Perfluorocarbon Nanoparticles Incorporating Ginkgolide B: Artificial O₂ Carriers with Antioxidant Activity and Antithrombotic Effect

Abstract Ischemic stroke primarily leads to insufficient oxygen delivery in ischemic area. Prompt reperfusion treatment for restoration of oxygen is clinically suggested but mediates more surging reactive oxygen species (ROS) generation and oxidative damage, known as ischemia‐reperfusion injury (IRI). Therefore, the regulation of oxygen content is a critical point to prevent cerebral ischemia induced pathological responses and simultaneously alleviate IRI triggered by the sudden oxygen restoration. In this work, we constructed a perfluorocarbon (PFC)‐based artificial oxygen nanocarrier (PFTBA‐L@GB), using an ultrasound‐assisted emulsification method, alleviates the intracerebral hypoxic state in ischemia stage and IRI after reperfusion. The high oxygen solubility of PFC allows high oxygen efficacy. Furthermore, PFC has the adhesion affinity to platelets and prevents the overactivation of platelet. The encapsulated payload, ginkgolide B (GB) exerts its anti‐thrombosis by antagonism on platelet activating factor and antioxidant effect by upregulation of antioxidant molecular pathway. The versatility of the present strategy provides a practical approach to build a simple, safe, and relatively effective oxygen delivery agent to alleviate hypoxia, promote intracerebral oxygenation, anti‐inflammatory, reduce intracerebral oxidative stress damage and thrombosis and caused by stroke.

Vibrational-Energy Redistribution in Single-Atom Manipulation by Scanning Tunneling Microscope

A pairwise phenomenon is observed when we attempt to extract a single H atom from a monohydride Si(100)-2×1:H surface. This is attributed to the final vibrational excitation induced by the tunneling electrons and the desorption of two H atoms on one Si dimer simultaneously as an H 2 molecule. The pair effect is thought to be direct evidence of vibrational-energy transfer on the surface.

Association between a TCF4 Polymorphism and Susceptibility to Schizophrenia

The basic helix-loop-helix (bHLH) transcription factor 4 ( TCF4 ) had been identified as a susceptibility gene associated with schizophrenia (SCZ) by GWAS, but inconsistent results have been found in other studies. To validate these findings and to reveal the effects of different inheritance models, rs2958182, rs1261085, rs8766, and rs12966547 of the TCF4 gene were genotyped in the Northwest Han Chinese population (448 cases and 628 controls) via a multiplex polymerase chain reaction SNPscan assay. Single SNP, genotype, and association analyses with three different models were performed. We observed genotype and allele distributions of four SNPs that showed nonsignificant associations in the Northwest Han Chinese population. However, published datasets (51,892 cases and 68,498 controls) were collected and combined with our experimental results to ascertain the association of the TCF4 gene SNPs and SCZ, which demonstrated that rs2958182 (P=0.003) was a significant signal based on a systematic meta-analysis. To clarify the biological role of rs2958182, it is important to improve the understanding of the pathophysiology of SCZ.

Do Electrospun Polymer Fibers Stick?

Adhesion between electrospun polycaprolactone (PCL) fibers was directly measured in a cross-cylinder geometry using a nanoforce tensile tester. The surface roughness of fibers was determined by an atomic force microscope (AFM), and the structural factors were characterized by differential scanning calorimeter (DSC) and wide-angle X-ray diffraction (WAXD). "Pull-off" force was found to be in the order of 10−6 N, and the adhesion energy was 190 ± 7 mJ/m2. Adhesion increases with decreasing fiber radius. The experimental data are analyzed by the classical Johnson−Kendall−Roberts (JKR) contact mechanics model. The study provides fruitful insights into future development of bio-inspired adhesives and devices.
