The pandemic caused by a new strain of SARS-CоV-2 coronavirus has swept the whole world, however, despite the developed strategic directions for the treatment of coronavirus infection and intensive research in all countries, effective methods for treating this severe pathology have not yet been created. The list of drugs against COVID-19 practically does not use compounds that affect the renin-angiotensin system, in the functioning of which the ACE2 coronavirus binding receptor plays a central role. It is assumed that the virus, causing a decrease in the density of ACE2 receptors, leads to disruption of RAS activity. This review presents current research on the response of the immune system to infection with the SARS-CoV-2 virus, describes adaptive and innate cellular mechanisms, and describes a number of predictors of severe COVID-19. To write this review, a search was made in the PubMed database and the scientific electronic library eLibrary.ru. The selection of articles was carried out manually with the main goal of synthesizing data and describing the mechanisms of influence of SARS-CoV-2 on the renin-angiotensin system, and, as a result, on the activation of the adaptive and innate immune response. This review includes 53 publications, including methodological recommendations of the Ministry of Health of the Russian Federation, data from ongoing clinical trials and patents. Data from selected scientific sources were structured and visualized.

YB-1 Protein Prevents Age Decline in Plasma Estradiol in 5xFAD Transgenic Aging Female Mice

Alzheimer’s disease (AD) is an incurable neurodegenerative disease that is the main cause of dementia in the elderly. When looking for new treatments for AD, attention was drawn to the multifunctional Y-box-binding protein 1 (YB-1). Previously, we revealed a positive effect of intranasal administration of YB-1 on learning and spatial memory, along with a decrease in the content of cerebral β-amyloid and the intensity of plaque initiation, with an improvement in the survival of neurons in the cortex and hippocampus of male AD mice. However, AD affects women twice as often as men, so it is of great interest to study the effects of YB-1 on aging females. Estrogens and androgens are necessary for the maintenance of cognitive function during aging and, apparently, may prevent the development of AD. In this work, peripheral levels of estradiol (E2) and cytokines were studied after intranasal administration of YB-1 to aging female 5xFAD transgenic mice and control non-transgenic animals. In intact aging animals of both groups, a violation of the estrous cycle and a decrease in the level of E2 in blood plasma were revealed. Mice treated with YB-1 did not show a characteristic age-related decrease in plasma E2 levels. The introduction of YB-1 did not affect the peripheral level of cytokines. Thus, a novel, previously undescribed effect of YB-1 on plasma E2 levels in aging female mice is shown. These data indicate that YB-1 may be a promising compound in the prevention and treatment of neurodegenerative diseases. However, further experiments are needed to gain insight into the detailed mechanisms of YB-1 action.

Germinal proto-mitochondria from rat liver

A number of properties of the smallest (less than 0.2 μm) germinal proto-mitochondria (PRMC) from rat liver have been studied. These PRMC were obtained by filtering the light fraction of hepatic mitochondria (MC) through calibrated millipore membranes. Germinal PRMC contain in general the same proteins as MC. However, they have the reduced content of flavoproteins and zero cytochrome oxidase. Germinal PRMC, in contrast to MC, almost does not contain the "aging pigment" - lipofuscin. They have DNA; the DNA/protein ratio in them is much higher than in MC, i.e. they are poor in protein. The obtained results support the earlier assumption that MC in specialized animal cells can arise from germinal PRMC - particles smaller than 0.2 μm containing DNA. It is assumed that the DNA molecules enter to cytoplasm during degradation of old MC serves as a seed for the formation of PRMC (with the connection of nuclear DNA).

Phase synchronization of oscillations in cardiovascular and respiratory systems in humans

Phase synchronization between blood flow oscillations of left and right forearm skin sites, heart rate variability (HRV) and breath rate were studied from healthy volunteers at rest. The degree of synchronization between the phases of the analyzed signals was estimated from the value of the wavelet phase coherence. High medians of values of phase wavelet coherence function were obtained for the endothelial, neurogenic, myogenic and cardiac intervals. Significant phase synchronization were demonstrated between HRV and skin blood flow oscillations in both left and right forearms in a wide frequency range from 0.04 to 0.4 Hz. Six participants exhibited low phase synchronization (< 0.5) between the breath rate and HRV, while nine participants had high phase synchronization (> 0.5). This distribution was not affected by the sex or sympathovagal status of volunteers. Participants with low phase synchronization between breath rate and HRV featured low phase synchronization (< 0.5) between breath rate and blood flow oscillations in both forearms. Contrariwise, in subjects with high phase synchronization between respiratory rhythm and HRV both low and high phase synchronization between breath rate and blood flow oscillations in both forearms was observed. The results obtained allow us to suggest that the organism possesses a mechanism mediating the synchronization of blood flow oscillations in the skin microvasculature with all other periodical processes across the cardiovascular system, in particular, with HRV and breath rate over a wide frequency range.

Unique Properties of Synaptosomes and Prospects for Their Use for the Treatment of Neurodegenerative Diseases (Review)

Alzheimer’s disease (AD) is a severe neurodegenerative disease that affects millions of people around the world. The increasing prevalence of AD correlates with increasing life expectancy and aging populations in developed countries. Since AD is a multifactorial disease and includes various pathological processes, such as: synaptic dysfunction, neuroinflammation, oxidative stress, protein misfolding, etc., an integrated approach aimed simultaneously at several targets may be effective and slow down the progression of the disease. Cell therapy and its further development in the form of transplantation of cellular vesicles and especially mitochondria are a very promising approach for the treatment of neurodegeneration. The use of synaptosomes, due to the uniqueness of their content, may become a new stage in the development of complex therapy for neurodegenerative diseases and AD in particular. This review discusses the preparation and composition of synaptosomes, as well as the possibilities and advantages of their use as transporters for the delivery of synaptic mitochondria and other biologically active substances to the brain.

Mitochondrial Fragmentation as a Key Driver of Neurodegenerative Disease

Mitochondrial form and function are intricately linked through dynamic processes of fusion and fission, and disruptions in these processes are key drivers of neurodegenerative diseases, like Alzheimer’s. The inability of mitochondria to transition between their dynamic forms is a critical factor in the development of pathological states. In this paper, we focus on the importance of different types of mitochondrial phenotypes in nervous tissue, discussing how mitochondria in Alzheimer's disease are “stuck” in certain patterns and how this pattern maintains itself. Understanding the specific roles and transitions between mitochondrial forms, including tiny, networked, and hyperfused, is crucial in developing new therapies aimed at restoring mitochondrial homeostasis. By targeting these dynamics, we may be able to intervene early in the disease process, offering novel avenues for preventing or treating neurodegeneration.
