Microalgal lipids: A review of lipids potential and quantification for 95 phytoplankton species

Phytoplankton have great potential for biodiesel production and offer promises and opportunities in the long term. Phytoplankton species reach higher growth rates, and thus productivity, than conventional forestry or agricultural crops and other aquatic plants. The oil yield in phytoplankton is an order of magnitude larger than terrestrial oleaginous crops. To meet the potential of phytoplankton-based biodiesel there is a need to radically increase lipid yields, which are generally produced under adverse conditions. Nutrients stress and alterations of cultivation conditions are commonly used as lipid enhancement strategies. It is difficult to get a clear picture of the most efficicent factors affecting lipid accumulation and productivity from the abundant literature on this topic, dispatched into a large variety of species and stresses. This article seeks to summarize the widely reported information on TAGs accumulation in phytoplankton and to decipher the regulation mechanisms triggered along the diversity of enhancement strategies. Most of the factors affecting lipid content and composition were analyzed, such as nutrient starvation, temperature, irradiance, salinity, oxidative stress, metals, CO2 flux, pH and metabolic engineering. In this review, we compiled 213 experiments with lipid analysis, dealing with 95 marine and freshwater phytoplankton (microalgae and cyanobacteria) species. Quantitative indicators (lipid content and productivity), stress level and exposure time, are presented. This review highlights the complexity of comparison between phyla due to differences in culture conditions, analytical methods and/or growth phase. It provides valuable tools for triggering phytoplanktonic lipid biosynthesis and opens the door for enhanced quality and quantity of phytoplankton-based biodiesel.

Microalgae have received much attention in the context of renewable fuel production, due to their ability to produce in high quantities carbon storage molecules such as lipids and carbohydrates. Despite significant research effort over the last decade, the production yields remain low and need to be optimized. For that, a thorough understanding of carbon storage metabolism is necessary. This paper develops a constrained metabolic model based on the dFBA framework to represent the dynamics of carbon storage in microalgae under a diurnal light cycle. The main assumption here is that microalgae adapt their metabolism in order to optimize their production of functional biomass (proteins, membrane lipids, DNA, RNA) over a diurnal cycle. A generic metabolic network comprised of 160 reactions representing the main carbon and nitrogen pathways of microalgae is used to characterize the metabolism. The optimization problem is simplified by exploiting the right kernel of the stoichiometric matrix, and transformed into a linear program by discretizing the differential equations using a classical collocation technique. Several constraints are investigated. The results suggest that the experimentally observed strategy of accumulation of carbon storage molecules during the day, followed by their depletion during the night may indeed be the optimal one. However, a constraint on the maximal synthesis rate of functional biomass must be added for consistency with the biological observations.

Sensors network diagnosis in anaerobic digestion processes using evidence theory

Instrumentation defines a sensors network on a process. Hardware sensors indeed allow one to get different information sources that can be often cross-checked to provide reliable data. However, each of these sources of information contains some uncertainties, either due to the hardware sensors' measurement principles, to their possible fouling, to the estimated parameters of the models used in software sensors and/or to the specific structures of the software sensors. This paper demonstrates that, in this context, the evidence theory is a very well suited formalism for fault detection and diagnosis. This theory indeed allows one to take into account the exact knowledge supported by each source of information and to combine them in order to detect the occurring faults. Moreover, this combination guarantees the best fault isolability from a practical point of view and is suitable for multiple faults occurring at the same time. Finally, the evidence theory is a highly modular formalism since new information sources can be very easily added and old ones can be removed. Validation is performed using real-life experiments from a 1 m3 anaerobic digestion fixed bed process used applied to the treatment of winery wastewaters.
