[Combination of acupuncture therapy and kinesiotherapy: an example of pain rehabilitation].

The effect mechanism and the characteristics of clinical application of the combination of acupuncture therapy and kinesiotherapy are explored for pain rehabilitation. Analgesia of acupuncture therapy and kinesiotherapy is characterized by multiple targets and dimensions. The paper summarizes the effect mechanisms of these two therapies and their synergistic action and investigates the current situation and clinical characteristics of their combination in treatment of musculoskeletal pain, visceral pain and postoperative pain so that the references can be provided for the selection of pain rehabilitation programs.

The leading edge during dorsal closure as a model for epithelial plasticity: Pak is required for recruitment of the Scribble complex and septate junction formation

Dorsal closure (DC) of the Drosophila embryo is a model for the study of wound healing and developmental epithelial fusions, and involves the sealing of a hole in the epidermis through the migration of the epidermal flanks over the tissue occupying the hole, the amnioserosa. During DC, the cells at the edge of the migrating epidermis extend Rac- and Cdc42-dependent actin-based lamellipodia and filopodia from their leading edge (LE), which exhibits a breakdown in apicobasal polarity as adhesions are severed with the neighbouring amnioserosa cells. Studies using mammalian cells have demonstrated that Scribble (Scrib), an important determinant of apicobasal polarity that functions in a protein complex, controls polarized cell migration through recruitment of Rac, Cdc42 and the serine/threonine kinase Pak, an effector for Rac and Cdc42, to the LE. We have used DC and the follicular epithelium to study the relationship between Pak and the Scrib complex at epithelial membranes undergoing changes in apicobasal polarity and adhesion during development. We propose that, during DC, the LE membrane undergoes an epithelial-to-mesenchymal-like transition to initiate epithelial sheet migration, followed by a mesenchymal-to-epithelial-like transition as the epithelial sheets meet up and restore cell-cell adhesion. This latter event requires integrin-localized Pak, which recruits the Scrib complex in septate junction formation. We conclude that there are bidirectional interactions between Pak and the Scrib complex modulating epithelial plasticity. Scrib can recruit Pak to the LE for polarized cell migration but, as migratory cells meet up, Pak can recruit the Scrib complex to restore apicobasal polarity and cell-cell adhesion.

The approximately 300 distinct neurons comprising each hemineuromere of the Drosophila embryonic central nervous system are derived from a segmentally reiterated array of approximately 30 progenitor cells, neuroblasts (NBs). Each NB has a unique identity and undergoes repeated cell divisions to produce several smaller secondary precursor cells, ganglion mother cells (GMCs); each GMC divides once to produce two neurons and/or glia, thereby generating a specific lineage of neurons/glia. Understanding the generation of neuronal diversity requires not only elucidation of the molecules and mechanisms that specify NB identity but also those that act to differentiate between the cell types produced within one NB lineage. Here we show that the Drosophila Zn finger protein Klumpfuss (Klu), which shows sequence similarities to the mammalian Wilm's tumor suppressor (WT-1), acts to differentiate between the identities of the first two secondary precursor cells produced from one NB lineage. Klu is expressed in the NB4-2 lineage only after two rounds of NB cell division, in the second born GMC (GMC4-2b). In loss-of-function mutant embryos, the first born GMC (GMC4-2a) as well as its progeny neurons are duplicated; we show that this duplication of the GMC4-2a sublineage arises because GMC4-2b adopts the identity of GMC4-2a and divides to produce the GMC4-2a progeny. Moreover, when Klu is ectopically expressed in GMC4-2a, it fails to acquire its normal identity and fails to produce correctly specified progeny. klu therefore acts to specify the identity of GMC4-2b and to make it distinct from GMC4-2a. Our findings further suggest that the determination of GMC cell fate occurs in two steps; the initial GMC identity is the consequence of inheritance from the maternal NB, however, the subsequent stabilization of this identity requires functions like klu in the GMC.

Phosphotyrosyl phosphatase activator facilitates Miranda localization through dephosphorylation in dividing neuroblasts

The mechanism for the basal targeting of the Miranda (Mira) complex during the asymmetric division of Drosophila neuroblasts (NBs) is yet to be fully understood. We have identified conserved Phosphotyrosyl Phosphatase Activator (PTPA) as a novel mediator for the basal localization of the Mira complex in larval brain NBs. In ptpa NBs, Mira remains cytoplasmic during early mitosis where its basal localization is delayed until anaphase. Detailed analyses indicate that PTPA acts independently of, and prior to, aPKC activity to localize Mira. Mechanistically, our data show that the phosphorylation status of the Thr591 (T591) residue determines the subcellular localization of Mira and that PTPA facilitates the dephosphorylation of T591. Furthermore, PTPA associates with the Protein Phosphatase 4 complex to mediate Mira localization. Based on these results, a two-step process for Mira basal localization during NB division is revealed where PTPA/PP4-mediated cortical association followed by apical aPKC-mediated basal restriction.

Experimental of Longhua capsule on the thrombocytopenia model caused by CTX

Objective To observe the effects of Longhua Capsule on the thrombocytopenia Model caused by CTX.Methods The 40 SD rats were randomly divided into 4 groups: the control group,the model group,the Longhua capsule group and the interleukins-11 group,each group with 10 rats.Except the control group,the other groups were made into models with the injection of CTX into the abdominal cavities.After the success of making models,the rats of the Longhua capsule group were administered by gavages with the Longhua capsule solution(460 mg/kg),once a day.The rats in the interleukins-11 control group were injected subcutaneously with interleukins-11[13 μg/(kg·d)].The rats in the control group and model group were administered by gavages with the same volume of 0.9% sodium chloride solution,once a day,for 14 days.The platelet count,marrow megalocaryocyte count,serum TPO and IL-3 of the rats in each group were compared before and after the administration.Results There were marked difference between the rats of the Longhua capsule group,the interleukins-11 group and model group on the platelet count,marrow megalocaryocyte count,,serum TPO and IL-3 levels(P0.05);there was no marked difference between the Longhua capsule group,the interleukins-11 group and control group(P0.05).Conclusion Longhua capsules may improve the general condition of thrombocytopenia,raise the platelet count,promote the rise of marrow megalocaryocyte count,and increase the levels of serum TPO and IL-3 of SD rats in the model group.

PRL-1 is required for neuroprotection against olfactory CO₂ stimulation in Drosophila

Abstract The Mammalian phosphatase of regenerating liver (PRL) family is primarily recognized for its oncogenic properties. Here we found that in Drosophila, loss of prl-1 resulted in CO2-induced brain disorder presented as irreversible wing hold up with enhancement of Ca2+ responses at the neuron synaptic terminals. Overexpression of Prl-1 in the nervous system could rescue the mutant phenotype. We show that Prl-1 is particularly expressed in CO2-responsive neural circuit and the higher brain centers. Ablation of the CO2 olfactory receptor, Gr21a, suppressed the mutant phenotype, suggesting that CO2 acts as a neuropathological substrate in absence of Prl-1. Further studies found that the wing hold up is an obvious consequence upon knockdown of Uex, a magnesium transporter, which directly interacts with Prl-1. Conditional expression of Uex in the nervous system could rescue the phenotype of prl-1 mutants. We demonstrate that Uex acts genetically downstream of Prl-1. Our findings provide important insights into mechanisms of Prl-1 protection against olfactory CO2 stimulation induced brain disorder at the level of detailed neural circuits and functional molecular connections.
