Alginate‐Boronic Acid: pH‐Triggered Bioinspired Glue for Hydrogel Assembly

Abstract The development of bioadhesives has become an emerging research field for tissue sealants, wound dressings, and hemostatic agents. However, assembling hydrogels using bioadhesive‐mediated attachment remains a challenging task. Significantly high water content (>90%) in hydrogels compared to that of biological tissues is the main cause of failure. Considering that hydrogels are primary testing scaffolds mimicking in vivo environments, developing strategies to assemble hydrogels that exhibit diverse properties is important. Self‐healing gels have been reported, but such gels often lack biocompatibility, and two gel pieces should be identical in chemistry for assembly, thus not allowing co‐existence of diverse biological environments. Herein, a mussel‐mimetic cis ‐diol‐based adhesive, alginate‐boronic acid, that exhibits pH‐responsive curing from a viscoelastic solution to soft gels is developed. Associated mechanisms are that 1) polymeric diffusion occurs at interfaces utilizing intrinsic high water content; 2) the conjugated cis ‐diols strongly interact/entangle with hydrogel chains; 3) curing processes begin by a slight increase in pH, resulting in robust attachment of diverse types of hydrogel building blocks for assembly. The findings obtained with alginate‐boronic acid glues suggest a rational design principle to attach diverse hydrogel building blocks to provide platforms mimicking in vivo environments.

Biologically Inspired Materials Exhibiting Repeatable Regeneration with Self‐Sealing Capabilities without External Stimuli or Catalysts

A new insect-cuticle- and fruit-browning-mimetic film exhibiting simultaneous self-healing and self-sealing properties only by ambient oxygen without external stimuli is developed. The film is formed at the liquid/air interface via crosslinking of phenolic compounds and poly(amine) chains. The film can be self-healed over a hundred times under ambient air at room temperature without exogenous materials and stimuli.

Galactosylated Lipidoid Nanoparticles for Delivery of Small Interfering RNA to Inhibit Hepatitis C Viral Replication In Vivo

Small interfering RNA (siRNA) delivery can provide an effective therapy for treating viral diseases by silencing genes involved in viral replication. In this study, a liver‐targeting formulation of lipidoid nanoparticle for delivery of siRNA that targets protein kinase C‐related kinase 2 (PRK2) to inhibit hepatitis C virus (HCV) replication is reported. The most effective, minimally cytotoxic lipidoid for siRNA delivery to hepatic cells is identified from a small library of alkyl epoxide–polyamine conjugates. In vitro transfection of PRK2 siRNA (siPRK2) using this lipidoid induces significant silencing of PRK2 (≈80%), suppressing HCV replication in human hepatic cells transfected with the HCV subgenomic replicon. Systemic administration of siPRK2 using the lipidoid nanoparticles results in significant reduction of host PRK2 in the mouse liver (≈60%). This treatment significantly suppresses HCV replication in an HCV‐xenograft mouse model. siRNA delivery to the liver is further improved via galactosylation of the lipidoid. Compared with the unmodified lipidoid formulation, galactosylated lipidoids induce greater silencing of host PRK2 in mouse livers (≈80%) and more rapid suppression of HCV replication in an HCV‐xenograft mouse. This study suggests that galactosylated lipidoid nanoparticles could provide a treatment for hepatitis C by mediating delivery of anti‐viral RNA interference therapeutics to the liver.

Dynamic Bonds between Boronic Acid and Alginate: Hydrogels with Stretchable, Self-Healing, Stimuli-Responsive, Remoldable, and Adhesive Properties

For the increasing demand of soft materials with wide ranges of applications, hydrogels have been developed exhibiting variety of functions (e.g., stretchable, self-healing, stimuli-responsive, and etc.). So far, add-in components such as inorganic nanoparticles, carbon materials, clays, and many others to main polymers have been used to achieve various unique functions of hydrogels. The multicomponent hydrogel systems often exhibit batch-dependent inconsistent results and problems in multicomponent mixings, require labors during preparations, and accompany unpredictable cross-talk between the added components. Here, we developed 'single polymeric component', alginate-boronic acid (alginate-BA) hydrogel to overcome the aforementioned problems. It exhibits unprecedented multifunctionalities simultaneously, such as high stretchability, self-healing, shear-thinning, pH- and glucose-sensitivities, adhesive properties, and reshaping properties. Multifunctionalities of alginate-BA hydrogel is resulted from the reversible inter- and intramolecular interactions by dynamic equilibrium of boronic acid–diol complexation and dissociation, which was proved by single molecule level Atomic Force Microscopy (AFM) pulling experiments. We also found that the alginate-BA gel showed enhanced in vivo retentions along gastrointestinal (GI) tract. Our findings suggest that rational polymer designs can result in minimizing the number of a participating component for multifunctional hydrogels, instead of increasing complexity by adding various additional components.

STAPLE: Stable Alginate Gel Prepared by Linkage Exchange from Ionic to Covalent Bonds

A new Ca2+-alginate hydrogel with improved physical stability is prepared by covalent tethering of catechols. Ionic to covalent bond transition spontaneously occurs in physiological conditions. It is found that the kinetics of leaching out of Ca2+ ions from the chelated carboxylates and oxidative intercatechol cross-linking is similar, successfully maintaining the original physical shapes without conventionally performed treatment of cationic polymers on the surface Ca2+-alginate hydrogels.

On page 9961, H. Lee and co-workers present an O2-triggered self-sealing film with the repeatable capability of regenerations and sealings inspired by the chemical principles of insect cuticle and fruit browning processes. Simple dissolution of PEI and gallol in alkaline aqueous solutions results in generation of robust films called Re-SEAL at air/water interfaces.

Sprayable Ultrafast Polydopamine Surface Modifications

Significant acceleration in the speed of polydopamine coating is achieved by optimizing various physicochemical conditions. Using these conditions, sufficient amounts of polydopamine layers are deposited onto various surfaces within a minute. Moreover, the coating process is improved by combining it with a spray system, which allows the system to be used for large-scale surface modifications.
