Comparative efficacy of metronidazole and another 5-nitroimidazole, CG-10213-Go, in caecal amoebiasis in albino mice (Mus musculus)

In an experimental patent infection of Ancylostoma ceylanicum in golden hamsters the efficacy of seven anthelmintics was tested; bephenium hydroxynaphthoate, tetramisole, tetrachlorethylene, phenylene 1,4-diisothiocyanate, thiabendazole, parbendazole and mebendazole were studied. Mebendazole was the most effective, with parbendazole, phenylene 1,4-diisothiocyanate, thiabendazole and bephenium hydroxynaphthoate also satisfactory. Tetrachlorethylene and tetramisole were the least effective in our screen.

Bio fabrication of Copper Nanoparticles: A Next Generation Antibacterial Agent against Wound Associated Pathogens

Impaired wound healing is a major complication. A few factors such as blood glucose level, poor circulation, immune system deficiency, and infection are the root causes of impaired wound healing. The aim of the present study was to bio-synthesize copper nanoparticles with potential antibacterial activity against wound-associated pathogens.Copper nanoparticles were fabricated using the sol-gel method with the mixing of Syzigium cumini leaf extract in metal salt solution. The particles were then later characterized using UV spectroscopy, SEM, TEM, FTIR, and XRD, and evaluated for their antibacterial activity and its MIC against four wound-associated pathogens.The results obtained from TEM, SEM, and XRD characterization showed that the particle size was below 100 nm and of spherical shape. FTIR analysis showed the possibility of various biomolecules, which have a role in capping and stabilizing copper nanoparticles. The particles synthesized showed antibacterial activity against four wound-associated pathogens (P. mirabilis, S. saprophyticus, S. pyogenes, and P. aeruginosa).The biosynthesized copper nanoparticles showed potent antimicrobial activity, thus the antibacterial activity of the synthesized copper nanoparticles could be used in several biomedical applications. Additionally, they can be exploited as a better therapeutic agent for treating infection seen in impaired diabetic wounds. The particles synthesized by the biological route are eco-friendly, less toxic, feasible, and cost effective.

Nanotechnology is growing rapidly and its applications are enormous. A number of nano-based products are available currently on the market. Increased use of nanoproducts has become hazardous for both environmental and human life. Nanoparticles (NPs) have been increasingly released into environments through various means. Different NPs have different mechanisms of toxicity in various organisms. In the environment, water bodies in particular get severely deteriorated with discharged nanomaterials. This has severely impacted the flora & fauna. Various measures are needed for the selective removal of toxic nanomaterials from the environment. The present chapter provides concentric knowledge of toxic effects of NPs and nanoproducts. This chapter elaborates on NP exposure to humans via potential routes and also interaction of NPs to natural ecosystem,s plants as well as humans.

Nitroimidazoles: Part XIX. Structure-activity relationships

A variety of nitroimidazoles, mostly 1,2-disubstituted-5-nitroderivatives were examined for in vitro activity against E. histolytica and in early hepatic infection of infected golden hamsters. Many preparations carried a functionalised N atom at position 2. In vivo activity was found widely among 1-alkyl-5-nitroimidazoles carrying a substituted imidazolidinone (1) or imidazole (2). Out of these derivatives, l-methyl-sulphonyl-3-(1-methyl-5-nitro-2-imidazolyl)-2-irnidazolidinone(la;C 10213-Go) was found to be the best and to be superior to marketed nitroimidazoles against hepatic and caecal infections of E. histolytica ip the golden hamster and T. foetus infections in mice, and has been developed as a drug for treatment of amoebiasis, giardiasis and trichomoniasis.

Serum and blister‐fluid elevation and decreased epidermal content of high‐mobility group box 1 protein in drug‐induced Stevens–Johnson syndrome/toxic epidermal necrolysis

High-mobility group box 1 (HMGB1) is a damage-associated molecular-pattern protein. Stevens-Johnson syndrome (SJS)/toxic epidermal necrolysis (TEN) are serious, immune-mediated skin-blistering conditions.To determine serum and/or blister-fluid total HMGB1 levels in SJS/TEN cohorts, and HMGB1 expression in formalin-fixed, paraffin-embedded (FFPE) SJS/TEN skin vs. healthy and maculopapular exanthema (MPE) skin. Methods Serum HMGB1 was quantified in Malawian nevirapine-induced hypersensitivity, Taiwanese SJS/TEN and Spanish SJS/TEN cohorts. FFPE skin (healthy skin, MPE, SJS/TEN) was stained and assessed for HMGB1 expression.Serum total HMGB1 was not significantly elevated in patients with nevirapine-induced SJS/TEN (3·98 ± 2·17 ng mL-1 ), MPE (3·92 ± 2·75 ng mL-1 ) or drug reaction with eosinophilia and systemic symptoms (4·73 ± 3·00 ng mL-1 ) vs. tolerant controls (2·97 ± 3·00 ng mL-1 ). HMGB1 was significantly elevated in Taiwanese patients with SJS/TEN, highest during the acute phase (32·6 ± 26·6 ng mL-1 ) vs. the maximal (19·7 ± 23·2 ng mL-1 ; P = 0·007) and recovery (24·6 ± 25·3 ng mL-1 ; P = 0·027) phases. In blister fluid from Spanish patients with SJS/TEN, HMGB1 (486·8 ± 687·9 ng mL-1 ) was significantly higher than in serum (8·8 ± 7·6 ng mL-1 ; P <0·001). Preblistered SJS/TEN skin showed decreased epidermal nuclear HMGB1 expression in upper epidermis vs. healthy or MPE skin but retained basal/suprabasal expression.Epidermal HMGB1 expression was decreased in SJS/TEN skin. Retained basal/suprabasal epidermal HMGB1 expression may exacerbate localized injury in SJS/TEN.

Evaluation of Platelet and Red Blood Cell Parameters with Proposal of Modified Score as Discriminating Guide for Iron Deficiency Anemia and β -Thalassemia Minor

Iron Deficiency Anaemia (IDA) and β-Thalassaemia Minor (BTM) are considered to be important cause of microcytic hypochromic anaemia. Studies have evaluated various red cell parameters which are easily available on electronic cell counters for discrimination of IDA and BTM in different ethnic populations. The analysis of previously established red cell discriminative indices with new cut-off have also been done by studies which may be relevant in their set of population for differentiation.The study was conducted to propose a modified score considering the established red blood cell indices with a new cut off and to formulate index taking into consideration Red Blood Cell (RBC) and platelet parameters for early differentiation of IDA and BTM.The prospective study included cases with MCV< 80 fl and new modified score of 11 was proposed by statistically analysing the previous discriminative indices with new cut-off by giving score 0 for IDA and score 1 for BTM. The summation of all scores gave modified 11 T score. A new cut off for differentiation of IDA and BTM was proposed in the study by using ROC curve and analysing AUC which statistically corresponded to highest accuracy. An attempt to formulate a new index using the RBC and platelet parameters was also made for initial discrimination.The study included 153 cases and in addition to red blood cell parameters, mean platelet volume and platelet distribution width also showed statistical significant difference between IDA and BTM (p<0.05). Modified new 11 T score was 87.6% specific for BTM while proposed index showed 80.4% negative predictive value for BTM and correctly identified 75% of cases.The proposed new index and modified 11T score may be used for initial discrimination of BTM and IDA especially in resource limited regions. Apart from RBC parameters, mean platelet volume and platelet distribution width may also be useful in early differentiation. It is essential to determine cut off of every index in given population for differentiation of these two conditions.
