Chestnut Farming with Chinese, Japanese, and Eurojapanese Hybrid Cultivars

This chapter presents the main directions to establish Chinese, Japanese, and Eurojapanese hybrid chestnut orchards. Chinese chestnut plantations are widely established outside China thanks to its broad adaptability to different soil and climatic conditions, its productivity, and its relatively blight-resistance. Japanese chestnut is highly appreciated for its resistance to blight fungus but is less commercially valuable for nut quality than other species. Eurojapanese hybrids are widely appreciated, and the establishment of new cultivations is still increasing worldwide.

Sporogenesis and gametophyte development in the Chinese chestnut (Castanea mollissima Blume.).

【Summary】 Chinese chestnut (Castanea mollissima Blume.) is a widely distributed fruit tree and well known for its ecological and economic value. In order to evaluate obstacles to sexual reproduction in C. mollissima, microsporogenesis, megasporogenesis, and development of the male and female gametophytes of the C. mollissima ‘Yanshanzaofeng’ cultivar were examined by a paraffin section technique and scanning electron microscopy. Results showed that the anther wall was of the basic type and consisted of an epidermis, endothecium, middle layers, and tapetum. The tapetum was of the glandular type. Cytokinesis during meiosis of the microspore mother cell was of the simultaneous type, tetrads were tetrahedral, pollen grains were the 2-cell type, and development of the embryo sac was of the Polygonum type. In total, 16~18 ovules were found in the ovary axis, and the ovules were anatropous, bitegmic, and crassinucellate. Before fertilization, the 2 polar nuclei fused into a secondary nucleus. The mature embryo sac was made up of 7 cells. Abnormal embryo sacs or abortive ovules were observed in the ovary. The abortive ovules are likely the cause of the low

Castanea species show high levels of diversity (morphological and ecological traits, vegetative and reproductive habits, wood characteristics, adaptability and resistance to biotic and abiotic stresses, etc.), reflecting the adaptation of the genus to different environmental conditions. Tree shape and form are variable. C. dentata and C. sativa are upright, tall, and slender trees, whilst other species are smaller, with round foliage and branches that start from the base. Other species are dwarf shrubs. Plants can live and be regularly productive for centuries. Some species grow rapidly and re-sprout quickly when cut. Nut size, peeling, and pulp characteristics vary greatly among species and cultivars, and represent important features in the food market. Chestnut is a good source of nutrients, and it has a steady consumption in the main producing countries. Due to its mineral elements and bioactive compounds, chestnut can be considered a health-promoting food.

A Study of Microsporgenesis and Male Gametogenesis in <em>Camellia grijsii</em> Hamce

Camellia grijsii Hamce is used as a woody edible oil tree and wellknow for its commercial value in Southern China. The plants rarely set viable seeds and have little work on the reproduction biology. In order to verify whether there was any obstacle of male reproduction in the C. grijsii, microsporgenesis and male game to genesis in C. grijsii have been evaluated by paraffin section technique. The results are showed that the development of the anther wall belonged to a basic type and consisted of epidermis, endothecium, middle layers and tapetum. The tapetum conformed to the glandular type. Cytokinesis during meiosis of the microspore mother cell was simultaneous type. A majority of the microspores were arranged in tetrahedral tetrads. The mature pollen grains were 2-cell type and had three germ pores. Anthers were dehiscent and pollen grains shed on the early-February. Based our results, we did not find the abnormal male flower in the C. grijsii, suggesting that male gametes were fertile and male sterility was not the major cause of the low seed set in the C. grijsii.

Design, Synthesis, and Structure–Activity Relationship Studies of Bisamide Derivatives of Amphotericin B with Potent Efficacy and Low Toxicity

Amphotericin B (AMB, 1) is the most powerful antibiotic in treating potentially life-threatening invasive fungal infections (IFIs), though severe toxicity derived from self-aggregation greatly limits its clinical application. Herein, we applied a bisamidation strategy at the C16-COOH and C3'-NH2 to improve the therapeutic properties by suppressing self-aggregation. It was found that basic amino groups at the residue of C16 amide were beneficial to activity, while lipophilic fragments contributed to toxicity reduction. Additionally, N-methyl-amino acetyl and amino acetyl moieties at C3' amide could help keep the fungistatic effectiveness. The modification work culminated in the discovery of 36 (ED50 = 0.21 mg/kg), which exerted a 1.5-fold stronger antifungal efficacy than amphamide, the optimal derivative theretofore, in mice, low self-aggregation propensity, and thus low acute toxicity. With the improvement in therapeutic index and good PK profile, 36 is promising for further development as a second-generation polyene antifungal agent.

Self-Sterility in Camellia oleifera May Be Due to the Prezygotic Late-Acting Self-Incompatibility

In this report, self-sterility in Camellia oleifera was explored by comparing structural and statistical characteristics following self-pollination (SP) and cross-pollination (CP). Although slightly delayed pollen germination and pollen tube growth in selfed ovaries compared to crossed ovaries was observed, there was no significant difference in the percentages of pollen that germinated and pollen tubes that grew to the base of the style. There was also no difference in morphological structure after the two pollination treatments. However, the proportions of ovule penetration and double fertilization in selfed ovules were significantly lower than in crossed ovules, indicating that a prezygotic late-acting self-incompatible mechanism may exist in C. oleifera. Callose deposition was observed in selfed abortive ovules, but not in normal. Ovules did not show differences in anatomic structure during embryonic development, whereas significant differences were observed in the final fruit and seed set. In addition, aborted ovules in selfed ovaries occurred within 35 days after SP and prior to zygote division. However, this process did not occur continuously throughout the life cycle, and no zygotes were observed in the selfed abortive ovules. These results indicated that the self-sterility in C. oleifera may be caused by prezygotic late-acting self-incompatibility (LSI).
