Comparative proteomic analysis on human L-02 liver cells treated with varying concentrations of trichloroethylene

To determine the differential proteomic expressions in human L-02 liver cells induced by varying concentrations of trichloroethylene (TCE), comparative proteomic analysis was performed on human L-02 liver cells which were treated with varying concentrations of TCE. According to the result of MTT test, we designed four different groups, in which the cells were treated with 0 μM (control group), 3, 10 or 40 μM TCE for 24 h, respectively. Comparative analysis of approximately 800 spots resolved by two-dimensional gel electrophoresis (2DE) in the soluble proteomes of L-02 cells from the four different groups resulted in 10 differential proteins. To identify the differential spots, matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) was carried out; if the results from the tool were insufficient, tandem MS (MALDI-TOF-TOF-MS) was then performed. The raw data of peptide mass fingerprints (PMFs) and MS/MS spectra were searched against the IPI human data base for exact matches. Then western blot was employed to verify the result of proteomic analysis, the following result confirmed that the results of proteomic analysis were reliable. These results might provide an insight into the underlying mechanism of TCE intoxication and find biological markers for diagnosis and therapy of TCE-induced diseases.

Construction and Identification of shRNA Eukaryotic Expression Vector Targeting Annexin A3

[Objective] The aim of this paper was to construct and identify the siRNA eukaryotic expression vector targeting annexin A3.[Method]s Four shRNAs were designed according to the coding sequence of annexin A3 gene,and cloned into the downstream of H1 promoter of psiRNA-hH1-neo.The constructed recombinant was analyzed and identified by Ase Ⅰendonuclease digestion and DNA sequencing.[Result]s The constructed psiRNA plasmid digested with Ase Ⅰwas linearized.The sequencing result confirmed that the sequence of inserted fragment was correct.[Conclusion] Eukaryotic expression vector of siRNA targeting annexin A3 gene was successfully constructed.

[Effect of trichloroethylene on expressions of Rho GDI alpha, ANXA3 and GLO1 in L-02 liver cells].

目的研究三氯乙烯(trichloroethylene,TCE)处理L-02肝细胞后,Rho鸟苷二磷酸分离抑制因子(Rho GDI α)、膜联蛋白A3(ANXA3)及乙二醛酶Ⅰ(GLO1)的表达变化.方法以40 μmol/LTCE对L-02肝细胞进行染毒处理,DMSO做溶剂对照.分别采用蛋白质印迹(Western blot)法和实时荧光定量反转录一聚合酶链反应(real-time quantitative RT-PCR)法检测Rho GDIα、ANXA3及GLO1在蛋白质和mRNA水平的表达变化.结果 Western blot法分析结果显示,TCE处理后Rho GDI α、ANXA3和GLO1蛋白表达水平均上调,差异有统计学意义(P<0.05);实时荧光定量RT-PCR结果显示,TCE处理后ANXA3和GLO1 mRNA表达的相对值分别为2.786±0.492和1.893±0.402,而Rho GDIα的mRNA表达的相对值为0.563±0.106,与对照组比较,差异有统计学意义(P<0.05).结论 TCE刺激后,RhoGDI α、ANXA3和GLO1蛋白分别在蛋白质和转录水平发生了相应变化,为进一步研究和揭示TCE致肝细胞损伤的分子机制奠定了基础。

Expression changes of glutathione transferase omega 1 induced by low dose trichloroethylene

OBJECTIVE To determine the expression of glutathione transferase omega 1 (GSTO-1) during hormesis induced by low dose trichloroethylene (TCE). METHODS Based on the dose-response of hormesis induced by TCE, the total protein and mRNA from four groups was extracted, and expression of GSTO-1 was detected by western blotting and RT-PCR technology. RESULTS The study showed that increases of GSTO-1 protein expression were induced by TCE at low dose (0.1 mmol/L) TCE, but there was no significant different between the high-dose group and the control (P < 0.05). RT-PCR analysis presented the expression of GSTO-1 gene in low-dose group was increased, but the opposite effect was observed in the high-dose group. CONCLUSION GSTO-1 may be play a key role in hormesis induced by TCE.
