Soybean GmSAUL1, a Bona Fide U-Box E3 Ligase, Negatively Regulates Immunity Likely through Repressing the Activation of GmMPK3

E3 ubiquitin ligases play important roles in plant immunity, but their role in soybean has not been investigated previously. Here, we used Bean pod mottle virus (BPMV)-mediated virus-induced gene silencing (VIGS) to investigate the function of GmSAUL1 (Senescence-Associated E3 Ubiquitin Ligase 1) homologs in soybean. When two closely related SAUL1 homologs were silenced simultaneously, the soybean plants displayed autoimmune phenotypes, which were significantly alleviated by high temperature, suggesting that GmSAUL1a/1b might be guarded by an R protein. Interestingly, silencing GmSAUL1a/1b resulted in the decreased activation of GmMPK6, but increased activation of GmMPK3 in response to flg22, suggesting that the activation of GmMPK3 is most likely responsible for the activated immunity observed in the GmSAUL1a/1b-silenced plants. Furthermore, we provided evidence that GmSAUL1a is a bona fide E3 ligase. Collectively, our results indicated that GmSAUL1 plays a negative role in regulating cell death and immunity in soybean.

Functional analysis of the seven in absentia ubiquitin ligase family in tomato

Abstract Seven in absentia (SINA) protein is one subgroup of ubiquitin ligases possessing an N‐terminal cysteine‐rich really interesting new gene (RING) domain, two zinc‐finger motifs, and a C‐terminal domain responsible for substrate‐binding and dimerization. In tomato ( Solanum lycopersicum ), the SINA gene family has six members, and we characterize in this study all tomato SINA ( SlSINA ) genes and the gene products. Our results show that SlSINA genes are differentially regulated in leaf, bud, stem, flower, and root. All SlSINA proteins possess RING‐dependent E3 ubiquitin ligase activity, exhibiting similar specificity towards the E2 ubiquitin‐conjugating enzyme. SlSINA1/3/4/5/6 are localized in both cytoplasm and nucleus, whereas SlSINA2 is exclusively localized in the nucleus. Moreover, all SlSINAs can interact with each other for homo‐ or hetero‐dimerization. The functionality of SlSINA proteins has been investigated. SlSINA4 plays a positive role in defense signalling, as manifested by elicitation of E3‐dependent hypersensitive response‐like cell death; the other SlSINAs are negative regulator and capable to suppress hypersensitive response cell death. Transgenic tomato plants overexpressing SlSINA2 exhibit pale‐green leaf phenotype, suggesting SlSINA2 regulates chlorophyll level in plant cells, whereas transgenic tomato plants overexpressing SlSINA5 have altered floral structure with exserted stigma, implicating SlSINA5 plays a role in flower development.

Probiotics in 14-day triple therapy for Asian pediatric patients with Helicobacter pylori infection: a network meta-analysis

Although 14-day triple therapy has been widely administrated for eradicating Helicobacter pylori (H. pylori) in Asia, its antibiotic-associated side effects restrict the effectivity of eradication therapy in pediatric patients. Therefore, a network meta-analysis (NMA) was conducted to compare the efficacy and safety of probiotics supplemented in 14-day triple therapy in Asian pediatric patients.Randomized controlled trials (RCTs) were retrieved comprehensively in electronic databases (such as PubMed, Cochrane library, Embase, CNKI, Wan fang database, VIP database and CBM) until April 2017. Additional references were obtained from reviewed articles. NMA was performed using a random-effects model under a frequentist framework.Seventeen RCTs were included. NMA indicated that Bifidobacterium infantis+Clostridium butyricum was most beneficial for H. pylori eradication rates (P-score = 0.82) and Bacillus mesentericus+Clostridium butyricum+Streptococcus faecalis for total side effects (P-score = 0.77). Taken together, Bacillus mesentericus+Clostridium butyricum+Streptococcus faecalis was the best one to supplement in 14-day triple therapy due to its efficacy (P-score = 0.72) and safety (P-score = 0.77). Additionally, pairwise meta-analysis indicated that probiotics supplemented 14-day triple therapy significantly increased H. pylori eradication rates (RR: 1.16, 95%CI: 1.07-1.26) and reduced the incidence of total side effects (RR: 0.40, 95%CI: 0.34-0.48) compared with placebo.Bacillus mesentericus+Clostridium butyricum+Streptococcus faecalis is the optimal probiotic regime of reducing total side effects and improving eradication rates when supplemented 14-day triple therapy. Further direct evidence is needed to warrant it.

The changes in the production of cytokines in spenocytes of BALB/c mice after immunization with recombinant Bifidobacterium bifidum(Bb)-ESAT-6-MPT64 vaccine of Mycobacterium tuberculosis were dynamically observed in the present study,in which BALB/c mice were immunized with recombinant Bb-ESAT-6-MPT64 vaccine intranasally and subcutaneously each 4 mice were killed before and 2,4,6,8 and 10 weeks after immunization respectively,and the lymphocytes suspensions from spleen were prepared and cultivated in vitro with MTB-Ag and Con-A to induce the production of IL-12,IL-10 and IFN-γ,while,MTB and LPS were used to induce the production of TNF-α.ELISA assay was used to detect the presence of various cytokines in the lymphocyte suspensions.The experimental results showed that the levels of IFN-γ,IL-12,TNF-α and IL-10 in group of mice inoculated with vaccine rose up 2-10 weeks after immunization,reaching their peaks during 6-8 weeks.On the same time point,the cytokine levels induced by Con-A or LPS were comparatively higher than that induced by MTB-Ag(P0.05 or 0.01).It is concluded that the mixed type of immne responses is induced at early course of immunization after inoculation with recombinant Bifidobacterium bifidum-ESAT-6-MPT64 vaccine in mice.

A highly efficient transient protoplast system for analyzing defence gene expression and protein–protein interactions in rice

SUMMARY The transient assay system based on mesophyll or cultured cell-derived protoplasts has been exploited in several plant species and has become a powerful tool for rapid gene functional analysis and biochemical manipulations. However, the system has not been widely used in rice owing to the difficulties in large-scale isolation of viable rice protoplasts from leaves or suspension-cultured cells. Here, we describe a significantly improved method to isolate a large number of protoplasts from stem and sheath tissues of both young and mature plants. High-level coexpression of multiple constructs and efficient suppression of exogenous and endogenous genes were observed in the stem- and sheath-derived protoplasts. A transient green fluorescent protein and luciferase-based reporter system for defence-related genes expression analysis has been established, which is useful for screening and characterizing genes involved in rice defence signalling pathways. Furthermore, a protoplast-based bimolecular fluorescence complementation (BiFC) system for the detection of protein-protein interactions in living rice cells was developed. The YFP complementation of two split-YFP halves mediated by homodimerization of the GUS and SPIN1, a cell-death related protein, was observed in transfected protoplasts. In combination with genetic, genomic and proteomic approaches, the established versatile protoplast transient assay system will facilitate large-scale functional analysis of defence-related genes in rice.
