Stimulation of decidua development by transplantation of endometrial stem cells

On all terms of pregnancy, insolvency of decidual reaction of endometrial cells is one of the reasons of miscarriages and fetal growth delay. The insufficient decidualization of endometrum leads to infertility in such pathologies, as Asherman’s syndrome and an endometrium atrophy. However, there are data on successful application of autologous bone marrow MSCs for Asherman’s syndrome treatment. The aim of this work was to assay the effect of endometrial mesenchymal stem cell (eMSC) transplantation for decidualization process in pseudopregnant rat. Our study showed that injection of human eMSC suspension into the uterine lumen of pseudopregnant rats facilitated more intensive development of decidua in comparison with phosphate buffed saline (PBS) injection in the control uterine horn. Histological analysis of decidua sections did not reveal any alterations in cell differentiation or tissue structure. In conclusion, we demonstrated for the first time that eMSC transplantation assists the development of all decidual tissue elements. It opens the possibility that eMSCs may be applied for cell therapy of infertility associated with decidualzation insufficiency.

[Effect of bone marrow cells transplantation on the decidua formation in pseudopregnant rats].

One of the most common causes of the current pregnancy loss is the failure of the decidual reaction of endometrial cells. It is assumed that a partial source of decidual cells in endometrial tissue is bone marrow cells (BMCs). In the present work, we have studied possible effect of BMCs transplantation on the process of decidualization using the model of pseudopregnancy in rats. BMCs were flushed from the rat femurs and tibias. The obtained suspension of single BMCs was injected into one of rat uterine horns on the 5th day of pseudopregnancy. PBS without cells was injected into the contralateral horn served as the control. Rats were sacrificed on the 11th day of pseudopregnancy. Decidua formed in the experimental uterine horn showed an increase in the meso-antimezometral direction of their diameter of about 1.5-2 times as compared with a control horn. The weight of decidual tissue in the experimental horn exceeded 3 times the weight of the control one. The presence of transplanted BMCs in decidual tissue was documented by preliminary double staining of BMCs with membrane dye PKH 26 Red and nuclear dye Hoechst 33342. Histological analysis of decidua sections after transplantation revealed any alterations neither in cell differentiation nor in tissue structure. We conclude that BMCs transplantation stimulates decidualization in animals.

The decidual tissue performs numerous functions during pregnancy to provide for the progress of pregnancy and embryo development. These numerous functions initiate the participation of several cell types in the realization of decidual functioning. Decidual cells replace uterine epithelium and form a new cell barrier between the mother and the fetus. Proliferation activity of endometrial cells of front and back walls of uterus is changing different types during the implantation. The implantation of embryo into a back wall of uterus is a signal for increasing of proliferation activity of endometrial cells into the front wall of uterus. Unlike from this, the implantation of embryo into the front wall of uterus is not accompanied with increasing proliferation activity of endometrial cells of back wall of uterus. While pregnancy continue the process of placentation starts and it is accompanied with decreasing of proliferation activity of endometrial cells of a back wall of uterus.The placentation into the front wall of uterus result in high level of endometrial cells death into the front wall of uterus.

[Intracellular localization and tissue specificity of human uracil-DNA glycosylase].

Uracil-DNA glycosylase (UDG) purified by various procedures from the human placenta was used to obtain immune antisera with specific antibodies, the antibodies being affinity-purified on UDG-sepharose. Two immunoreactive polypeptides were found in crude extracts of the human placenta with the help of the antibodies. Their apparent molecular masses were about 37,000 and 34,000 dalton. Only the former polypeptide was found in crude extracts of the human embryonal heart, liver and in HeLa cells. The indirect immunofluorescent staining shows both slight and intensive fluorescence of HeLa cell nuclei. The similarity of antigenic properties of the human and rat UDG was confirmed.
