Changes in Properties of Pulp Mold Depending on the Forming Conditions of Wet Pulp Mold Manufacturing

In this study, the effects of process conditions of wet pulp mold manufacturing on the properties of the pulp mold were analyzed with a pilot wet pulp mold simulator. Two kinds of pulp stock, HwBKP and ONP pulp, were used in this study. The mesh hole size of the forming wire and the stock concentration were also changed. The results showed that the higher stock concentration led to a higher grammage pulp mold. The ONP pulp stock resulted in a lower grammage pulp mold which could be originated from the lower freeness of the ONP pulp. The grammage of pulp mold was not significantly affected by the mesh hole size of the forming wire. However the mesh hole size had influence on the pulp mold bulk was and the smaller hole size of the mesh resulted in the higher bulk of the pulp mold, which could be caused by changes in the retention of the small components such as ash.

Scalable production of uniform and mature organoids in a 3D geometrically-engineered permeable membrane

The application of organoids has been limited by the lack of methods for producing uniformly mature organoids at scale. This study introduces an organoid culture platform, called UniMat, which addresses the challenges of uniformity and maturity simultaneously. UniMat is designed to not only ensure consistent organoid growth but also facilitate an unrestricted supply of soluble factors by a 3D geometrically-engineered, permeable membrane-based platform. Using UniMat, we demonstrate the scalable generation of kidney organoids with enhanced uniformity in both structure and function compared to conventional methods. Notably, kidney organoids within UniMat show improved maturation, showing increased expression of nephron transcripts, more in vivo-like cell-type balance, enhanced vascularization, and better long-term stability. Moreover, UniMat's design offers a more standardized organoid model for disease modeling and drug testing, as demonstrated by polycystic-kidney disease and acute kidney injury modeling. In essence, UniMat presents a valuable platform for organoid technology, with potential applications in organ development, disease modeling, and drug screening.

Tenuidraconema tongaense n. sp. (Nematoda: Draconematidae), a new free-living marine nematode from a seamount in the southwest Pacific Ocean

Abstract A new species of free-living marine draconematid nematode, Tenuidraconema tongaense n. sp., is described. This new species was collected from shallow subtidal dead corals and detritus from a seamount in a hydrothermal area off the Tonga Islands, southwest Pacific Ocean. Tenuidraconema tongaense n. sp. differs from the known species of the genus by the following combination of characters: longer body (1060-1265 μ m), position of 12 cephalic adhesion tubes (anterior six cephalic adhesion tubes inserted on rostrum and posterior six adhesion tubes inserted on anterior body annule in male; all 12 cephalic adhesion tubes inserted on rostrum in female), number of posterior sublateral adhesion tubes (11 in male, 12 in female) and posterior subventral adhesion tubes (9-11 in male, 10-11 in female) with somatic setae intermingled between the two anteriormost sublateral adhesion tubes, longer spicule length (43-50 μ m), and slender body (a = 41.1-48.6 in male and 29.4-35.1 in female). A pictorial key and a dichotomous identification key for males and females are provided for the species. A table comparing the major differential diagnostic characteristics of the species of Tenuidraconema is presented.

Application of co-culture technology of epithelial type cells and mesenchymal type cells using nanopatterned structures

Various nanopatterning techniques have been developed to improve cell proliferation and differentiation efficiency. As we previously reported, nanopillars and pores are able to sustain human pluripotent stem cells and differentiate pancreatic cells. From this, the nanoscale patterns would be effective environment for the co-culturing of epithelial and mesenchymal cell types. Interestingly, the nanopatterning selectively reduced the proliferative rate of mesenchymal cells while increasing the expression of adhesion protein in epithelial type cells. Additionally, co-cultured cells on the nanopatterning were not negatively affected in terms of cell function metabolic ability or cell survival. This is in contrast to conventional co-culturing methods such as ultraviolet or chemical treatments. The nanopatterning appears to be an effective environment for mesenchymal co-cultures with typically low proliferative rates cells such as astrocytes, neurons, melanocytes, and fibroblasts without using potentially damaging treatments.

Improved chondrogenic performance with protective tracheal design of Chitosan membrane surrounding 3D-printed trachea

Abstract In recent tracheal tissue engineering, limitations in cartilage reconstruction, caused by immature delivery of chondrocyte-laden components, have been reported beyond the complete epithelialization and integration of the tracheal substitutes with the host tissue. In an attempt to overcome such limitations, this article introduces a protective design of tissue-engineered trachea (TraCHIM) composed of a chitosan-based nanofiber membrane (CHIM) and a 3D-printed biotracheal construct. The CHIM was created from chitosan and polycaprolactone (PCL) using an electrospinning process. Upon addition of chitosan to PCL, the diameter of electrospun fibers became thinner, allowing them to be stacked more closely, thereby improving its mechanical properties. Chitosan also enhances the hydrophilicity of the membranes, preventing them from slipping and delaminating over the cell-laden bioink of the biotracheal graft, as well as protecting the construct. Two weeks after implantation in Sprague–Dawley male rats, the group with the TraCHIM exhibited a higher number of chondrocytes, with enhanced chondrogenic performance, than the control group without the membrane. This study successfully demonstrates enhanced chondrogenic performance of TraCHIM in vivo. The protective design of TraCHIM opens a new avenue in engineered tissue research, which requires faster tissue formation from 3D biodegradable materials, to achieve complete replacement of diseased tissue.
