Carbon Dot with pH Independent Near-Unity Photoluminescence Quantum Yield in an Aqueous Medium: Electrostatics-Induced Förster Resonance Energy Transfer at Submicromolar Concentration

We report the synthesis and dynamical behavior of a carbon dot (CD) with near 100% photoluminescence quantum yield in water for a very large pH range (1-12). This CD exhibits a rotational correlational time of only ∼130 ps, signifying the whole CD is not exhibiting photoluminescence. Unlike most carbon-based nanoparticles (which act as a quencher of fluorescence), this CD could act as a donor, and the Förster model could account for the experimental observables for the resonance energy transfer (RET) experiment quite well. Based on two dynamical measurements, it could be shown that the fluorescing moiety is located inside the core of the CD. Importantly, for this CD, RET experiments could be performed with a very low concentration (500 nM) of the acceptor. This kind of electrostatics-driven RET at very low concentration is quite important in bioimaging. This ultrabright CD is nontoxic and useful for bioimaging in mesenchymal stem cells.

A 3D In Vitro Triculture Hybrid Model Recapitulating Tumor Stromal Interaction of Triple‐Negative Breast Cancer as a High Throughput Anticancer Drug Screening Platform

Abstract Breast cancer (BC) progression is substantially driven by cellular cross‐talk between tumor and stromal cells within the tumor microenvironment (TME). However, lack of precise recapitulation of the heterocellular complexity, interactions in oversimplified 2D models, and physiological differences in animal models lead to discrepancy in anticancer drug response. Three‐dimensional (3D) in vitro bioengineered models that physiologically resemble in vivo TME are apt to overcome the discrepancy in the preclinical drug testing outcome. Here, a compartmentalized 3D in vitro triculture triple‐negative breast cancer (TC‐TNBC) model is bioengineered using silk‐fibroin (SF) scaffold and GelMA hydrogel for recapitulating stromal and tumor extracellular niche, respectively. The model features the cellular heterogeneity of stromal niche by incorporating most representative cell types in breast tissue, i.e., adipocytes and endothelial cells, and MDA‐MB‐231 cells in the tumor niche. The TC‐TNBC model exhibits enhanced tumorigenic potential in the presence of stromal cells. Screening of model anticancer drugs (doxorubicin (Dox) and cisplatin (Cis)) exhibits an increase in IC 50 concentration in TC‐TNBC as compared to monoculture, suggesting crucial role of stromal cells in drug response. Drug sensitivity and cytotoxicity behavior mimick the in vivo like drug response. This model provides a facile and adaptable platform to represent tumor‐stromal heterogeneity and high‐throughput anticancer drug screening.

Room-Temperature, Copper-Free, and Amine-Free Sonogashira Reaction in a Green Solvent: Synthesis of Tetraalkynylated Anthracenes and In Vitro Assessment of Their Cytotoxic Potentials

The multifold Sonogashira coupling of a class of aryl halides with arylacetylene in the presence of an equivalent of Cs2CO3 has been accomplished using a combination of Pd(CH3CN)2Cl2 (0.5 mol %) and cataCXium A (1 mol %) under copper-free and amine-free conditions in a readily available green solvent at room temperature. The protocol was used to transform several aryl halides and alkynes to the corresponding coupled products in good to excellent yields. The rate-determining step is likely to involve the oxidative addition of Ar-X. The green protocol provides access to various valuable polycyclic aromatic hydrocarbons (PAHs) with exciting photophysical properties. Among them, six tetraalkynylated anthracenes have been tested for their anticancer properties on the human triple-negative breast cancer (TNBC) cell line MDA-MB-231 and human dermal fibroblasts (HDFs). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed to find out the IC50 concentration and lethal dose. The compounds being intrinsically fluorescent, their cellular localization was checked by live cell fluorescence imaging. 4',6-Diamidino-2-phenylindole (DAPI) and propidium iodide (PI) staining was performed to check apoptosis and necrosis, respectively. All of these studies have shown that anthracene and its derivatives can induce cell death via DNA damage and apoptosis.

Canine Breast Carcinomas: Recent Advances in Diagnostic and Treatment Strategies

Abstract Breast cancer in canines is one of the leading causes of death globally due to client misinterpretation and improper diagnosis and treatment. In past centuries, the diagnosis and treatment of breast carcinoma in dogs followed conventional techniques adopted from human oncology. However, with increasing demand and scientific advancements in the upcoming future, there is an emerging necessity to modernize the diagnostic and treatments in canine breast cancer (CBC) patients. This review explores recent advances in diagnostic techniques and novel therapeutic approaches such as adjuvant‐based targeted therapy, nanomaterial therapy, immune‐based therapy, adoptive cell therapy, tumor vaccine, oncolytic virotherapy, and the use of noncoding RNAs in CBCs. In addition, the review discusses the healthcare policies aimed at improving diagnostic and therapeutic efficacy and future directions for translation from human oncology into veterinary oncology. By adopting these modern strategies, the quality of care can be significantly enhanced by translating them into practical applications with better outcomes and improved survival rates for canine patients.

Tunable and improved antiproliferative activity of Pt (II)‐based organometallics bearing alkynyls and 1,2,3‐triazole moieties

Abstract Synthesis and characterization of a set of four Pt (II)‐based organometallic compounds ( 7–10 ) are being described. These molecules are unique since each of these contain three biologically active structural motifs, in the form of a pyrazine ring that connects a Pt‐alkynyl and a triazole ring. These organoplatinum molecules have been characterized using multinuclear nuclear magnetic resonance (NMR) ( 1 H, 13 C{ 1 H}, and 31 P{ 1 H}), Fourier transform infrared spectroscopy (FT–IR), and mass spectrometry (MS). Additionally, the antiproliferative activities of 7–10 were probed against human triple‐negative breast cancer cells (MDA‐MB‐231) and normal human dermal fibroblast cells (HDF).

Isolation and Establishment of Mesenchymal Stem Cells from Wharton’s Jelly of Human Umbilical Cord

Mesenchymal stem cells (MSCs) are currently considered as 'medicinal signaling cells' and a promising resource in regard to cell-based regenerative therapy. Umbilical cord is a human term perinatal tissue which is easily attainable, and a promising source of stem cells with no associated ethical concerns. MSCs have been isolated from different regions of the umbilical cord and Wharton's jelly (WJ) is the gelatinous matrix that surrounds and provides protection to the umbilical cord blood vessels. Being more primitive, MSCs from human umbilical cord exhibit greater proliferative capacity and immunosuppressive ability as compared to adult stem cells which gives them a therapeutic advantage. To meet the requirements for cell therapy, it is important to generate MSCs at a clinical scale by following steps which are not time consuming or labor intensive. Here we present a simple, efficient protocol for isolation of MSCs from WJ of human umbilical cord by explant culture method which is reproducible and also, cost effective.
