Whale Sharks, Rhincodon typus, Aggregate around Offshore Platforms in Qatari Waters of the Arabian Gulf to Feed on Fish Spawn

Whale sharks, Rhincodon typus, are known to aggregate to feed in a small number of locations in tropical and subtropical waters. Here we document a newly discovered major aggregation site for whale sharks within the Al Shaheen oil field, 90 km off the coast of Qatar in the Arabian Gulf. Whale sharks were observed between April and September, with peak numbers observed between May and August. Density estimates of up to 100 sharks within an area of 1 km2 were recorded. Sharks ranged between four and eight metres' estimated total length (mean 6.92±1.53 m). Most animals observed were actively feeding on surface zooplankton, consisting primarily of mackerel tuna, Euthynnus affinis, eggs.

Genetic Stability Assessment of Three Tomato Hybrids (Lycopersicon esculentum) grown in the Libyan Green Mountains Using SRAP and RAPD Techniques

The genetic diversity of F1 tomato hybrid (Nada, Mona, and Dania) grown in Libyan Green Mountain (LGM) was investigated using SRAP and RAPD molecular marker techniques while the coefficient of variation C.V was used to investigate F1 seed purity. The coefficient of variation C.V., in the three cultivars Nada, Mona and Dania are 31.88, 51.66 and 44.18, respectively, indicating the homogeneity of Nada cultivar over Dania and Mona. The SRAP marker provided a total number of 29 amplified DNA bands with an average of 9.67 bands per primer, 26 of which were polymorphic (67% polymorphism), while the RAPD marker provided a total of 73 bands with an average of 14.6 bands per primer; all of them were 100% polymorphic. The genetic diversity for the three studied cultivars was 0.31, whereas Nada cultivar was the most homogeneous recording a genetic diversity of 0.24 compared with Mona 0.25 and Dania 0.28 Dania. Principal coordinate analysis (PcoA) divided tomato F1 hybrid samples into two main groups, the first (A) includes all the plants of a Nada hybrid cultivar, while group (B) consists of overlapping of Dania and Mona cultivars, reflecting the genetic relatedness.

RAPD-based detection of genomic instability in cucumber plants derived from somatic embryogenesis

Random amplified polymorphic DNA (RAPD) markers were used to evaluate genetic stability of regenerants of cucumber plants obtained through somatic embryogenesis. Somatic embryo plants and plants of F1 hybrids, from which they were derived, were compared during weaning, early growth, flowering, fruiting and at maturity. No differences in phenotype were observed, by simple observation. Banding patterns were scored for the presence (+) or absence (-) of a DNA band, there were no visually detectable differences between the somatic embryo derived plants compared to their F1 parents in the Random Amplified Polymorphic DNA (RAPD) test using five primers OP-C10, OP-G14, OP-H05, OP-Y03 and OP-AT01. The results indicate the usefulness of RAPD markers to detect genetic instability in cucumber primary regenerant plants derived from somatic embryogenesis, and as a certification tool for monitoring genetic stability during the generation process.

Efficiency of RAPD and ISSR Markers for the Detection of Polymorphisms and Genetic Relationships in Date Palm

Background and Objective: Several DNA markers have been used to detect the genetic variation among date palm cultivars, selecting of molecular markers for their specific purposes is the main challenge for researchers, the aim of this study was to evaluate the efficiency of RAPD and ISSR markers in assessing genetic diversity of date palm.Methodology: The DNA of 18 female date palms, comprising Khalas cultivar representing the diversity of date palm genotypes in Qatar was assessed using 29 ISSR primers and 8 RAPD primers, the data were then computed with the PowerMarker software to detect the major allele frequency, number of alleles, gene diversity and Polymorphism Information Content (PIC) value.Results: Twenty nine ISSR primers produced a total of 132 DNA fragments of which 72 fragments (54.5%) were reproducible polymorphic amplified fragments, while 8 RAPD primers produced a total of 32 DNA fragments, of which 22 fragments (68.75%) were polymorphic across all the genotypes.The most effective ISSR and RAPD primers were BT14 and OPC-11 respectively, which generated 7 bands all of which were 100% polymorphic.The RAPD marker exposed 22% gene diversity and 18% (PIC), while ISSR exposed 14 and 12% gene diversity and Polymorphism Information Content (PIC) respectively.Conclusion: The molecular markers obtained by the ISSR technique revealed a remarkable molecular discrimination among date palm compared to the RAPD, while RAPD provides perfect outcomes for date palms genetic diversity.

DNA Finger Printing of <i>Prosopis cineraria</i> and <i>Prosopis juliflora</i> Using ISSR and RAPD Techniques

Diversity within and among the populations of Prosopis cineraria and Prosopis juliflora collected from different loca-tion of Qatar were explored using Inter Simple Sequence Repeat (ISSR) and Random Amplified Polymorphic DNA (RAPD) markers. A total of one hundred and nine bands were generated from twenty nine ISSR and nineteen bands from seven RAPD primers with an average polymorphism of more than ninety nine percent across all the genotypes. ISSR techniques were capable of distinguishing between P. cineraria and P. juliflora, through twenty one bands. How-ever, of the seven RAPD markers, only three bands were able to distinguish between Prosopis species. The dendro-grams for the analysis of genetic similarity show that the individuals from both species can be separated in two highly related groups. Our observations suggest that genetic variations among different accessions of Prosopis are identified using ISSR and RAPD analysis.

Investigation of EMS effect on Vicia faba seed germination and seedling growth under saline condition

The effect of Ethyl-methane Sulphonate (EMS) on seed germination and seedling characters of broad beans (Vicia faba L.) was analyzed in this investigation. The seeds were soaked in different percentages and durations of (EMS), then after sowing were irrigated with a range of NaCl concentration. The effect of EMS was observed based on the seed germination (%), and seedling characters such as: total fresh weight, root fresh weight and root length. Germination percentage generally decreases while concentrations of NaCl increases, however the interaction effect of EMS clearly appeared on concentration 6000 ppm NaCl, which positively increased the germination from 40% in absence of EMS to 60%, when 0.5 or 1% EMS were used. 24 hours soaking in EMS decreased the fresh weight from 33.36 to 13.55 gm per plant, in addition to the decrease of the root length from 22.0cm to 14.2cm per plant. The root nodulation of broad bean is not affected during the increase of NaCl, possibly due to the interactional effect of mutagenesis agent.

New microsatellite markers for assessment of genetic diversity in date palm (Phoenix dactylifera L.)

New primer pairs of genomic DNA microsatellite markers were tested to assess the genetic diversity of eleven date palm genotypes. The results indicated that out of thirty, only seven primers (23.3%) failed to amplify the expected PCR fragments, while thirteen primers (43.3%) amplified monomorphic banding patterns and the remaining ten primers (33.4%) generated polymorphic banding patterns. A total of 77 alleles have been observed with a mean of 7.7 alleles per locus. The average of gene diversity was 0.80 ranging from 0.6 (in marker DP168) to 0.9 (in two markers DP157 and DP175). These new co-dominant markers will be a starting point for researchers making use of the markers for genetic mapping and diversity analysis of date palm.

De novo genome sequencing and comparative genomics of date palm (Phoenix dactylifera)

Date palm is one of the most economically important woody crops cultivated in the Middle East and North Africa and is a good candidate for improving agricultural yields in arid environments. Nonetheless, long generation times (5-8 years) and dioecy (separate male and female trees) have complicated its cultivation and genetic analysis. To address these issues, we assembled a draft genome for a Khalas variety female date palm, the first publicly available resource of its type for a member of the order Arecales. The ∼380 Mb sequence, spanning mainly gene-rich regions, includes >25,000 gene models and is predicted to cover ∼90% of genes and ∼60% of the genome. Sequencing of eight other cultivars, including females of the Deglet Noor and Medjool varieties and their backcrossed males, identified >3.5 million polymorphic sites, including >10,000 genic copy number variations. A small subset of these polymorphisms can distinguish multiple varieties. We identified a region of the genome linked to gender and found evidence that date palm employs an XY system of gender inheritance.

Genetic diversity of Qatari date palm using SSR markers

The genetic diversity in the date palm germplasm of 59 female accessions representing 12 cultivars from different locations in Qatar was investigated using 14 loci of simple-sequence repeat (SSR) primers.A total of 94 alleles, with a mean of 6.7 alleles per locus, were scored.The number of alleles per locus varied from 3 (primer mPdCIR090) to 11 (primers mPdCIR010 and mPdCIR015).The amplified SSR band sizes ranged from 104 to 330 bp.The mean gene diversity was 0.66 and ranged from 0.39 (locus mPdCIRO93) to 0.86 (locus mPdCIR015), indicating that the Qatari date palm collection has a high degree of genetic diversity.The heterozygosity ranged from 0 (marker mPdCIR090) to 98% (marker mPdCIR010).Forty-four percent of the variability is explained at the inter-population level, while 56% of the variability is maintained within individuals.However, the loci mPdCIR044, mPdCIR057, mPdCIR090, and mPdCIR093 revealed that the total gene diversity is explained at the inter-population level.The Qatari populations Khalas, Shishi, Barhi, Hillali, Khnaizi, Gar, and Jabri showed significant differentiation compared to all other populations.The average fixation index was 0.24814, showing that about 24.81% of Genetic diversity of Qatari date palm ©FUNPEC-RP www.funpecrp.com.
