Lightsheet optical tweezer (LOT) for optical manipulation of microscopic particles and live cells

Abstract Optical trapping and patterning cells or microscopic particles is fascinating. We developed a light sheet-based optical tweezer to trap dielectric particles and live HeLa cells. The technique requires the generation of a tightly focussed diffraction-limited light-sheet realized by a combination of cylindrical lens and high NA objective lens. The resultant field is a focussed line (along x -axis) perpendicular to the beam propagation direction ( z -axis). This is unlike traditional optical tweezers that are fundamentally point-traps and can trap one particle at a time. Several spherical beads undergoing Brownian motion in the solution are trapped by the lightsheet gradient potential, and the time (to reach trap-centre) is estimated from the video captured at 230 frames/s. High-speed imaging of beads with increasing laser power shows a steady increase in trap stiffness with a maximum of 0.00118 pN/nm at 52.5 mW. This is order less than the traditional point-traps, and hence may be suitable for applications requiring delicate optical forces. On the brighter side, light sheet tweezer (LOT) can simultaneously trap multiple objects with the distinct ability to manipulate them in the transverse ( xy ) plane via translation and rotation. However, the trapped beads displayed free movement along the light-sheet axis ( x -axis), exhibiting a single degree of freedom. Furthermore, the tweezer is used to trap and pattern live HeLa cells in various shapes and structures. Subsequently, the cells were cultured for a prolonged period of time (> 18 h), and cell viability was ascertained. We anticipate that LOT can be used to study constrained dynamics of microscopic particles and help understand the patterned cell growth that has implications in optical imaging, microscopy, and cell biology.

High resolution multiple excitation spot optical microscopy

We propose fundamental improvements in three-dimensional (3D) resolution of multiple excitation spot optical microscopy. The excitation point spread function (PSF) is generated by two interfering counter-propagating depth-of-focus beams along the optical axis. Detection PSF is obtained by coherently interfering the emitted fluorescent light (collected by both the objectives) at the detector. System PSF shows upto 14-fold reduction in focal volume as compared to confocal, and almost 2-fold improvement in lateral resolution. Proposed PSF has the ability to simultaneously excite multiple 3D-spots of sub-femtoliter volume. Potential applications are in fluorescence microscopy and nanobioimaging.

Multiphoton induced cubic-to-quartic intensity dependent transition of anti-Stokes Raman signal

We show an interesting cubic-to-quartic pump intensity dependent crossover of anti-Stokes Raman signal. This is achieved by multiphoton induced excitation of higher vibronic states. The steady state solution of the population dynamics displays a fourth power dependence for anti-Stokes Raman signal in the pump dominated regime, while Stokes Raman signal continues to show cubic power dependence. Proposed nonlinear intensity dependence has the potential for realizing strong enhancement in anti-Stokes Raman signal and an efficient rejection of background noise.

The Representation Theory of Light

Light fluctuation is strongly associated with its state. A good example of this is coherent light, for which the associated probability distribution is Poisson. This strongly suggests that the description of light and the associated fluctuations can also be understood using a distribution function. This necessitates that light be described purely on the basis of a distribution function, popularly known as the “representation theory” of light. This theory treats light as an entity that exhibit fluctuations with rich statistical significance, which justifies the description of light in the realm of quantum statistics and the development of a quasi-classical distribution theory. This is essentially an extension of the quantum theory that incorporates radiation fields with a non-standard distribution function. An equivalent and powerful approach to understanding light makes use of the Fourier domain, which is associated with the characteristic function. In this chapter, we set out to explore radiation fields, including thermal, random, and squeezed light, based on the representation theory.

Fluorescence-based multifunctional light sheet imaging flow cytometry for high-throughput optical interrogation of live cells

Abstract Multifunctional light sheet imaging flow cytometry of a large population of live cells at high throughput is challenging and requires new technological advancement. Existing cytometry techniques are limited due to point-based illumination that does not allow volume interrogation and biophysical parameter estimation on the go. Here, we propose a multifunctional (multichannel, multisheet and multicolor) imaging cytometry (M3IC) system that employs vertically-aligned multi-sheet array (VAMSA) illumination for interrogating cells flowing simultaneously through multiple microfluidic channels. We studied cancer cells (volume interrogation with organelle-level resolution and high signal-to-background-ratio(SBR)) at high throughput (~2500 nl/min). M3IC system demonstrates organelle-level resolution with a SBR comparable to that of confocal, especially at low flow rates. In addition, the multicolor imaging capability of the system facilitates multi-organelle investigation, determination of critical biophysical parameters, and drug (Paclitaxel) treatment studies on cancer cells. M3IC system is expected to advance the field of fluorescence microscopy, cell biophysics, disease biology and optical physics.

Correlation Single Molecule Localization Microscopy (corrSMLM) Detects Fortunate Molecules for High Signal-to-Background Ratio and Better Localization Precision

Single-molecule localization microscopy can decipher fine details that are otherwise not possible using diffraction-limited microscopy. Often the reconstructed super-resolved image contains unwanted noise, random background and is prone to false detections. This cause spurious data that necessitates several trials, multiple experimentations, and repeated preparation of specimens. Moreover, this is not suitable for experiments that require time-lapse imaging and real-time microscopy. To overcome these limitations, we propose a technique (corrSMLM ) that can recognize and detect fortunate molecules (molecules with long fluorescence cycles) from the recorded data. The technique uses correlation between two or more consecutive frames to extract fortunate molecules that blink for longer than the standard blinking time. Accordingly, strongly-correlated spots (single molecule signatures) are compared in consecutive frames, followed by data integration (mean centroid position and the total number of photons) and estimation of critical parameters (position and localization precision). The technique addresses two major problems that plague SMLM : (1) random noise due to false detection that contributes to strong background, and (2) poor localization precision offered by standard SMLM techniques. On the brighter side, corrSMLM allows only fortunate molecules contribute to the super-resolved image, thereby suppressing the background and improving localization precision by a factor of 2-4 times as compared to standard SMLM. To substantiate, corrSMLM is used for imaging fixed cell samples (Dendra2-Actin and Dendra2-Tubulin transfected NIH3T3 cells). Results show multi-fold reduction in noise and localization precision with a marked improvement in overall resolution and SBR. We anticipate corrSMLM to improve overall image quality and offer a better understanding of single molecule dynamics in cell biology.
