[Sensitizing and protective activity of a staphylococcal vaccine made of water-soluble antigens, depending on the method and schedule of administration].

An experiment on guinea pigs immunized with staphylococcal vaccine prepared from water-soluble antigens revealed that the degree of developing sensitization and specific resistance was essentially determined by the method and schedule of the administration of the preparation. The intranasal administration of the vaccine induced a lesser degree of sensitization in comparison with its subcutaneous injection. The optimum response to the administration of the vaccine (a low sensitization level and a high degree of protection from infection) was observed in the animals immunized first intranasally and then by subcutaneous injection. The subcutaneous injection of the preparation in combination with its subsequent intranasal application induced a more pronounced degree of sensitization and a lesser degree of protection from infection.

[Protective effect of the aerosol immunization of mice with the polycomponent vaccine Immunovac VP-4 after the challenge of the animals with Klebsiella pneumoniae virulent strain].

Used four schemes of the administration of the preparation with different time of the exposition of the animals in an aerosol chamber were tested with their subsequent intraperitoneal challenge with K. pneumoniae virulent strain K16. Irrespective of the number of immunization courses, the administration of the preparation made at intervals of 1 day, or daily, did not ensure any protective effect, but only led to an insignificant increase in their survival time in comparison with nonimmunized animals. After intervals between immunizations were increased to 3 days the protective effect of aerosol immumization was obtained (the survival rate was 65-80 % and considerably differed from that of the controls). The protective effect of aerosol immunization thus obtained was comparable with the effectiveness immunization made in a single subcutaneous injection. Aerosol immunization resulted in low antibody titers to the antigens contained in the vaccine, while after a single subcutaneous injection high antibody titers to Klebsiella and Proteus antigens were detected. The antigen-stimulated blast transformation of spleen lymphocytes in mice subjected to aerosol immunizations in 5 exposures was high. After subcutaneous immunization significant changes in such characteristics were detected on day 15. The data thus obtained were indicative of good prospects in the development Immunovac VP-4 as the medicinal form intended for use in aerosols.

[The effect of immunotherapy with cell-free vaccines made from the antigens of opportunistic microorganisms on the dynamics of DNA antibody formation].

The sera of patients subjected to immunotherapy with staphylococcal vaccine and with multicomponent vaccine (i.e. the mixture of the antigenic preparations of Staphylococcus aureus, Klebsiella pneumoniae, Proteus vulgaris, Escherichia coli) were studied by the method of the enzyme immunoassay on the basis of cattle spleen DNA. Immunotherapy with staphylococcal vaccine was given to patients with dermal diseases, chronic obstructive bronchitis and pulmonary abscess. Multicomponent vaccine was introduced to patients with the infectious allergic form of bronchial asthma, moderate or severe. Immunotherapy with both preparations under study was shown to produce no accumulation of antibodies to native and denatured DNA.

[Experimental study of the protective activity of a complex staphylococcal antigen and toxoid when administered in combination].

The data obtained in the experimental study of the protective activity of an antigenic staphylococcal complex prepared by the method of aqueous extraction and staphylococcal toxoid (native and adsorbed) are presented. The study carried out on the model of septic staphylococcal infection in rabbits indicated that after immunization with the mixture of the above-mentioned preparations the survival time of the rabbits increased to a greater extent than after immunization with each of these preparations separately. This regularity was especially pronounced under the aggravated conditions of infection with the mixture of strains.

The results of the study of rabbit antisera to meningococci A, B, C in the double diffusion in gel, passive hemagglutination test and enzyme immunoassay with antigenic preparations isolated from S. aureus strains are indicative of the presence of common antigenic determinants of protein and polysaccharide nature in S. aureus and N. meningitidis.

[Experimental, clinical and immunologic assessment of acellular staphylococcal vaccine "Staphylovac"].

Results of experimental, clinical and immunological effects of acellular dry staphylococcal vaccine Staphylovac developed in Mechnikov Research Institute of Vaccines and Sera are presented. Original mildly virulent strains of Staphylococcus aureus having high immunogenicity, and intra- and interspecies protective activity against different representatives of opportunistic microflora were used for construction of the preparation. Low-toxicity and weak anapylactogenicity of the vaccine were established. In experiments on mice, guinea pigs and rabbits significant protective, antigenic and immunomodulate activity of the preparation was revealed with low sensitization of animals. Clinical trials performed in different centers showed that inclusion of vaccinotherapy in complex treatment of chronic staphylococcal infections (chronic pyodermia, lung abscess etc.) resulted in prolonged pathologic locus, decrease of number and severity of exacerbations, prolongation of remission, and complete recovery in significant number of patients. Activation of innate and adaptive immunity was revealed in the same patients. It was shown on the large group of athletes that administration of the vaccine by aerosol route prevents disruption of immunologic adaptation occurring due to excess physical activity and stress situations during competitions.

[ChemicaL and immunochemical characteristics of antigenic preparations from Staphylococcus aureus and Klebsiella pneumoniae as the components for an associated vaccine].

S. aureus aqueous extract and K. pneumoniae hydroxylamine vaccine were studied by means of chemical and immunochemical analytical techniques. The preparations were found to contain, respectively, 7.0% nad 53.5% of neutral monosaccharides, 6.5% and 0.7% of nucleic acids, as well as protein in approximately equal amounts (11.63-14.0%). In experiment of immunodiffusion, immunoelectrophoresis and rocket immunoelectrophoresis in homologous systems with hyperimmune antimicrobial sera the preparations were characterized by serological heterogeneity. After their combination with Escherichia coli aqueous extract and Proteus hydroxylamine preparation their serological characteristics remaIned unchanged. The study of cross reactions of all components of the combined preparations with hyperimmune rabbit sera to the corresponding microorganisms revealed that only Klebsiella component of the combined vaccine reacted with all hyperimmune sera. The preparation of Proteus showed the lowest activity, it reacted only with hyperimmune sera to K. pneumoniae. Besides, no reaction of S. aureus component with sera to E. coli and no reaction of the preparation of E. coli with antistaphylococcal serum were observed.

[Toxicity and the protective activity of a complex staphylococcal antigen obtained by water extraction].

The toxicity and the protective activity of a complex staphyloccus antigen was studied. Its insignificant toxicity for mice when injected intraperitoneally and protective activity under conditons of experimental infection on mice, rabbits, and guinea pigs were demonstrated. In particular it was shown that the antigen obtained in doses under study protected rabbits from septic staphylococcus infection more than antitoxin and corpuscular vaccine. Under conditons of local experimental infection on mice the complex antigen promoted a more rapid than corpuscular vaccine elimination of the local focus of infection.
