Synthesis and in Vivo Evaluation of Triphenylphosphonium Conjugated Trimetazidine with Enhanced Cardioprotection and Ability to Restore Mitochondrial Function

Trimetazidine exhibits great therapeutic potential in cardiovascular diseases and mitochondria-mediated cardioprotection by trimetazidine has been widely reported. In this study, to enhance its cardioprotection, the triphenylphosphonium-based modification of trimetazidine was conducted to deliver it specifically to mitochondria. Fifteen triphenylphosphonium (TPP) conjugated trimetazidine analogs were designed and synthesized. Their protective effects were evaluated in vivo using a tert-butyl hydroperoxide (t-BHP) induced zebrafish injury model. Structure-activity relationship correlations revealed the best way to couple the TPP moiety to trimetazidine, and led to a new conjugate (18a) with enhanced therapeutic properties. Compared to trimetazidine, 18a effectively protects against heart injury in the zebrafish model at a much lower concentration. Further study in t-BHP treated zebrafish and H9c2 cells demonstrated that 18a protects against cardiomyocyte death and damage by inhibiting excessive production of ROS, maintaining mitochondrial morphology, and preventing mitochondrial dysfunction. Consequently, 18a can be regarded as a potential therapeutic agent for cardioprotection.

Advanced Recursive Spherical Quantization for Image Retrieval

In recent years, quantization technology has proven to be very effective in the field of supervised image retrieval, owing to its capacity to provide both high accuracy and swift retrieval speeds. However, the challenge of balancing retrieval accuracy and speeds, as denoted by variable code lengths, persists as a fundamental consideration across different applications. Recently, there are a few methods that employ recursive strategies for quantization encoding, and there remains substantial room for improvement in their effectiveness. In this paper, we introduce the concept of a Recursive Spherical Quantization (RSQ) architecture, capable of generating sequential binary codes. It can reuse unit vector codebook to recursively quantize real vectors. Empirical findings from benchmark datasets demonstrate that our model is competitive or even superior performance in image retrieval, while simultaneously demanding a significantly reduced number of parameters.

Discovery of New D‐Ring Modified Isosteviol Derivatives as Potent Cardioprotective Agents against Oxidative Stress‐Trigged Damage

Cardiovascular diseases (CVDs) are a major global health concern, and oxidative stress is known to play a central role in their pathogenesis. The identification of new agents capable of inhibiting oxidative stress presents a promising strategy for preventing and treating CVDs. Natural products and their derivatives offer a valuable source for drug discovery, and isosteviol, a readily available natural product, is known to exhibit cardioprotective effects. In this study, 22 new D-ring modified isosteviol derivatives were synthesized and evaluated for their cardioprotective effect in vivo using the zebrafish cardiomyopathy model. The findings revealed that derivative 4e exhibited the most potent cardioprotective effect, surpassing its parent compound isosteviol and the positive drug levosimendan. At 1 μM, derivative 4e significantly protected the cardiomyocytes from injury, while at 10 μM it effectively maintained normal heart phenotypes, preventing cardiac dysfunction in zebrafish. Further investigation demonstrated that 4e protected cardiomyocytes from oxidative stress-induced damage by inhibiting reactive oxygen species overaccumulation, activating superoxide dismutase 2 expression, and enhancing the endogenous antioxidant defense system. These results suggest that isosteviol derivatives, particularly 4e, have the potential to serve as a novel class of cardioprotective agents for the prevention and treatment of CVDs.

Lycojapodine A, a novel C16N-type Lycopodium alkaloid with an unprecedented 6/6/6/7 tetracyclic ring system, was isolated from the club moss Lycopodium japonicum. The structure and relative stereochemistry were elucidated on the basis of spectroscopic data and were further confirmed by X-ray analysis. A possible biosynthetic pathway for 1 was proposed. Its inhibitory activity on acetylcholinestrease and anti-HIV-1 activity were also evaluated.

A Comprehensive Survey on Underwater Image Enhancement Based on Deep Learning

Underwater image enhancement (UIE) is a challenging research task in the field of computer vision. Although hundreds of UIE algorithms have been proposed, a comprehensive and systematic review is still lacking. To promote future research, we summarize the UIE task from multiple perspectives. First, the physical models, data construction processes, evaluation metrics, and loss functions are introduced. Second, according to the contributions brought by different literatures, recent proposed algorithms are discussed and classified from six perspectives, namely network architecture, learning strategy, learning stage, assistance task, domain perspective and disentanglement fusion, respectively. Third, considering the inconsistencies in experimental settings in different literatures, a comprehensive and fair comparison does not yet exist. To this end, we quantitatively and qualitatively evaluate state-of-the-art algorithms on multiple benchmark datasets. Finally, issues worthy of further research in the UIE task are raised. Abstract Seven new phenolic glucosides, 2′‐ O ‐acetylhenryoside ( 1 ), 2′,3′‐di‐ O ‐acetylhenryoside ( 2 ), 2′,6′‐di‐ O ‐acetylhenryoside ( 3 ), 2′,3′,6′‐tri‐ O ‐acetylhenryoside ( 4 ), 2′,3′,4′,6′‐tetra‐ O ‐acetylhenryoside ( 5 ), 2‐[(2,3‐di‐ O ‐acetyl‐ β ‐ D ‐glucopyranosyl)oxy]‐6‐hydroxybenzoic acid ( 6 ), and 6‐hydroxy‐2‐[(2,3,4,6‐tetra‐ O ‐acetyl‐ β ‐ D ‐glucopyranosyl)oxy]benzoic acid ( 7 ), were isolated from the leaves and stems of Viburnum cylindricum , along with 26 known compounds (henryoside=2‐( β ‐ D ‐glucopyranosyloxy)‐6‐hydroxybenzoic acid [2‐( β ‐ D ‐glucopyranosyloxy)phenyl]methyl ester). The structures of the new compounds were established on the basis of chemical and spectroscopic evidences.
