weeks intervals. The sera of experimental groups were collected three weeks after the last immunization and then specific antibodies were evaluated by ELISA method.Successful cloning of mecA was confirmed by colony-PCR, enzymatic digestion, and sequencing. SDS-PAGE and western blot analysis showed that recombinant protein with molecular weight of 13 kDa is over expressed. In addition, high titer of specific antibody against PBP2a in vaccinated mice was developed as compared with the control group and confirmed the immunogenicity of the vaccine candidate.Results suggest that PBP2a recombinant induced specific antibodies and can be used as Staphylococcal vaccine candidate after further studies.
