Intratumoral collagen cross-links heighten stromal stiffness and stimulate tumor cell invasion, but it is unclear how collagen cross-linking is regulated in epithelial tumors. To address this question, we used Kras(LA1) mice, which develop lung adenocarcinomas from somatic activation of a Kras(G12D) allele. The lung tumors in Kras(LA1) mice were highly fibrotic and contained cancer-associated fibroblasts (CAF) that produced collagen and generated stiffness in collagen gels. In xenograft tumors generated by injection of wild-type mice with lung adenocarcinoma cells alone or in combination with CAFs, the total concentration of collagen cross-links was the same in tumors generated with or without CAFs, but coinjected tumors had higher hydroxylysine aldehyde-derived collagen cross-links (HLCC) and lower lysine-aldehyde-derived collagen cross-links (LCCs). Therefore, we postulated that an LCC-to-HLCC switch induced by CAFs promotes the migratory and invasive properties of lung adenocarcinoma cells. To test this hypothesis, we created coculture models in which CAFs are positioned interstitially or peripherally in tumor cell aggregates, mimicking distinct spatial orientations of CAFs in human lung cancer. In both contexts, CAFs enhanced the invasive properties of tumor cells in three-dimensional (3D) collagen gels. Tumor cell aggregates that attached to CAF networks on a Matrigel surface dissociated and migrated on the networks. Lysyl hydroxylase 2 (PLOD2/LH2), which drives HLCC formation, was expressed in CAFs, and LH2 depletion abrogated the ability of CAFs to promote tumor cell invasion and migration.CAFs induce a collagen cross-link switch in tumor stroma to influence the invasive properties of tumor cells.
Recent studies showing significant changes in valvular matrix composition with age offer design criteria for age-specific tissue-engineered heart valves. However, knowledge regarding aging-related changes in valvular material properties is limited. Therefore, 6-week, 6-month, and 6-year-old porcine aortic valves (AV) and mitral valves (MV) were subjected to uniaxial tensile testing. In addition to standard material parameters, the radius of transition curvature (RTC) was measured to assess the acuteness of the transition region of the tension-strain curve. Radially, the MV had greater stiffness and a smaller RTC compared with the AV. Circumferentially, the center of the MV anterior leaflet (MVAC) had the highest stiffness (MVAC > AV > MV free edge [MVF]), greater stress relaxation (MVAC > MVF/AV), lowest extensibility (MVAC < AV < MVF), and smaller RTC compared with MVF (AV < MVAC < MVF). AV and MV radial strips had a larger RTC compared with circumferential strips. Aging elevated stiffness for MV and AV radial and circumferential strips, elevated stress relaxation in AV and MVF circumferential strips, and increased RTC for MV radial and MVF circumferential strips. In conclusion, there are significant age-related differences in the material properties of heart valves, which parallel differences in tissue composition and structure, likely impact valve function, and highlight the need for age-specific design goals for tissue-engineered heart valves.

Extensive research exists on arterial mechanical properties and how they change in disease conditions, but substantially less is known about venous mechanics in healthy and disease states. Although the mechanics of both vessel types are determined by the unique layered composition of the vessel wall, the precise distribution of the layers differs greatly between arteries and veins. Thus, vein mechanics must be analyzed and understood independently from those of arteries. This review discusses the compositional attributes that are unique to veins, how these attributes contribute to venous mechanics, and the alterations that occur to both composition and material properties during venous thrombosis and insufficiency. In general, changes to the venous wall during thrombosis increase wall stiffness and decrease extensibility. During venous insufficiency, however, both the stiffness and the extensibility of the venous wall decrease. Characterizing these changes is essential to better understand disease progression and build vein-specific devices for treating venous disease.
Objective: The mitral valve is a highly heterogeneous tissue composed of two leaflets, anterior and posterior, whose unique composition and regional differences in material properties are essential to overall valve function. While mitral valve mechanics have been studied for many decades, traditional testing methods: imit the spatial resolution of measurements and can be destructive. Optical coherence elastography (OCE) is an emerging method for measuring viscoelastic properties of tissues in a noninvasive, nondestructive manner. Methods: In this study, we employed air-pulse OCE to measure the spatial variation in mitral valve elastic properties with micro-scale resolution at 1 mm increments along the radial length of the leaflets. We analyzed differences between the leaflets, as well as between regions of the valve. Results: We found that the anterior leaflet has a higher elastic wave velocity, which is reported as a surrogate for stiffness, than the posterior leaflet, most notably at the annular edge of the sample. In addition, we found a spatial elastic gradient in the anterior leaflet, where the annular edge was found to have a greater elastic wave velocity than the free edge. This gradient was less pronounced in the posterior leaflet. These patterns were confirmed using established uniaxial tensile testing methods. Conclusions: Overall, the anterior leaflet was stiffer and had greater heterogeneity in its mechanical properties than the posterior leaflet. This study measures differences between the two mitral leaflets with greater resolution than previously feasible and demonstrates a method that may be suitable for assessing valve mechanics following repair or during the engineering of synthetic valve replacements.
