The antifilarial activity of a synthetic agent, methyl-5 (6) piperidino-carbonyl benzimidazole-2-carbonate (C.D.R.I. Comp. No. 81/504) was evaluated against three different filarial species in rodents at various dose levels. It exerted 100 percent microfilaricidal action against Litomosoides carinii at 3 mg/kg, i.p. x 5 days in cotton rats and at 50 mg/kg i.p. x 5 days against Dipetalonema viteae and Brugia malayi in mastomys, on day 35,42 and 56 respectively. All the adults of L. carinii died at 10 mg/kg while worms of other two species, D. viteae and B. malayi were killed respectively at 12.5 and 50 mg/kg x 5 days. At 3000 mg/kg by oral route and at 300 mg/kg by i.p. route, the agent did not produce any untoward reactions in mice. The compound thus deserves further investigations.

Protein kinases in different life stages of Brugia malayi and other filarial worms.

The investigation of protein kinase activity in different life stages of a few filarial worms indicates the presence of different protein kinases and of dissimilarities between the distribution pattern of these enzymes during the development. Phosvitin turned out to be the preferred exogenous acceptor protein, when homogenates of adults of Brugia malayi, Dipetalonema viteae, Setaria cervi and Litomosoides carinii were used as source of enzyme. In contrast to adults the L3-larval and the microfilarial stages demonstrated much lower phosvitin phosphorylating activity and the preference for acidic and basic proteins as phosphate acceptors was about equal. The occurrence of cyclic AMP-dependent protein kinase was established for adults of B. malayi.

Comparative antifilarial efficacy of the N-oxides of diethylcarbamazine and two of its analogues.

The N-oxides of 1-diethyl carbamoyl-4-methyl piperazine (DEC), 3-ethyl-8-methyl, 1,3,8-triazabicyclo (4,4,0) decan-2-one (centperazine) and 1-methyl-4-(pyrrolidin-1-yl-carbonyl) piperazine (CDRI Comp. 72/70) have been evaluated against Limotosoides carinii infection in cotton rats to establish whether conversion of the three antifilarials into N-oxides would lead to exertion of better antifilarial activity. Of the three N-oxides, N-oxide of DEC showed significantly more suppressive effect on circulating microfilariae in comparison to its parent compound. However, adult worms were unaffected. It was observed that basicity of the N-CH3 group did not play major role in exertion of activity of DEC and related compounds. Nevertheless, two other N-oxides were inactive.

A new approach in study of bioenergetics of macrofilariae using Nuclear magnetic resonance(NMR) spectroscopy

Nuclear magnetic resonance (NMR) offers new and exciting ways for examining biological phenomena in vivo and H, 13C and 31P NMR spectroscopy has been employed in metabolic studies on intact cells, tissues and organs as well as on whole organisms. In the present study 31PNMR studies were conducted to understand the effect of various drugs on the energy metabolism of filarial parasite. The 31PNMR at 22(C during perfusion with Ringer's solution saturated with N2 containing 5% CO: were composed of 12 peaks. Based on the chemical shift analysis of the intact parasites, the phosphorus component included sugar phosphates, nucleoside diphosphosugars, glycerophosphoryl choline, glycerophosphoryl ethanolamine, phosphoenolpyruvate and nucleotides mono, di and tri phosphates etc. Further standardisation of the perfusion method with normal adult filariids and male, female adult filarial worms separately has been carried out. In vivodrug administration using DEC (standard microfilaricidal drug for filariasis) at 250 mg/kg, i.p. x 5 days showed changes in the content of the 31PNMR peaks as compared with spectra of normal parasites.

Stage-specific antibody response of Mastomys natalensis during the course of Dipetalonema viteae infection.

Stage-specific haemagglutinating (HA) and precipitin antibodies (PA) to infective larvae (L3), adult worms and microfilariae (mf) have been demonstrated in sera of Mastomys natalensis during the course of Dipetalonema viteae infection using indirect haemagglutination (IHA) and Ouchterlony's gel-diffusion tests. L3-specific HA antibody titre was shown to be quite low and lasted for shorter period whereas adult-specific antibody response was significantly higher and persisted for longer duration (beyond day 240 post exposure). No precipitin antibody to L3 stage could be detected, however, significant adult-specific antibody was detected which ultimately tapered off by day 210 post exposure (p.e.). In contrast, mf-specific PA which appeared at the beginning of patency, never disappeared even at the late stage of infection. Mf-specific HA antibody appeared at early incubation period (day 15 p.e.) and after exhibiting two peaks, one in the midst of prepatent period (day 30 p.e.) and the other on day 180 p.e. persisted at low level during the late stage of infection when amicrofilaraemia developed.
