Gigantol Alleviates IL-1β-Induced Inflammation and Catabolism in Mouse Osteoarthritis via PI3K/Akt/NF-κB Pathways In Vivo and In Vitro

Abstract Osteoarthritis (OA), a prevalent disabling disease, is characterized by irreversible cartilage degradation and persistent inflammation. The etiology as well as pathogenesis of OA are not completely unclear and need further investigation. Gigantol, is a bibenzyl derivative extracted from Dendrobium plants and has been found exhibit multiple effects such as anti-inflammatory effects. Nevertheless, the biological function of gigantol on osteoarthritis (OA) is still uncertain. This study aimed at examining the anti-inflammatory effects and latent mechanisms of gigantol in IL-1β-mediated OA progression. In vitro, we identified that gigantol treatment suppressed tumor necrosis factor-alpha (TNF-α), nitric oxide (NO), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), inducible nitric oxide synthase (iNOS) and interleukin-6 (IL-6) in interleukin-1 beta (IL-1β) mediated mouse OA chondrocytes. Gigantol was also shown to dose dependently downregulate the metalloproteinase 13 (MMP13) as well as thrombospondin motifs 5 (ADAMTS5) levels. Moreover, IL-1β-mediated AKT and PI3K phosphorylation as well as NF-κB activation were inhibited by gigantol. Meanwhile, in vivo, we detected that gigantol treatment inhibited degradation of the cartilage degradation and lowered the Osteoarthritis Research Society International scores (OARSI) in OA mouse. Therefore, gigantol is a promising therapeutic option for OA.

DNA-Mediated Graphene Oxide/Platinum Nanozymes for Label-Free Colorimetric Detection of Hg²⁺

Mercury ions (Hg2+) have persistent bioaccumulation and high toxicity, which can severely damage the ecological system and human health. Therefore, sensitive methods are required for Hg2+ analysis. Herein, a DNA-mediated growth strategy was developed for the synthesis of platinum nanoparticles (PtNPs) on the surface of graphene oxide (GO). The resulting GO-DNA-PtNPs displayed excellent peroxidase-mimicking activity in a sequence-dependent manner. The peroxidase-mimicking activity could be significantly inhibited by Hg2+ because of the specific trapping of Hg2+ by thymine bases (T–Hg2+–T) and in situ reduction of Hg2+ by PtNPs on the GO surface. Under optimal conditions, there was a good linear range from 1 pM to 1 μM with a detection limit of 2.2 pM for Hg2+ analysis. This sensor demonstrated excellent analytical performance for assessing Hg2+ in complex lake waters. This work provides an effective strategy for the preparation of PtNP-based nanozymes and broadens the applications of PtNPs for biosensing.

High-Performance Supercapacitor Electrode Materials from Cellulose-Derived Carbon Nanofibers

Nitrogen-functionalized carbon nanofibers (N-CNFs) were prepared by carbonizing polypyrrole (PPy)-coated cellulose NFs, which were obtained by electrospinning, deacetylation of electrospun cellulose acetate NFs, and PPy polymerization. Supercapacitor electrodes prepared from N-CNFs and a mixture of N-CNFs and Ni(OH)2 showed specific capacitances of ∼236 and ∼1045 F g–1, respectively. An asymmetric supercapacitor was further fabricated using N-CNFs/Ni(OH)2 and N-CNFs as positive and negative electrodes. The supercapacitor device had a working voltage of 1.6 V in aqueous KOH solution (6.0 M) with an energy density as high as ∼51 (W h) kg–1 and a maximum power density of ∼117 kW kg–1. The device had excellent cycle lifetime, which retained ∼84% specific capacitance after 5000 cycles of cyclic voltammetry scans. N-CNFs derived from electrospun cellulose may be useful as an electrode material for development of high-performance supercapacitors and other energy storage devices.

Preparation of Copper (II) Containing Phosphomolybdic Acid Salt as Catalyst for the Synthesis of Biodiesel by Esterification

Copper (II) containing phosphomolybdic acid (PMA) catalysts were synthesized by ion exchange method and characterization using various physico-chemical techniques such as X-ray diffraction (XRD), fourier transform infrared spectroscopy (FT-IR), thermogravimetric (TG) and scanning electron microscopy (SEM). The characterization results showed that the Keggin ions were retained in the catalysts and possessed well thermal stability. The catalytic esterification of lauric acid with methanol could be easily achieved about 78.7% conversion under optimum condition, the catalyst also contributed to the stability of the catalyst in which it can be reused for a certain time. This study demonstrated an alternative approach to biodiesel production with high efficiency by Cu (II) ion exchanged phosphomolybdic acid catalyst in the esterification catalytic.
