NATURAL ANTIOXIDANTS AS DEFENSE SYSTEM AGAINST CANCER

In living cells, the production of free radicals that comprise both reactive oxygen species (ROS) and reactive nitrogen species is highly regulated that help the cells to sustain redox homeostasis. Overproduction of ROS from mitochondrial electron transport chain leakage or excessive stimulation of xanthine oxidase and other oxidative enzymes leads to the uncontrolled production of free radicals leading to oxidative stress that can mediate damage to cell structures. This damage can be repaired by the antioxidant defense system. Antioxidants are capable of stabilizing, or deactivating, free radicals before they attack cellular components such as DNA, proteins, and lipids. The use of antioxidants in cancer prevention is a rapidly evolving research area where antioxidants scavenge free radicals and thus, indirectly help in the prevention of cancer. A wide range of antioxidants such as glutathione, N-acetylcysteine, coenzyme Q10, lycopene, flavonoids, and isoflavones when used in combination with chemotherapy and radiotherapy, result in the reduction of drug toxicity and enhanced efficacy of anticancer agents. This review aims at the use of these exogenous antioxidants as disease-oriented therapy and elucidating the relation of antioxidant enzymes with different types of cancers to overcome the harmful effects of cancer treatment.

An Overview of Synthesis and Photocatalytic Application of Carbon Quantum Dots-Based Nanocomposites

Carbon quantum dots (CQDs) is an emerging category of carbonaceous substance engaging the broad scientific community due to good chemical stability, high dispersibility, biocompatibility, facile functionalization and low toxicity. CQDs are blended with certain specific photoluminescence properties showing upconversion phenomena. In upconversion photoluminescence, CQDs emit light of a shorter wavelength than the excitation source, which is especially employed for the generation of electron-hole pairs at the neighboring photocatalysts. These phenomena, thereby, empowered large bandgap photocatalysts to be functional under a broad spectral range. Excellent light-harvesting and high charge-transfer capability facilitate CQDs in various photocatalytic applications. The photo efficiency of CQDs can be tuned by varying size, dopant and functionalization on the surface and by heterojunction construction. This chapter focuses on demonstrating a current progression in the fabrications method followed by various applications of photocatalysis, such as photodegradation, hydrogen production and CO2 photoreduction. Various techniques, like arc discharge, laser ablation, electrochemical, acid-exfoliation, ultrasonic-exfoliation, hydrothermal, microwave, pyrolytic and template method, used to synthesize CQDs are also discussed. Furthermore, photocatalytic application of CQDs-based nanohybrids with a conclusion describing direction to achieve highly efficient, sustainable and environmentally friendly CQDs to provide the current state and new vision are elaborated. We hope the literature survey successfully distributes and recapitulates the information on CQDs.

In this paper, the microwave-assisted synthesis of carboxymethyl cellulose-cl-poly(lactic acid–co-itaconic acid)/ZnO-Ag nanocomposite [CMC-cl-p(LA-co-IA)/ZnO-Ag] has been discussed. Lactic acid (LA) and itaconic acid (IA) monomers were grafted onto carboxymethyl cellulose (CMC) using potassium persulphate and N, N'-methylene-bis-acrylamide (MBA) as initiator and crosslinker, respectively at optimized conditions of temperature and pressure. The nanocomposite was characterized using different techniques such as Fourier transform infrared spectroscopy (FTIR), field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), thermogravimetric analysis (TGA) and energy dispersive X-ray (EDX). XRD, TEM and FTIR spectral analysis confirmed the formation of the nanocomposite. The release of amoxicillin drug-using nanocomposite as a function of pH and time has been investigated. The maximum drug release of 94.64% was pragmatic at pH 2.2 after 6 h. The degradation of congo red using nanocomposite followed the pseudo-first-order reaction model with the regression coefficient (R2) values of 0.99312. The nanocomposite was also explored for anticancer behavior against yeast cells.

A comparative molecular dynamic simulation study on potent ligands targeting mTOR/FRB domain for breast cancer therapy

Our study aimed to develop and find out the best drug candidate against the mechanistic target of rapamycin (mTOR/FRB) domain having a critical role in the aetiology of breast cancer. The FKBP12-rapamycin-binding (FRB) domain in the essential phosphoinositide 3 kinase/mammalian target of rapamycin (PI3K/AKT/mTOR) pathway has been a vital player in the disease progression in breast cancer. By using structure-based drug designing , the best possible targets have been identified and developed. The three-dimensional structure of the target protein was generated using I-TASSER. The ligands were generated against the most suitable target active site using standard tools for active site identification. Furthermore, the seed molecule was drawn using Chemsketch, which was then grown into the pocket using Ligbuilder. The obtained ligands were further validated using online programs for bioavailability and toxicity, followed by molecular dynamic simulations. The study concludes that the equilibrated NVT-NPT complexes indicate LIG2 stability over LIG3. RMSD and RMSF have shown that the complex of LIG2 is more stable than LIG3. LIG2 has the potential antagonistic properties to target the mTOR/FRB domain and has therapeutic implications for breast cancer.
