The combined effect of UV radiation and ultrasonic cavitation on saprophytic and conditionally pathogenic microflora of cold-water ultrasound

The paper considers the combined effect of ultraviolet radiation (UV) and ultrasonic cavitation (ultrasonic) as part of ultrasonic filtration systems on representatives of saprophytic and conditionally pathogenic microflora of water. Ultrasound examination (25 kHz) is a promising method of purification in aquaculture due to the possibility of inactivation of microorganisms in agglomerates that are not exposed to UV. As a result of the study of the cultivated microflora, it was found that ultraviolet radiation and the combined effect of UV and ULTRASOUND have a significant effect on the quantity and quality of saprophytic microflora of water. It is shown that the studied modes of operation led to a significant decrease (at p <0.05) in the occurrence of the genus Escherchia relative to the control (without exposure to ultrasound and UV). The total contamination of UV water decreased to 1.2×102 CFU/ml with UV operation and to 1.1×102 CFU/ml with the combined action of UV and ultrasound. The UV sterilizer showed low efficiency in relation to Enterobacter cloacae, while the combined effect of UV and ultrasound led to a significant decrease in CFU to 0.94 ± 0.05 Log10.

Evaluation of secretory activity in carp pancreatocytes (Cyprinus carpio L.) when using lactobacillus-based chelate and probiotic supplemental feed complexes

The experimental research was carried out on the juvenile carp (Cyprinus carpio L.). The impact from supplemental feeds consisting of variable concentrations of chelate compounds, biogenic trace elements and probiotic lactobacillus-based product Bacillus subtilis VKPM B-2335 was evaluated. Optical and qualitative parameters of the lactobacillus base were studied in order to identify the major group of substances potentially able to influence the end result. The purpose of this research was to identify changes in the structure of the zymogen granules and their dimensions at which supplemental feeds produce a stimulating effect on the synthesis of zymogens in exogenous cells of the secretory part of pancreas. At the outcome of the study, for the first time, it was possible to prove that the integrated action of chelates and lactobacillus-based probiotics complemented each other. Metal chelate compounds contributed to enlargement of the zymogen granules, if compared to the control values. The bacterial products accelerated production of the zymogen granules in acinar cells diffusely located in carp hepatopancreas.

Evaluating the UV radiation effectiveness in industrial aquaculture

Abstract The paper presents an analysis of the bacterial purification effectiveness by UV radiation of wastewater in modern industrial aquaculture recirculation systems. the effectiveness of water purification in the ultrasound system by a set of filters for mechanical treatment and ultraviolet radiation is experimentally established. UV lamp allows to process water with rays that effectively destroy the organic matter formed in it. Pathogenic and conditionally pathogenic genera, such as: Aeromonas, Pseudomonas, Staphylococcus, Vibrio, Enterobacteria were differentiated.

Amphibian and fish eye lens used as biomarker of remote and chronic environmental contamination

The lens of vertebrates has a general structural plan and reacts similarly to various factors of anthropogenic impact. Rainbow trout (Oncorhynchus mykiss) and grass frog (Rana temporaria L.) are the most well-studied objects of aquatic toxicology in Russia, which react to the level of exposure to toxicants in the urbanized environment and retain the consequences of this exposure. The study aims to determine the possibility of using the lens of fish and amphibians as a biomarker of chronic and long-term background environmental contamination. The fundamental importance of the work lies in the fact that for the first time the mechanisms of the response of the epithelial cells of the eye lens and lens fibers to the spectrum of various toxicants entering the environment with industrial effluents are considered. In chronic experiments, the sensitivity of the lens of Oncorhynchus mykiss fingerlings to three toxicants (dibutyl sebacate-DBS, polyterpenes-PTP, and 3-aminopropyltriethoxysilane-AGM-9) in concentrations close to the normalized valuesin the environment was studied. On adult individuals of Rana temporaria, the appearance of abnormities in the lens was studied over a long period, depending on the total environmental contamination. To assess remote environmental contamination, studies were carried out on the constantly growing lens of the grass frog, which can preserve the resulting fiber abnormities throughout its life in the form of local opacities from the harmful environmental factors impacts. On the epithelium of the lens of fish, it was found that under the influence of certain pollutants, the mitotic index (MI) in the germinal area can change over a wide range. With the combined effect of toxicants, inhibition of mitosis is also observed. Using biomicroscopy of the eye, it was possible to show that frogs from the urbanized zone of Moscow have 1.5 times more genotypic and modification changes in the lens than their relatives taken from an ecologically clean zone of the Moscow region, Mozhaisky district in the water area of the Ozerninskoye reservoir. For the first time, the sensitivity of the lens of aquatic organisms to chronic effects under experimental and background contamination of the terrestrial and aquatic environment has been shown. Therefore, taking into account the uniformity of the structure of the eye lens of fish and amphibians, as well as in other vertebrates, the lens can be used as a biomarker for assessing chronic and remote environmental contamination. The research results can be applied for a comprehensive assessment of the action of anthropogenic factors.

CHANGES IN ONTOGENETIC PARAMETERS IN DAPHNIA MAGNA INFLUENCED BY SOLUTIONS OF BENZENE COMPOUNDS

The article presents the research results of biological parameters of Daphnia magna development at the critical stages of ontogenesis influenced by the aqueous solutions of ecotoxicants: nitrobenzene and trichlorobenzene. The periods of reproduction and development in Daphnia magna are stated to be decisive, among other parameters (physiological, biochemical). In fact, the entire period of embryogenesis in Daphnia is quite vulnerable, compared to the further stages of development. Under the action of solutions of ecotoxicants taken for the research Daphnias have a decreased fertility and a delayed oogenesis. The next stage of sensitivity is the number of cast carapaces. The least sensitive parameter is the survival rate of adults. Increasing the remote toxic effect in three generations of Daphnias was not detected in the course of research. The Daphnia magna test object showed high sensitivity to nitrobenzene and trichlorobenzene acting as water environment pollutants in all the experiments. For the main ontogenetic parameters in the assessment of water quality it is preferable to take fecundity and violation of development processes at the pre-natal stages of Daphnia magna. According to the survival parameter, the maximum permissible concentration of the studied benzene compounds was 0.01 mg/l, which was determined by the most sensitive parameters and critical stages of ontogenesis

Toxicity of metal chelates mixture in aquatic environment at Danio rerio

Abstract In this paper, the toxic effect of a complex chelate mixture containing microelements (Fe, Mn, Zn, Se, I, Cu) on Danio rerio was investigated. Chelated compounds are used to detoxify elements in the aquatic environment, as well as feed additives for various types of farm animals. The effect of chelate complexes was studied according to the following parameters: LC50, survival in chronic experience (30 days), embryotoxicity and genotoxicity by the micronucleus test method. The established LC50 value was 2.73 mg/l, the maximum allowable concentration that does not have a negative effect on adults fish and eggs was 0.5 mg/l. The genotoxic effect on erythrocytes of fish blood was not found in the entire range of sublethal concentrations. Comparison of the data obtained on the toxicity in this study allows asserting that the toxicity of chelates is lower than that of the ionic forms of the elements, subject to their complex effect. To accurately determine the safe level of exposure to chelates, additional studies on other organisms of the hydrobiocenosis are required.

Temperature differentiation of aquatic microflora of a closed water supply system by the example of incubation of microbiological crops at 21 and 37 °C

Abstract In this paper, the dependence of the temperature conditions of incubation of microbiological water cultures taken directly from the pool of a closed water supply system on the activity and species diversity of the resulting microflora is analyzed. The object of the study was clarias catfish (Clarias gariepinus), which was grown at a temperature of 22-24 °, Ph 6.5-7.5, which corresponds to the optimal requirements for breeding this type of fish. The study was conducted on the basis of the Russian-Norwegian Center for Research in the field of Innovation and Aquaculture of the MSUTM named after K.G. Razumovsky. As a result of the study, a reliable regularity of the activity of aquatic microflora in CFU / ml, as well as generic and species affiliation, was revealed.

SPECIFIC FEATURES OF EMBRYONIC DEVELOPMENT OF POND SNAIL (LYMNAEA STAGNALIS L.) UNDER ANTHROPOGENIC INFLUENCE OF CHLORIDE COMPOUNDS IN WATER

The article highlights the research of embryonic development of hydrobionts under the influence of anthropogenic factors of water solutions – chlorobenzene and tetrachloroethylene. To detect the toxicity of chloride pollutants in a freshwater reservoir, representatives of benthic mollusks (big pond snail Lymnaea stagnalis L.) were used as a model organism. According to the results of research, it was found that at a concentration of 0.01 mg/l of chlorobenzene solution individual anomalies of embryonic development of the pond fish are observed, but at higher concentrations its development is delayed by 5-6 stages in comparison with the control. As an integral parameter of pond fish development, we can take indicators of the biological toxicity criterion: Hatchling and its survival over a 10-day period after leaving the egg shells. Changes in the physiological parameters of pond fish embryos begin with a chlorobenzene concentration of 0.01 mg/l; the maximum permissible concentration for tetrachloroethylene for the considered biological indicators should be considered 0.05 mg/l. It is noted that in big pond snail (Lymnaea stagnalis L.) the hatch of juveniles from egg shells is the most sensitive indicator when determining the toxicity of an aqueous solution; and less sensitive indicators are the number of eggs laid and the survival of juveniles in the first 10 days after hatching. The test object used is a large pond, which has shown a high sensitivity to contamination of the water environment with chlorinated ecotoxicants and can be recommended for a comprehensive quality assessment when biotesting freshwater waters

Biocenosis of cold-water and warm-water biofilter in recirculating aquaculture system

Abstract This article presents data on the species composition of periphyton organisms in biofilms of cold-water biological filtration systems intended for the cultivation of rainbow trout ( Oncorhynchus mykiss ) and warm-water species for the cultivation of ( Clarias gariepinus ) in a recirculating aquaculture system. Data on the occurrence of certain groups of periphyton organisms are presented, and their role in the process of water purification is indicated. A significant difference in the species composition of biofilter biofilm organisms is shown, depending on: temperature regime and planting density of aquaculture objects. The species composition of aquasysets is similar to that in urban wastewater treatment systems.
