Isatin‐Triazole‐Functionalized Rhodamine: A Dual Sensor for Cu²⁺ and Fe³⁺ Ions and Its Application to Cell Imaging

Abstract A new isatin‐triazole tethered rhodamine based fluorescent probe R1 (1‐(2’’‐(4’‐(((3’’’,6’’’‐ bis (diethylamino)‐3’’‐oxospiro[isoindoline‐1’’,9’’’‐xanthen]‐2’’‐yl)amino)methyl)‐1 H ‐1’,2’,3’‐triazolyl)ethyl)indoline‐2,3‐dione) has been synthesized using click chemistry approach. Probe R1 exhibits a dual sensor property for Cu 2+ and Fe 3+ ions through turn‐on fluorescence response. A prominent colour change from colourless to pink allows the naked eye identification of aforementioned metal ions. R1 forms a 1:2 complex with Cu 2+ and Fe 3+ ions with binding constants 8.0 × 10 8 and 2.93 × 10 7 M −2 , respectively. The binding mode is scrutinised through various spectroscopic techniques which is further supported by theoretical calculations. The detection limit of R1 for Cu 2+ and Fe 3+ ions is found to be 12.2 nM and 0.33 μM, respectively. Further, R1 has been evaluated for its potential to detect Cu 2+ ions in biological systems using fluorescence cell imaging studies.

Evaluation of effectiveness of polymeric nanoparticles based vaccine delivery system over varying time intervals using tetanus toxoid as model antigen

At present choice of adjuvants for human vaccination reflects a compromise between a requirement for adjuvanticity and an acceptable level of side-effects. To overcome the side effects, encapsulation of antigen in the biodegradable polymeric nanoparticles (NPs) may be a promising approach, which may be developed as adjuvants. Moreover, small size of these nano-formulations may help to project the antigen directly to the antigen presenting cells (APCs) for stimulating cell mediate immunity. The present study was directed towards the evaluation of PLA and PCL based nanoformulations as potential adjuvants by using tetanus toxoid as model antigen. The particles size was observed in the range from 92.9 ± 2.6 to 124.6 ± 3.7 nm having zeta potential of -12.4 ± 1.2 to -3.4 ± 1.5. The loading efficiency of different formulations ranges from 46.3 - 56.8 %. Highest loading of 56.8% and burst release (82 % in 48 days) was observed in PLA-PEG based nanoformulation. In terms of anti-tetanus antibodies (determined by ELISA) titer of > 0.5 IU/ml was observed upto 70 days in all the survived mice. Efficacy of nanoformulations was studied in mice by challenge method over different time intervals wherein tetanus loaded PLA, PLA-PEG and PCL NPs shows maximum efficacy at 28, 56 and 21 days respectively. The study shows that the biodegradable nanoparticle based formulations has no toxicity, comparable efficacy and therefore has a strong potential as vaccine adjuvant and delivery system.

Cell replacement therapy holds a promising future in the treatment of degenerative diseases related to neuronal, cardiac and bone tissues. In such kind of diseases, there is a progressive loss of specific types of cells. Currently the most upcoming and trusted cell candidate is Mesenchymal Stem Cells (MSCs) as these cells are easy to isolate from the tissue, easy to maintain and expand and no ethical concerns are linked. MSCs can be obtained from a number of sources like bone marrow, umbilical cord blood, umbilical cord, dental pulp, adipose tissues, etc. MSCs help in tissue repair and regeneration by various mechanisms of action like cell differentiation, immunomodulation, paracrine effect, etc. The future of regenerative medicine lies in tissue engineering and exploiting various properties to yield maximum output. In the current review article, we have targeted the repair and regeneration mechanisms of MSCs in neurodegenerative diseases, cardiac diseases and those related to bones. Yet there is a lot to understand, discover and then understand again about the molecular mechanisms of MSCs and then applying this knowledge in developing the therapy to get maximum repair and regeneration of concerned tissue and in turn the recovery of the patient.

Linking Organic Metabolites as Produced by Purpureocillium Lilacinum 6029 Cultured on Karanja Deoiled Cake Medium for the Sustainable Management of Root-Knot Nematodes

Root-knot nematodes pose a serious threat to agriculture and forest systems, causing significant losses of the crop worldwide. A wide range of chemical nematicides has traditionally been used to manage phyto-nematodes. However, due to their ill effects on the environment, biological control agents (BCAs) like Purpureocillium lilacinum that exhibit antagonistic effects on root-knot nematodes are preferred. The current study focused on identifying nematicidal metabolites produced by the fungus Purpureocillium lilacinum cultivated on akaranja deoiled cake-based liquid medium through bioactivity-guided fractionation against Meloidogyne incognita. Column chromatography of the ethyl acetate extract of fungal filtrate exhibited the most potent fraction (fraction 14–15), giving 94.6% egg mass hatching inhibition on the 5th day and a maximum nematicidal activity of 62% against second-stage juveniles after 48 h at 5000 mg/L. Gas chromatography coupled with mass spectrometry (GC-MS) analysis of this fraction revealed five major compounds, viz., 2-ethyl butyric acid, phenyl ethyl alcohol, benzoic acid, benzene acetic acid, and 3,5-Di-t-butylphenol. These biocompounds have potential biocontrol applications in agriculture, but further in vivo studies are warranted.
