Influence of meclofenoxate hydrochloride on proliferation of human retinal pigment epithelial cells

Objective To study the effects of meclofenoxate hydrochloride on cultured human retinal pigment epithelial cells in vitro.Methods The effects of meclofenoxate hydrochloride on the proliferation of cultured hRPE cells were examined by tetrazolium salt(MTT) assay and AgNORs staining.Results Ranging from 10～1000 mg·L~ -1 ,meclofenoxate hydrochloride promoted hRPE proliferation and had a dose-dependent and time-dependent effect.AgNORs staining results showed the numbers of AgNORs in the nuclear increased when treated by meclofenoxate hydrochloride.Treated with 10 mg·L~ -1 meclofenoxate hydrochloride for 24 hours,the mean number of AgNORs was 2.0(P=0.029);and with 100 mg·L~ -1 meclofenoxate hydrochloride for 12 hours it was 2.0(P=0.029).Conclusion Meclofenoxate hydrochloride promoted human retinal pigment epithelial cells proliferation and had a dose-dependent and time-dependent effect.

Small Molecule Inhibition of Rab7 Impairs B Cell Class Switching and Plasma Cell Survival To Dampen the Autoantibody Response in Murine Lupus

IgG autoantibodies mediate pathology in systemic lupus patients and lupus-prone mice. In this study, we showed that the class-switched IgG autoantibody response in MRL/Faslpr/lpr and C57/Sle1Sle2Sle2 mice was blocked by the CID 1067700 compound, which specifically targeted Ras-related in brain 7 (Rab7), an endosome-localized small GTPase that was upregulated in activated human and mouse lupus B cells, leading to prevention of disease development and extension of lifespan. These were associated with decreased IgG-expressing B cells and plasma cells, but unchanged numbers and functions of myeloid cells and T cells. The Rab7 inhibitor suppressed T cell-dependent and T cell-independent Ab responses, but it did not affect T cell-mediated clearance of Chlamydia infection, consistent with a B cell-specific role of Rab7. Indeed, B cells and plasma cells were inherently sensitive to Rab7 gene knockout or Rab7 activity inhibition in class switching and survival, respectively, whereas proliferation/survival of B cells and generation of plasma cells were not affected. Impairment of NF-κB activation upon Rab7 inhibition, together with the rescue of B cell class switching and plasma cell survival by enforced NF-κB activation, indicated that Rab7 mediates these processes by promoting NF-κB activation, likely through signal transduction on intracellular membrane structures. Thus, a single Rab7-inhibiting small molecule can target two stages of B cell differentiation to dampen the pathogenic autoantibody response in lupus.

Activation and Denitrosylation of Procaspase-3 in KA-induced Excitotoxicity

Background: It has been reported that activation of glutamate kainate receptor subunit 2 (GluK2) subunit-containing glutamate receptors and the following Fas ligand(FasL) up-regulation, caspase-3 activation, result in delayed apoptosis-like neuronal death in hippocampus CA1 subfield after cerebral ischemia and reperfusion. Nitric oxide-mediated S-nitrosylation might inhibit the procaspase activation, whereas denitrosylation might contribute to cleavage and activation of procaspases. Objectives: The study aimed to elucidate the molecular mechanisms underlying procaspase-3 denitrosylation and activation following kainic acid (KA)-induced excitotoxicity in rat hippocampus. Methods: S-nitrosylation of procaspase-3 was detected by biotin-switch method. Activation of procaspase-3 was shown as cleavage of procaspase-3 detected by immunoblotting. FasL expression was detected by immunoblotting. Cresyl violets and TdT-mediated dUTP Nick-End Labeling (TUNEL) staining were used to detect apoptosis-like neuronal death in rat hippocampal CA1 and CA3 subfields. Results: KA led to the activation of procaspase-3 in a dose- and time-dependent manner, and the activation was inhibited by KA receptor antagonist NS102. Procaspase-3 was denitrosylated at 3 h after kainic acid administration, and the denitrosylation was reversed by SNP and GSNO. FasL ASODNs inhibited the procaspase-3 denitrosylation and activation. Moreover, thioredoxin reductase (TrxR) inhibitor auranofin prevented the denitrosylation and activation of procaspase-3 in rat hippocampal CA1 and CA3 subfields. NS102, FasL AS-ODNs, and auranofin reversed the KAinduced apoptosis and cell death in hippocampal CA1 and CA3 subfields. Conclusions: KA led to denitrosylation and activation of procaspase-3 via FasL and TrxR. Inhibition of procaspase-3 denitrosylation by auranofin, SNP, and GSNO played protective effects against KA-induced apoptosis-like neuronal death in rat hippocampal CA1 and CA3 subfields. These investigations revealed that the procaspase-3 undergoes an initial denitrosylation process before becoming activated, providing valuable insights into the underlying mechanisms and possible treatment of excitotoxicity.

Relationship between expression of P-gp170 and apoptosis in bladder transitional cell carcinoma

AIM To study the relationship between the expression of P glycoprotein (P gp170) and apoptosis in primary bladder transitional cell carcinoma. METHODS In 40 cases with primary bladder transitional cell carcinoma, we analyzed the expression of P gp170 by immunohistochemistry, and detected the apoptosis by in situ apoptotic cell labeling method of TUNEL, then calculated the Apoptosis Index (AI) by images analysis system. RESULTS Mean expression rate of P gp170 in 40 cases of bladder transitional cell carcinoma was 48%, and no significant statistic difference was found ( P 0.05). AI in higher grade tumors were significantly higher than in lower grade tumors ( P 0.05). AI in the P gp170 positive bladder transitional cell carcinoma group was significantly lower than that in the P gp170 negative group ( P 0.01). CONCLUSION P gp170 over expression in primary bladder transitional cell carcinoma may inhibit the process of cell apoptosis.

Wnt1 Positively Regulates CD36 Expression via TCF4 and PPAR-γ in Macrophages

Background: Scavenger receptors including CD36 control the phagocytosis of oxidized low-density lipoprotein and play an important role in macrophage physiology, but the underlying molecular mechanism by which CD36 is regulated in macrophages or during macrophage differentiation from monocytes remains to be determined. Methods: Here, we investigated the relationship between Wnt1 and CD36 during macrophage differentiation. CD36 was suppressed following knockdown of Wnt1 by siRNA, while it was increased by ectopic overexpression of Wnt1 in macrophages. Using a β-catenin inhibitor, peroxisome proliferator-activated receptor gamma (PPAR-γ) siRNA, and transcription factor 4 (TCF4) siRNA, we demonstrated that Wnt1 regulates the expression of CD36 through TCF4 and PPAR-γ. Co-immunoprecipitation, chromatin immunoprecipitation, and immunofluorescence experiments showed that β-catenin interacted with PPAR-γ and that PPAR-γ and TCF4 colocalized in the nucleus. Furthermore, Pax3 regulated Wnt1 via binding to the first binding site in the Wnt1 promoter. Results: Our study demonstrated that during macrophage differentiation from monocytes, Wnt1 promotes CD36 expression via activation of PPAR-γ and TCF4. Conclusions: Our findings suggest that Wnt1 plays an important role in macrophage physiology via activation of the canonical Wnt pathway.
