Studied are phenotypic and genetic properties of anthrax agent strains, isolated from earthworms retained in infected soil for a period of 20 and 30 days. Discovered is the fact that the presence of worms in the biotope reduces the number of Bacillus anthracis spores by 30-50 %. However, the rest of the spores preserve original properties, and biological and genetic virulence dominants.

Biosafety Cabinets Used in the Work with Biological Agents of I-II Pathogenicity Groups

At present, in compliance with sanitary and epidemiologic regulations in force, equipping of microbiological laboratory with modern engineering and technical systems and equipment is considered to be the important condition ensuring adherence with the biological safety requirements. The abovementioned systems and equipment, together with the rules and techniques of the safe work with pathogenic microorganisms, should provide the set of biological safety measures for the laboratory personnel. Biological safety cabinets of the second and the third classes serve as additional protection means. Considered are peculiarities of the design and benefits of different biosafety cabinets used in BSL 2-3 laboratories, and some of their shortcomings are determined.

Usage of chi-Sequence for Differentiation between the Strains of Anthrax Etiological Agent and Closely Related Bacilli

Objective of the study was to carry out comparison between the microorganisms appurtenant to six different species of Bacilli genus as regards mobility of the amplicons obtained using the primer that contained chi-sequence of Bacillus subtilis . Therewith, analyzed were 30 Bacillus anthracis strains and closely related bacilli. Cultures grew on L-agar and L-broth at 37 °C. DNA was isolated from vegetative B. anthracis and Bacillus spp . cultures introducing lysozyme and phenol de-proteinizing. In order to perform single-primer PCR constructed was ChiBs primer - 5’-CTAGGAGCGGG-3’. Single-primer amplification (30 cycles) was carried out at annealing temperature of 47 °C. Comparative analysis of individual electrophoretic amplicon profiles, obtained by means of ChiBs-primer PCR, allows for identification of genetic intraspecific variability and differentiation of B. anthracis strains from the atypical ones and closely related species of bacilli.

The role of earthworms in the spread of anthrax

The article discusses the issue of the danger of earthworms in the spread of anthrax in connection with the long-term persistence of the causative agent of the disease in the soil of cattle burial grounds. The results of assessing the effect of earthworms on soil cleansing from the anthrax pathogen and the properties of microorganisms isolated from worms are presented. Studies have shown that under the influence of worms, the amount of anthrax pathogen in the soil decreases by 30–50%, but the remaining spores did not change their properties and retained all the main biological and genetic factors of pathogenicity. Designed is a differential-diagnostic solid nutrient media for isolation and identification of anthrax agent. This media contains: nutrient base (enzymic hydrolysate of fishbone powder), agar-agar, D (+)-sorbite, pH indicator bromthymol blue, and broad-spectrum antibiotic (polymyxin). The media makes it possible to differentiate virulent (capsular) and avirulent (non-capsular) Bacillus anthracis strains, as well as closely related saprophyte microorganisms, depending upon the color and colony morphology.
