Natural History study to characterize CC011Unc/J spontaneous Mouse Model of Colitis

Abstract Objective: Inflammatory bowel disease (IBD), including ulcerative colitis and Crohn’s disease, is a complex disorder characterized as an autoimmune response to the host’s intestinal microflora. We evaluate the progression of the colitis using CC01UncJ mice, a recombinant inbred mouse with similar characteristics to human IBD (Rogala AR et al., Mamm Genome, 2014. 25:95-108). Methods: CC011Unc/J mice (JAX stock# 018854) were monitored weekly. Mice were euthanized at 8, 25, 30, 35 or 40 weeks of age, and colons were fixed and stained with hematoxylin and eosin. Slides were analyzed for leukocyte density, glandular hyperplasia, goblet cell loss, vasculitis, crypt abscesses, mucinous lakes, ulceration, and extent of lesion on a scale from 0-4. Results: CC011Unc/J mice showed mild to moderate inflammation characterized by multifocal lymphocytes and plasma cell aggregates. A statistically significant scoring increase was observed between 8 weeks and 30 weeks (p<0.01), with a plateau reached at 35-40 weeks of age. Significant differences between males and females were not observed for any of the scored parameters. Conclusion: In contrast to the publications, CC011Unc/J mice showed only a moderate inflammation, suggesting an important role of environmental conditions to the development of the disease. Additional studies will be performed at JAX preclinical services to fully characterize these mice to offer this model as a platform for performing pre-clinical testing.

Effectiveness of a media campaign promoting health care provider tobacco dependence treatment

The New York State Department of Health (NYSDOH) developed a provider-focused media campaign to encourage provision of evidence-based, clinical tobacco dependence treatment (TDT). The purpose of this study was to assess providers' awareness of the campaign and the relationship between campaign awareness and changes in campaign-related beliefs and clinical TDT intervention. We conducted a longitudinal, mailed survey of health care providers in New York State (n = 851; AAPOR3 RR: 24.6%). We estimated descriptive statistics and used multivariable regression analyses to assess whether changes in key outcomes (campaign-related beliefs and clinical TDT) from pre- to post-campaign vary by self-reported campaign awareness. Approximately 12% of providers were aware of the campaign. In multivariable analyses, changes from pre- to post-campaign in provider beliefs that the nicotine patch and gum are very effective at helping patients quit were greater for providers aware of the campaign compared with those not aware of the campaign (For patch: OR 2.17, CI 1.06-4.45, p = 0.03; for gum: OR 2.78, CI: 1.24-6.27, p = 0.01), but not for provider behavior. After seeing the NYSDOH campaign, providers' beliefs about the effectiveness of the patch and gum increased. Many state tobacco control programs and health care organizations are implementing tobacco-related policies and systems to facilitate the provision of clinical TDT; this study suggests that a digital and print provider-focused media campaign has the potential to complement health systems change interventions. Future studies should seek to identify ways to modify ad delivery to increase campaign awareness to maximize potential campaign impact.

Eff ect of the fi rst federally funded US antismoking national media campaign

Findings 3051 smokers and 2220 non-smokers completed baseline and follow-up assessments. 2395 (78%) smokers and 1632 (74%) non-smokers recalled seeing at least one Tips advertisement on television during the 3-month campaign. Quit attempts among smokers rose from 31·1% (95% CI 30·3–31·9) at baseline to 34·8% (34·0–35·7) at follow-up, a 12% relative increase. The prevalence of abstinence at follow-up among smokers who made a quit attempt was 13·4% (95% CI 9·7–17·2). N ationally, an estimated 1·64 million additional smokers made a quit attempt, and 220 000 (95% CI 159 000–282 000) remained abstinent at follow-up. Recommendations by non-smokers to quit grew from 2·6% at baseline to 5·1% at follow-up, and the prevalence of people talking with friends and family about the dangers of smoking rose from 31·9% (95% CI 31·3–32·5) to 35·2% (34·6–35·9), resulting in an estimated 4·7 million additional non-smokers recommending cessation services and more than 6 million talking about the dangers of smoking. Interpretation The high-exposure Tips media campaign was eff ective at increasing population-level quit attempts. The growth in smokers who quit and became sustained quitters could have added from a third to almost half a million quality-adjusted life-years to the US population. Expanded implementation of similar campaigns globally could accelerate progress on the WHO F ramework Convention on Tobacco Control and reduce smoking prevalence globally.

The impact of anti-tobacco industry prevention messages in tobacco producing regions: evidence from the US truth® campaign

Adolescents who live in tobacco producing regions may not respond favourably to anti-industry ads.To examine whether state level involvement in tobacco production appears to limit the effectiveness of anti-industry ads to prevent tobacco use among adolescents in the USA.Time trend analyses were done using repeated cross sectional data from six waves of the Legacy Media Tracking Survey, which were collected between 1999 and 2003.28,307 adolescents, ages 12-17 years, were classified as living in: tobacco producing states (TPS) (n = 1929); non-tobacco producing states (non-TPS) with low tobacco control funding comparable to TPS (n = 5323); non-TPS with relatively high funding (n = 15,076); and non-TPS with established anti-industry ad campaigns (n = 5979).Reactions to anti-industry ads; strength of anti-industry attitudes/beliefs; changes in anti-industry attitudes/beliefs over time.Ad reactions did not differ by state type. Multivariate adjusted time trend analyses indicated significant, comparable increases in anti-industry attitudes/beliefs since the onset of the truth campaign, in both TPS and non-TPS. Mediation analyses indicated that these increases were due, in part, to campaign exposure.Adolescents who live in tobacco producing regions appear to be as responsive to anti-industry ads as their counterparts in non-tobacco producing regions. This study provides further evidence for the effectiveness of such ads.

Consequences of dramatic reductions in state tobacco control funds: Florida, 1998-2000

Objective: This study assessed whether dramatic funding reductions to the Florida Tobacco Control Program (FTCP) influenced trends in recall of the Florida “truth” anti-smoking media campaign, anti-industry attitudes and non-smoking intentions among Florida teens. Methods: We used an interrupted time series technique to test for differences in the rates of change in Florida “truth” recall, anti-industry beliefs and non-smoking intentions before and after the FTCP budget cuts using the Florida Anti-tobacco Media Evaluation (FAME) survey, a repeated cross-sectional telephone survey of Florida teens. Results: Recall of the Florida “truth” anti-smoking campaign, anti-industry attitudes, and non-smoking intentions increased dramatically between April 1998 and May 1999. Florida “truth” recall declined after FTCP budget cuts in June 1999. Anti-industry beliefs and non-smoking intentions plateaued or began to decline after the budget cuts. The launch of the national “truth” campaign in February 2000 may have offset otherwise deleterious effects of the budget cuts on anti-industry beliefs, but not smoking intentions. Conclusion: Reductions in tobacco control funding have immediate effects on programme exposure and cognitive precursors to smoking initiation. There is a critical need to maintain and enhance funding for state tobacco control programmes to continue nationwide progress in preventing youth from initiating cigarette smoking.
