Zao-Jiao-Ci (ZJC), a traditional Chinese medicine, is considered as a promising candidate to treat laryngeal squamous cell carcinoma (LSCC). However, the underlying molecular mechanism remains unclear.Gene expression profiles of GSE36668 were available from the GEO database, and differentially expressed genes (DEGs) of LSCC were obtained by R package; subsequently, enrichment analysis on KEGG and GO of DEGs was performed. The active ingredients of ZJC were screened from the TCMSP database, and the matched candidate targets were obtained by PharmMapper. Furthermore, we constructed protein-protein interaction (PPI) networks of DEGs and candidate targets, respectively, and we screened the core network from the merged network through combining the two PPI networks using Cytoscape 3.7.2. The key targets derived from the core network were analyzed to find out the associated KEGG signal enrichment pathway. By the GEPIA online website, Kaplan-Meier analysis was used to complete the overall survival and disease-free survival of the selected genes in the core module.We identified 96 candidate targets of ZJC and 86 DEGs of LSCC, the latter including 50 upregulated genes and 36 downregulated genes. DEGs were obviously enriched in the following biological functions: extracellular structure organization, the extracellular matrix organization, and endodermal cell differentiation. The 60 key targets from the core network were enriched in the signal pathways including transcriptional misregulation cancer, cell cycle, and so on. We found that LSCC patients with high expression of HIST1H3J, HIST1H3F, and ITGA4 had worse overall survival, while higher expression of NTRK1, COPS5, HIST1H3A, and HIST1H3G had significantly worse disease-free survival.It suggested that the interaction between ZJC and LSCC was related to the signal pathways of transcriptional misregulation cancer and cell cycle, revealing that it may be the mechanism of ZJC in the treatment of LSCC.
Recently developed optogenetic technology, which allows high-fidelity control of neuronal activity, has been applied to investigate the neural circuits underlying sensory processing and behavior. The nasal cavity is innervated by the olfactory nerve and trigeminal nerve, which are closely related to common symptoms of rhinitis, such as impairment of smell, itching, and sneezing. The olfactory system has an amazing ability to distinguish thousands of odorant molecules at trace levels. However, there are many issues in olfactory sensing mechanisms that need to be addressed. Optogenetics offers a novel technical approach to solve this dilemma. Therefore, we review the recent advances in olfactory optogenetics to clarify the mechanisms of chemical sensing, which may help identify the mechanism of dysfunction and suggest possible treatments for impaired smell. Additionally, in rhinitis patients, alterations in the other nerve (trigeminal nerve) that innervates the nasal cavity can lead to hyperresponsiveness to various nociceptive stimuli and central sensitization, causing frequent and persistent itching and sneezing. In the last several years, the application of optogenetics in regulating nociceptive receptors, which are distributed in sensory nerve endings, and amino acid receptors, which are distributed in vital brain regions, to alleviate overreaction to nociceptive stimuli, has gained significant attention. Therefore, we focus on the progress in optogenetics and its application in neuromodulation of nociceptive stimuli and discuss the potential clinical translation for treating rhinitis in the future.
To study the effect of n-3 polyunsaturated fatty acids (EPA, DHA) on the production of NO, expression of iNOS mRNA and DNA-binding activity of NFkappaB in human monocyte.RT-PCR and enzyme reduction method were used to measure the expression of iNOS mRNA and production of NO respectively. The DNA-binding activity of NFkappaB was assessed by EMSA.EPA, DHA can decrease the production of NO, expression of iNOS mRNA and DNA-binding activity of NFkappaB between the dosage of 10microg/ml and 20microg/ml in human peripheral blood monocyte in vitro.EPA, DHA have inhibitory effects on NO production, iNOS mRNA expression and DNA-binding activity of NFkappaB in monocyte. It indicates EPA, DHA suppress the production of inflammatory mediators via the NFkappaB signal transduction pathways in human monocyte, accordingly suppress inflammatory responses.
Background: Epithelial barrier disruption is the initial pathogenesis of various diseases. We previously reported that acupoint catgut embedding (AE) improved tight junction proteins in rats with allergic rhinitis. However, whether AE benefits the epithelial barrier in local allergic rhinitis (LAR ) is unknown. Methods: In the present study, we used LAR model rats with or without AE treatment to investigate the effect of AE on nasal mucosal barrier function. Then, the LAR model rats were treated with capsaicin or tunicamycin (TM) to investigate the mechanism of AE on LAR. Results: AE ameliorated symptoms and pathological changes of nasal mucosa in LAR rats. In addition, AE reduced inflammatory factors (IL4, IL5, IL13) and inhibited endoplasmic reticulum stress (ERS) by reducing substance P (SP). Finally, AE improved the epithelial barrier function in LAR by suppressing ERS. Conclusion: the present study strongly confirms that AE is an effective method to improve nasal barrier function and prevent LAR barrier damage.
AIM: To explore the effects of oxidized low density lipoprotein (OxLDL) and one of its component— lysophosphatidylcholine (LPC) on cholesterol efflux from mouse macrophage foam cells. METHODS: (1) Cholesterol efflux induced by apoAI from mouse peritoneal macrophage foam cells loaded with OxLDL or acylated LDL(AcLDL) was measured. (2) Cholesterol efflux induced by LPC and apoAI from macrophage foam cells separated from normal or apoE gene deficient (E 0) mouse loaded with AcLDL were measured. RESULTS: (1) When the macrophage foam cells were incubated with apoAI, cholesterol efflux from AcLDL-induced macrophage foam cells increased significantly compared to that of OxLDL-induced macrophage foam cells. (2) LPC promoted cholesterol efflux from macrophage foam cells in relation to both dosage and time. When LPC was incubated with E 0 mouse macrophage foam cells, the released cholesterol mass was significantly lower than that of normal mouse macrophage foam cells. It was also found that cholesterol efflux induced by apoAI normally occurred in E 0 mouse macrophage foam cells. CONCLUSIONS: (1) OxLDL accumulated cholesterol in macrophages and impair cholesterol efflux. (2) LPC induced cholesterol efflux from macrophage foam cells, which may occur via apoE pathway.
Background Continuous comprehensive treatment is still needed after endoscopic sinus surgery (ESS) for chronic rhinosinusitis (CRS) to promote the recovery of sinus mucosal morphology and function. Traditional Chinese medicine (TCM) nasal irrigation is a promising external treatment of TCM, but at present, the application of TCM nasal irrigation after ESS for CRS has not been recommended by the guidelines. Therefore, this article aims to develop a systematic overview and meta-analysis protocol to assess the effectiveness and safety of Chinese herbal nasal rinse for CRS recovery after ESS. Methods Seven databases shall be retrieved from their inception until December 2021. Eligible randomized controlled trials will be covered in the study. The outcome indicators of the survey will consist of efficacy, visual analogue scale score, Lund-Kennedy score for nasal endoscopy, Lund-Mackay score for sinus computed tomography and other secondary outcome indicators. The selection of literature, extraction of data, and methodological quality evaluation of literature shall be conducted by two researchers separately. If there is any dispute, it can be discussed and solved by a third researcher. Review Manager 5.3 software will be applied to data analysis. Results The article will make a detailed research programme to explore the efficacy and safety of TCM nasal irrigation on CRS recovery after ESS. Conclusion This protocol is suitable for evaluating the effectiveness and safety of TCM nasal rinse for CRS recovery after ESS, and can provide corresponding evidence-based medical evidence. Objective Ticagrelor may be an alternative to aspirin as it provides robust and consistent platelet inhibition. However, the effect of ticagrelor treatment in patients undergoing coronary artery bypass grafting (CABG) has not been well confirmed. We conducted a meta-analysis to appraise whether ticagrelor therapy affects outcomes in CABG patients. Methods We searched PubMed, Embase, EBSCO, and Cochrane databases from its inception up to 4 December 2020 for randomized controlled trials that assessed ticagrelor versus non-ticagrelor in patients undergoing CABG. The primary outcome was the incidence of saphenous vein graft (SVG) occlusion at 1 year after CABG. Secondary outcomes were SVG occlusion at 7 days, major adverse cardiovascular events (MACE), and bleeding requiring reoperation. Results Seven trials including 4305 patients (2153 randomized to ticagrelor therapy and 2152 to non-ticagrelor therapy) were included. One-hundred and thirty of 1140 patients (11.4%) randomized to the ticagrelor group versus 175 of 1220 patients (14.3%) randomized to the non-ticagrelor group experienced SVG occlusion at 1 year after CABG. Compared to the control group, ticagrelor therapy yielded a significantly lower risk of SVG occlusion [RR 0.79 (0.64–0.97), p = 0.03]. In the subgroup analysis, ticagrelor plus aspirin compared with aspirin alone did not decrease the risk of SVG occlusion after 1 year [RR 0.65 (0.40–1.07), p = 0.09]. There was no difference in the incidence of SVG occlusion at 7 days [RR 0.67 (0.42–1.06), p = 0.09], MACE up to 1 year [RR 0.99 (0.81–1.21), p = 0.90], or bleeding requiring reoperation [RR 1.16 (0.80–1.70), p = 0.44]. Conclusions Compared with non-ticagrelor therapy, ticagrelor decreased the risk of saphenous vein graft occlusion after 1 year in patients undergoing elective CABG with saphenous vein grafting.
To investigate the characteristics of calcium metabolism in relation to different levels of dietary calcium intakes in premenarche Chinese girls.Forty-nine healthy premenarche girls of Han ethic (9 - 11.5 years) were recruited, and divided into four groups respectively, receiving four different doses of calcium intakes for 6 days, 600 mg (usual diet), 900 mg (containing 250 ml of milk), 1200 mg (containing 250ml of milk and 750 mg of calcium carbonate) and 1500mg calcium (250 ml of milk and 1500 mg of calcium carbonate) per day. A 3-day urine and stool, and a 3-d duplicated food samples were collected to assess the calcium excretion in urine and feces and input of the dietary calcium during the treatment period.There were no significant differences in appearance calcium absorption among the four groups (55%, 53%, 52% and 52%). Urine calcium is correlated negatively with dietary protein and dietary phosphorus. And no correlation was found between dietary protein and calcium absorption.The appearance calcium absorption was (53 +/- 0.12) % in Chinese premenarche girls with dietary calcium intakes ranged between 800 to 1600 mg/d, high dietary protein intakes increase urine calcium secrete.
