HYPOGLYCEMIC EFFECTS OF BARLERIA NOCTIFLORA FRACTIONS ON HIGH FAT FED WITH LOW DOSE STREPTOZOTOCIN INDUCED TYPE-2 DIABETES IN RATS

Objective: To investigate in vitro and in vivo antidiabetic activity of Barleria noctiflora (B. noctiflora) fractions on high-fat diet (HFD) with low dose Streptozotocin (STZ) induced type-2 diabetes in rats. Methods: B. noctiflora were successively extracted and then fractionated by Ethyl acetate and n. butanol. The in vitro antidiabetic activity from α-amylase and α-glucosidase was used to evaluate the potential activity of the fractions. The in vivo antidiabetic activity is evaluated against HFD/STZ induced type-2 diabetic in rats at a dose of 100 mg/kg, 200 mg/kg and 400 mg/kg, p. o for 28days. Results: Ethyl acetate fraction of B. noctiflora (EAFBN) showed the highest antidiabetic activity and IC 50 values of α-amylase inhibition (114.7±0.15) and α-glucosidase inhibition (104.93±0.28) than other fraction. In HFD/STZ (40 mg/kg) diabetic rats the EAFBN showed a dose dependent significant hypoglycemic property from body weight, blood glucose, serum lipids, serum cholesterol, serum triglycerides, urea, creatinine, hepatic enzymes and liver glycogen levels. The EAFBN significantly (P<0.01) increase the level of serum insulin. Histologically, focal necrosis was observed in the diabetic rat pancreas; however, was less obvious in treated groups. Conclusion: The EAFBN is a potent hypoglycemic agent and beneficial in reducing the elevated blood glucose level, improve the lipid profile and insulin level and histopathological changes in the pancreas of HFD/STZ‐induced non‐genetic rat model of type-2 diabetes mellitus.

In the present study was to investigated that the Overall frequency of surface epidermal and appendageal tumors and different clinical presentation such as morphology, site and their associations with skin and other systemicconditions and also find out the histopathological features of various tumors.Skinis a complex organ in which precisely regulated cellular and molecular interactions govern many crucial responses to our environment.This is a Prospective Observational Study.Totally 100 cases of appendageal tumors were studied.All the patients were reported in the out-patient department of dermatology, Rajiv Gandhi Government General Hospital, Chennai.Majority of the patient who had skin and appendageal tumors were adults.Most common age group with maximum number of tumors was 21-30 years.Most common tumor in this study was found to be Seborrhoeic keratosis(14%) and most common malignant tumor of our study was Basal cell carcinoma (7%).It was more frequently seen among females than males.In the present study we conclude that the Skin and appendageal tumors are a relatively rare group of tumors with considerable clinical and histological overlap.In our study, basal cell carcinoma was the commonest malignant tumor which is a different finding from the other studies where sweat gland tumors were common.

LPS-induced sepsis disrupts brain activity in a region- and vigilance-state specific manner

Abstract Sepsis-associated encephalopathy (SAE) is a common complication of sepsis and the systemic inflammatory response syndrome that leads to lasting consequences in survivors. It manifests as early EEG changes, that are region-, time- and state-specific, possibly reflecting distinct mechanisms of injury. Here, we investigated the effects of 5mg/kg lipopolysaccharide (LPS) on hippocampal and cortical sleep-wake states, oscillatory and non-oscillatory neuronal activity, as well as on within and between state dynamics using state-space analysis. LPS induced rapid-onset severe temporal and spatial vigilance state fragmentation, which preceded all other spectral changes by ∼90 minutes. Thereafter, LPS led to specific destabilization and increased delta oscillatory activity in wakefulness, but not NREM sleep, although state transitions remained largely normal. Instead, reduced NREM delta power resulted from aperiodic spectrum changes. LPS specifically reduced higher frequency hippocampal gamma oscillations (60-80Hz peak) in wakefulness, but not cortical high gamma or lower frequency gamma oscillations. These results suggest that disruption of sleep-wake patterns could serve as an early indicator of sepsis and associated encephalopathy, independent of spectral changes. Moreover, treatment aimed at stabilizing vigilance states in early stages of sepsis might prove to be a novel option preventing the development of further pathological neurophysiology, as well as limiting inflammation-related brain damage.

Tricyclohexylphosphate—A Unique Member in the Neutral Organophosphate Family

Abstract Tricyclohexylphosphate (TcyHP) was synthesised and characterized. The extraction of U(VI) and Th(IV) by TcyHP in n‐dodecane was studied and compared with tributylphosphate (TBP). The feasibility of the separation of Th and U with TcyHP has been explored and reported. The solubility of TcyHP in water at room temperature has been measured. The stoichiometry of the TcyHP solvates with Th(NO3)4 and UO2(NO3)2 was evaluated. The enthalpy of extraction of U(VI) by TcyHP was measured. The results of column runs with TcyHP impregnated Amberlite XAD‐7 resin for studying the loading and elution behavior of uranium from nitric acid medium are also reported in this paper. Hexavalent actinides form third phase with neutral organophosphate extractants only at very high nitric acid concentrations and high metal loading. However the presence of the three closed ring alkyl moieties in TcyHP is seen to lead to certain unique features with regard to the third phase formation behavior. It has been observed, for instance, that U(VI) forms third phase with this extractant even with highly polar diluents like 1‐chlorooctane. Further, the tendency for third phase formation with U(VI) is even higher than in the case of Th(IV), in contrast to the trend observed so far with other extractants.

ABSTRACT This paper reports the data on the extraction of U(VI), Pu(IV) and Th(IV) from nitric acid by tri-isobutyl phosphate, tri-n-amyl phosphate, tri-isoamyl phosphate and tri-n-hexyl phosphate and provides a comparison of their extract ion behaviour with that of tri-n-butyl phosphate. Data on the third phase formation in the system Th(NO3) 4 −HNO3 −1.1 M trialkyl phosphate/n-dodecane are also presented.

Abstract As part of the studies on the development of alternate extractants for fast reactor fuel reprocessing, a mixer-settler facility for performing counter-current solvent extraction runs with radioactive materials has been set up and commissioned in our laboratory for flow sheet development experiments. Mixer-settler runs were carried out with 1.1 M TiAP/Heavy Normal Paraffin (HNP) as the solvent using the above facility to test the equipment as well as the behaviour of the solvent for continuous solvent extraction processes. Stage profile data generated for the extraction and stripping of nitric acid with TiAP based solvent have been reported. Studies carried out on the continuous counter-current extraction and stripping of U(VI) for 1.1 M TiAP/HNP-U(VI)-HNO 3 system under high solvent loading conditions are also reported in this paper. Overall and stage-wise mass balance data for the above runs are also discussed.
