Occurrence of plant hormones in cells of the phototrophic purple bacterium Rhodospirillum rubrum 1R

Plant hormones from biomass of the purple non-sulfur bacterium Rhodospiririllum rubrum were isolated for the first time. These compounds show high physiological activities (300–330%) in the cytokinin bioassay. All three detected cytokinins are adenine derivatives, according to spectral analysis. One of them was identified as 6-(4-hydroxy-3-methyl-2-trans-2-bytenylamino)-9-β-D-ribofuranosylpurine (zeatinriboside) as shown by thin-layer chromatography and reversed-phase high-performance liquid chromatography. The possible functions of bacterial cytokinins are also discussed.

Effect of 4-hydroxyphenethyl alcohol on growth and adaptive potential of barley plants under optimal and soil flooding conditions.

Barley (Hordeum vulgare L. cv. “Rudzik”) seeds were soaked in aqueous 10 -6 М 4-hydroxyphenethyl alcohol (4-HPEA) to estimate its influence on seed germination, growth of seedlings, and their adaptive potential under soil flooding conditions. The adaptive potential of the plants was estimated by the concentration of thiobarbituric acid reactive substances (TBARs) and the activity of guaiacol-dependent peroxidase (GPX). It was shown that the 4-HPEA had a stimulatory action on seed germination, shoot and root growth, and biomass production, expressed to a greater extent in the early stage of plant development. Pre-sowing soaking of barley seeds in aqueous 4-HPEA solution increased plant tolerance to the effect of soil flooding, too.

Novel peripheral light-harvesting (LH) complex designated as LL LH2 was isolated along with LH4 complex from Rhodopseudomonas palustris cells grown under low light intensity (LL). FPLC-MS/MS allowed to reveal PucABd and PucBabc apoproteins in LL LH2 complex, which is different from previously described LH4 complex containing PucABd, PucABa and PucBb. The main carotenoids in LL LH2 complex were rhodopin and 3,4-didehydrorhodopin. Three-dimensional modeling demonstrated which amino acid residues of all the β -subunits could interact with carotenoids (Car) and bacteriochlorophyll a (BChl a). Analysis of amino acid sequences of α -subunits of both LL complexes showed presence of different C-terminal motifs, IESSVNVG in α a subunit and IESSIKAV in α d subunit, in the same positions of C-termini, which could reflect different retention force of LL LH2 and LH4 on hydroxyl apatite, facilitating successful isolation of these complexes. Differences of these LL complexes in protein and carotenoid composition, in efficiency of energy transfer from Car to BChl a, which is two times lower in LL LH2 than in LH4, allow to assign it to a novel type of light-harvesting complex in Rhodopseudomonas palustris .
