Abstract:In comparison to traditional quantitative analytical methods (HPLC and HPTLC), qNMR analysis is based upon the identification of resolved signals of the molecules of interest in the mixture rather than achieving resolution of individual components. We here present the quantitation of major sesquiterpene lactones in two important medicinal plants, Inula racemosa and Saussurea lappa using qNMR employing trimethoxybenzene (TMB) as an internal standard. Alantolactone (1) and isoalantolactone (2) were used as markers for Inula racemosa and costunolide (3) and dehydrocostuslactone (4) for Saussurea lappa. The results of qNMR analysis were found to be in corroboration with results of HPLC and HPTLC analyses. The method was validated in terms of selectivity, specificity, linearity, recovery, precision, repeatability and robustness.Keywords: qNMRalantolactoneisoalantolactonecostunolidedehydrocostuslactone

Eugenia jambolana, commonly known as 'jamun' or Indian blackberry, is an important source of bioactive compounds. All parts of the plant like stem bark, leaves, flower, fruit pulp and seeds are traditionally used for many diseases. Metabolite profiling in medicinally important plants is critical to resolve the problems associated with standardization and quality control. Metabolite profiling of the fruit pulp of Jamun was performed by NMR, HPLC, MS, GC-MS and MALDI-TOF mass spectrometry. These hyphenated techniques helped in the identification of 68 chemically-diverse metabolites of the fruit pulp. These include anthocyanins, anthocyanidins, sugars, phenolics and volatile compounds. Five extracts of fruit pulp were prepared i.e. hexane, chloroform, ethylacetate, butanol and aqueous methanolic. Twenty-five metabolites identified and quantified in the n-butanol and aqueous-methanolic extracts of ripe jamun fruit by qNMR. LC-PDA-MS and MALDI-TOF spectrometry helped in deciphering thirty-nine metabolites out of which thirteen were quantified.

Antioxidant and Antiproliferative Activities of Anthocyanin/Ellagitannin-Enriched Extracts FromSyzygium cuminiL. (Jamun, the Indian Blackberry)

Colored fruits, particularly berries, are highly chemoprotective because of their antioxidant, antiproliferative, and antiinflammatory activities. We report the cancer chemoprotective potential of Syzygium cumini L., commonly known as jamun or Indian blackberry. Anthocyanins and other polyphenolics were extracted with acidic ethanol and enriched by amberlite XAD7/HP20 (1:1). The pulp powder was found to contain 0.54% anthocyanins, 0.17% ellagic acid/ellagitannins, and 1.15% total polyphenolics. Jamun seed contained no detectable anthocyanins but had higher amounts of ellagic acid/ellagitannins (0.5%) and total polyphenolics (2.7%) than the pulp powder. Upon acid hydrolysis, the pulp extract yielded 5 anthocyanidins by HPLC: malvidin (44.4%), petunidin (24.2%), delphinidin (20.3%), cyanidin (6.6%), and peonidin (2.2%). Extracts of both jamun pulp (1,445 ± 64 μmol of trolox equivalent (TE)/g) and seeds (3,379 ± 151 μM of TE/g) showed high oxygen radical absorbance capacity. Their high antioxidant potential was also reflected by 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)- and 2,2-diphenyl-1-picrylhydrazyl-scavenging and ferrous ion-chelating activities. We also analyzed antiproliferative activity of jamun extracts against human lung cancer A549 cells. The hydrolyzed pulp and seed extracts showed significant antiproliferative activity. However, unhydrolyzed extracts showed much less activity. These data showed that in addition to 5 anthocyanidins, jamun contains appreciable amounts of ellagic acid/ellagitannins, with high antioxidant and antiproliferative activities.

Diarrhoea refers to an increase in the frequency, fand/or volume of faeces, greater than normal for an individual, resulting from an imbalan ce between the secretory and absorptive forces in the intestines. The World Health Organiza tion (WHO) estimates that 3‐5 billion cases occur each year (1billion in children less than 5 y ears of age), and that approximately 5 million deaths are due to diarrhoea annually (2.5 million i n children less than 5 years of age). The incidence of diarrhoeal diseases still remains high despite the efforts of many governments and international organisations to curb it. Many synthe tic chemicals like diphenoxylate, loperamide and antibiotics are available for the treatment of diarrhoea but they have some side effects and increasing resistance of pathogen to the common ant ibiotics . In recent years, special attention is being given on alternative safe natural bio-remedies to cure the infectious diseases becaus e of their less or no side effects, high efficacy, less cost. In this paper an attempt has been made to give an overview of certain plants with their phyto constituents and mechanism of action which have been studied for their antidiarrhoel activity.

A Revolutionary Approach for Combating Efflux Transporter-mediated Resistant Epilepsy: Advanced Drug Delivery Systems

Epilepsy is a persistent neurological condition that affects 60 million individuals globally, with recurrent spontaneous seizures affecting 80% of patients. Antiepileptic drugs (AEDs) are the main course of therapy for approximately 65% of epileptic patients, and the remaining 35% develop resistance to medication, which leads to Drug-Resistant Epilepsy (DRE). DRE continues to be an important challenge in clinical epileptology. There are several theories that attempt to explain the neurological causes of pharmacoresistance in epilepsy. The theory that has been studied the most is the transporter hypothesis. Therefore, it is believed that upregulation of multidrug efflux transporters at the blood-brain barrier (BBB), such as P-glycoprotein (P-gp), which extrudes AEDs from their target location, is the major cause, leading to pharmacoresistance in epilepsy. The most effective strategies for managing this DRE are peripheral and central inhibition of P-gp and maintaining an effective concentration of the drug in the brain parenchyma. Presently, no medicinal product that inhibits P-gp is being used in clinical practice. In this review, several innovative and promising treatment methods, including gene therapy, intracranial injections, Pgp inhibitors, nanocarriers, and precision medicine, are discussed. The primary goal of this work is to review the P-gp transporter, its substrates, and the latest novel treatment methods for the management of DRE.

CMOS Pulse Oximeter

Abstract: This paper presents the design and implementation of a non-invasive pulse oximeter based on Complementary MetalOxide-Semiconductor (CMOS) technology, aimed at enhancing the accuracy and accessibility of blood oxygen saturation measurements. Pulse oximetry is a critical tool in clinical and personal health monitoring, providing vital information about respiratory function. The integration of CMOS technology allows for miniaturization and energy efficiency, facilitating the development of compact, wearable devices capable of continuous monitoring. The proposed system utilizes advanced signal processing techniques, including lock-in detection and dual-wavelength illumination, to improve the reliability of photoplethysmographic (PPG) signal acquisition under various conditions. Experimental validation demonstrates the device's capability to deliver accurate SpO2 readings even in the presence of motion artifacts and ambient light interference. Furthermore, the calibration methods employed ensure that measurements are consistent and precise across different users. By leveraging the advantages of CMOS technology, this study not only highlights the potential for improved patient comfort and compliance but also opens new avenues for the application of pulse oximetry in diverse environments. The findings underscore the effectiveness of CMOS-based pulse oximeters in advancing non-invasive health monitoring solutions, ultimately contributing to enhanced patient care and outcomes.

Herbal drugs from natural sources like plants are in used since the traditional system of medicine; they represent a precious reservoir of innovative bioactive molecules. Among them Bidens pilosa L is one of the important plant found in all tropical and subtropical region of the world. It is the herbaceous flowering plant, having white 'petals' around a intense bunch of orange florets, and it has been reported to possess effective pharmacological properties like Antibacterial activity, Anti-inflammatory and antiallergic activity, Antimalarial Activity, T helper cell modulator, Immunosuppressive antihyperglycemic, anti-hypertensive, antiulcerogenic, hepatoprotective, anti-leukemic, anticancer, antipyretic, anti-virus, anti-angiogenic, antirheumatic, antibiotic. The Bidens pilosa L. has various chemical constituents like polyacetylenes, Polyacetylenic glycosides, aurons, auron glycosides, p-coumeric acid derivatives, caffeoylquinic acid derivatives, pheophytins, diterpenes, tannins, phytosterols, ascorbic acid, carotene, essential oils, saponins, steroids and flavonoids and many others were recognized in this plant. In this review we provide an overview mainly on the pharmacognostic characteristics, conventional uses, phytochemistry and pharmacological actions of extracts or isolated compounds from Bidens pilosa L.
