Glycolipids and fatty acids of two dog kidney cell lines

Glycolipid and fatty acid compositions were studied in whole cells and plasma membranes from two dog kidney cell lines (Madin-Darby and SV40-transformed cells) grown in monolayer and suspension cultures. Glycolipids, which account for 5% or less of the total lipids in dog kidney cells, were substantially increased in plasma membranes relative to whole cells. Sialoglycolipids more complex than a Tay-Sachs-like ganglioside were not found in any whole-cell or plasma-membrane preparation of this study. Dog kidney cells transformed by SV40 virus contained primarily a less complex sialoglycolipid, haematoside. Neutral glycolipids comprised 26-43% of the total glycolipid content in Madin-Darby preparations, whereas in transformed cells and membranes neutral glycolipids constituted only 1-22% of the total glycolipid content. Ceramide trihexoside was found in Madin-Darby cultures, but not in transformed cultures. The values for short-chain fatty acids from neutral glycolipids and for saturated fatty acids were generally higher than the values for these fatty acids in calf serum.

Canine kidney cells. II. Neutral lipids, phospholipids and fatty acids of Madin-Darby canine kidney cells in suspension and monolayer culture.

Differences in lipid composition and the effects of monolayer and suspension culture were studied in Madin-Darby canine kidney cells and their plasma membranes isolated by the Warren fluorescein-mercuric acetate technique. Unesterified cholesterol was the major neutral lipid extracted from the whole cells and plasma membranes of cultures except for suspension-grown whole cells which had triglyceride as their major neutral lipid. Triglyceride increased about 7- to 20-fold and glycerol ethers increased about 3- to 20-fold in suspension grown whole cells and their plasma membranes compared to those from monolayer cultures. Total quantities of phospholipids are similar in whole cells of both cultures. Plasma membranes from monolayer cultures have about 1.4-fold more total phospholipids than those from suspension cultures. The distribution of fatty acids in the plasma membranes and whole cells generally follow the composition of calf serum used in the culture medium. However, the phospholipids of plasma membranes from monolayer cultures had substantially higher values for C18 unsaturated fatty acids and somewhat lower values for palmitoleic acid than the values for the fatty acids in calf serum. Also, the phospholipids of suspension-grown whole cells had substantially higher values for lauric acid and lower values for C18 unsaturated fatty acids when compared to calf serum.

Characterization of the Membranes of Thermoplasma acidophilum

Thermoplasma acidophilum grows optimally under aeration at 59 C and pH 2. Both intact cells and membranes disaggregate below pH 1 and above pH 5, producing no sedimentable particles. Increase in ionic strength at pH 5 or below results in cellular lysis and membrane disaggregation. Membranous components produced by lysis at alkaline pH reaggregate upon reduction of both pH and ionic strength. Osmotic environment plays little role in cellular stability. Membranes prepared by sonic lysis at pH 5 exhibit vesicular structures and are composed of multiple proteins. Although the amino acid composition of the membrane proteins is similar to other mycoplasmal membranes, the number of free amino and carboxyl groups is less than half of those in Acholeplasma . Reduction of the number of free carboxyl groups results in membrane stabilization over a wide range of pH. Increase in the number of free amino groups reverses the stability of membranes relative to pH. Acidophily in Thermoplasma can be related to a significant reduction in repulsing negative charges on the membrane proteins.

Canine kidney cells: III. Neutral lipids, phospholipids and fatty acids of primary canine kidney cells in monolayer culture.

Neutral lipids, phospholipids and fatty acids were studied in monolayer grown primary canine kidney whole cells and their plasma membranes which were isolated by the Warren fluorescein-mercuric acetate technique. Triglyceride and unesterified cholesterol were the predominant neutral lipids of whole cells, while cholesterol ester, free fatty acid, and unesterified cholesterol were the major neutral lipids of plasma membranes. The total neutral lipid value is about 2-fold greater in plasma membranes than in whole cells. Phosphatidyl ethanolamine and physophatidyl choline are the major phospholipids of whole cells and plasma membranes. The total phospholipid value is almost 1.8-fold greater in plasma membranes than in whole cells. The fatty acid profiles of both whole cells and plasma membranes differ from that of calf serum used in the growth medium. The amount of unsaturated fatty acids is considerably lower while the amount of saturated fatty acids is higher than the corresponding values obtained in calf serum.

The formation of serine from glycine and formaldehyde by cell free extracts of Clostridium acidi-urici.

Cells of Clostridium acidi-urici which were grown in a medium containing uric acid were harvested, disrupted by sonication and centrifuged. After centrifugation the supernatant which served as the cell free extract was used to study the synthesis of serine from 2-14C glycine and formaldehyde. Serine was isolated from the reaction mixture by column chromatography. After identification by paper chromatography, serine was degraded carbon by carbon to locate the position of the labelled carbon. Radioactivity was confined almost exclusively to the alpha carbon of serine which was derived from the alpha carbon of glycine. Formaldehyde, therefore, binds at the alpha carbon of glycine to form serine. Conversion of serine to pyruvate was prevented by adding EDTA to the reaction mixture.
