Polyelectrolyte Gel Swelling and Conductivity vs Counterion Type, Cross-Linking Density, and Solvent Polarity

A joint theoretical and experimental study is devoted to the swelling and electrical conductivity of highly charged polyelectrolyte (PE) gels in media of different polarity. Deprotected poly(tert-butyl carbamate l-alanine) gels provided with fluorine, bromine, chlorine, sulfate, and trifluoroacetic counteranions demonstrated different conductivity in aqueous medium as well as solvent uptake in water/methanol mixtures. Counterion specificity in PE gel properties was theoretically explained in the framework of the model which treats ion association within the network as a two-stage process. Manning-type condensation, being the first condensation stage not affected by counterion type, provides understanding of a gel maximal swelling at intermediate degrees of gel ionization f and a slight gel contraction at f tending to unity, which were earlier observed in experimental investigations. The second ion association stage is an ion pairing influenced by the kind of counterion. Since a considerable fraction of ions form pairs even in a swollen hydrogel, its properties are largely influenced by counterion type. Conductivity of the gel was calculated theoretically owing to the known fraction of free and condensed counterions. Developed theory containing no adjusting parameters allows for the first time to reach semiquantitative matching of experimental results on highly charged gel swelling and conductivity.

Side-Chain Amino Acid-Based Cationic Antibacterial Polymers: Investigating the Morphological Switching of a Polymer-Treated Bacterial Cell

Synthetic polymer-based antimicrobial materials destroy conventional antibiotic resistant microorganisms. Although these antibacterial polymers imitate the properties of antimicrobial peptides (AMPs), their effect on bacterial cell morphology has not been studied in detail. To investigate the morphology change of a bacterial cell in the presence of antimicrobial polymer, herein we have designed and synthesized side-chain amino acid-based cationic polymers, which showed efficient antibacterial activity against Gram-negative (Escherichia coli), as well as Gram-positive (Bacillus subtilis) bacteria. Morphological switching from a rod shape to a spherical shape of E. coli cells was observed by field emission-scanning electron microscopy analysis due to cell wall disruption, whereas the B. subtilis cell structure and size remained intact, but stacks of the cells formed after polymer treatment. The zone of inhibition experiment on an agar plate for E. coli cells exhibited drastic morphological changes at the vicinity of the polymer-treated portion and somewhat less of an effect at the periphery of the plate.

Amino acids, the natural building blocks of life, are highly functional small molecules with chirality and important catalytic properties. Imparting the exquisite properties of amino acids into the macromolecules allows us to prepare unique, well-defined, biocompatible, functional, and tailorable materials. They exhibit stimuli responsiveness, tunable optical characteristics, enhanced thermal, mechanical, and biological properties with desired cell-materials interactions. Considering the salient features and diverse applications of side-chain amino acid-containing polymers, in this feature article, we present a concise highlight on the recent trends (2015–present) of the synthesis and utilization of pendant amino acid-based polymeric architectures. This article covers the synthesis of different classes of side-chain amino acid-derived polymeric architectures, and their recent utilization in drug and gene delivery, antibacterial activity, anti-fouling activity, heavy metal ion sensing, tissue engineering, etc. We hope that this study will provide a comprehensive outline of key aspects associated with the construction of functional 'bio-inspired' polymeric materials with desired characteristics, and their enhanced potential applications.

Zwitterionic Polysulfobetaine Inhibits Cancer Cell Migration Owing to Actin Cytoskeleton Dynamics

Cell migration is an essential dynamic process for most living cells, mainly driven by the reorganization of actin cytoskeleton. To control actin dynamics, a molecular architecture that can serve as a nucleator has been designed by polymerizing sulfobetaine methacrylate. The synthesized zwitterionic polymer, poly(sulfobetaine methacrylate) (PZI), effectively nucleates the polymerization process of G-actin and substantially accelerates the rate of polymerization. Isothermal titration calorimetry (ITC) and bioinformatics analysis indicated binding between PZI and monomeric G-actin. Thus, in vitro actin dynamics was studied by dynamic light scattering (DLS), pyrene-actin polymerization assay, and total internal reflection fluorescence microscopy (TIRFM). Furthermore, a 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) fluorophore-containing monomeric unit was incorporated into the sulfobetaine zwitterionic architecture to visualize the effect of polymer in the cellular environment. The BODIPY-containing zwitterionic sulfobetaine polymer (PZI-F) successfully penetrated the cell and remained in the lysosome with minimal cytotoxicity. Confocal microscopy revealed the influence of this polymer on the cellular actin cytoskeleton dynamics. The PZI-F polymer was successfully able to inhibit the collective migration of the human cervical cancer cell line (HeLa cell) and breast cancer cell line (MDA-MB-231 cell), as confirmed by a wound healing assay. Therefore, polyzwitterionic sulfobetaine could be explored as an inhibitor of cancer cell migration.

Precision Control of Temperature Response by Copolymerization of Di(Ethylene Glycol) Acrylate and an Acrylamide Comonomer

Abstract The readily tuned thermoresponsive behavior of well‐defined statistical and block copolymers of di(ethylene glycol)ethyl ether acrylate (DEGA) and N , N ‐dimethylacrylamide (DMA) was investigated. DEGA and DMA were copolymerized with various comonomer feed ratios by reversible addition–fragmentation chain transfer (RAFT) polymerization. Reactivity ratios were determined, which allowed copolymers with precisely tuned compositions to be prepared. The cloud point of the resulting copolymers was observed to vary linearly with the mole fraction of hydrophilic DMA over the range of 10–90 °C. Block copolymers of DEGA were also prepared by chain extension of poly(DEGA) homopolymers with DMA. The resulting block copolymers formed well‐defined polymeric micelles when heated above the cloud point of the poly(DEGA) block. Thus, copolymerization of DEGA was shown to be a convenient route to thermoresponsive polymers with specific and predictable transition temperatures.

Cationic methacrylate polymers containing chiral amino acid moieties: controlled synthesis via RAFT polymerization

Two methacrylate containing amino acid based chiral monomers, Boc-L-alanine methacryloyloxyethyl ester (Boc-Ala-HEMA) and Boc-L-phenylalanine methacryloyloxyethyl ester (Boc-Phe-HEMA), were polymerized by the reversible addition–fragmentation chain transfer (RAFT) process to afford well-defined amino acid based polymers with controlled molecular weight, narrow molecular weight distribution, and precise chain end structure. Linear pseudo-first-order kinetics and number average molecular weight dependence on conversion were observed for the RAFT polymerizations. Di-block copolymers were prepared by RAFT polymerization of methyl methacrylate using poly(Boc-L-alanine methacryloyloxyethyl ester) or poly(Boc-L-phenylalanine methacryloyloxyethyl ester) as macro-chain transfer agents. Subsequent deprotection of the Boc group produced architectures with a primary amine moiety at the side chain, which showed pH responsiveness. The specific rotations of both the chiral monomers and corresponding homo- and block copolymers were studied and alteration of specific rotation due to transformation of monomer to polymer was noticed. In addition, the surface charge of various polymeric architectures was studied using dynamic light scattering (DLS) measurements. The self-assembly of block copolymer films was investigated by atomic force microscopy (AFM). These pH-responsive cationic polymers can potentially be extended for the delivery of small interfering RNA.

Helical Superstructures from the Hierarchical Self-Assembly of Coil–Coil Block Copolymer Guided by Side Chain Amyloid-β(17–19) LVF Peptide

The rational design of precisely controlled hierarchical chiral nanostructures from synthetic polymers garnered inspiration from sophisticated biological materials. Since chiral peptide motifs induce helix formation in macromolecules, herein we report the synthesis of a novel type of hybrid polymer consisting of a β-sheet forming a LVF [L = leucine, V = valine, and F = phenylalanine] tripeptide pendant polymethacrylate block and a poly[poly(ethylene glycol) methyl ether methacrylate] (PPEGMA) block. The designed block copolymer self-organized into helical superstructures with a left-handed twisting sense, as visualized by field emission scanning electron microscopy, transmission electron microscopy, and atomic force microscopy. This intriguing hierarchical self-assembly is driven by the minimalistic peptide motif that itself has a high propensity to adopt an antiparallel β-sheet conformation. We also report the generation of a diverse array of nanostructures, including spherical micelles, spindle micelles, rod-like micelles, vesicles, helical supramolecular fibers, and helical toroids via self-assembly of the designed block copolymer in tetrahydrofuran/water mixed solvents. To realize the observable helical superstructure, a twisted two-dimensional core–shell tape is proposed as a structure model in which the peptide segments form an antiparallel β-sheet with a polymer shell. The findings contribute to the advancement of a helical polymer or the superhelical self-assembly of polymers, paving the way for diverse applications in materials science and related fields.
