[Morphometry of hepatocyte mitochondrial apparatus in normal and cirrhotic rat liver].

Morphometric electron microscopy study of the hepatocyte mitochondrial apparatus and morphofunctional analysis of the degree of pathological alterations were carried out on the liver of rats with CCL4-cirrhosis (experimental group). Chronic poisoning of rats with CCL4 for 6 months led to a 4.2-fold increase in proportion of connective tissue and to a decrease in the number of hepatocytes in the liver by 21.8 %. Dry mass and ploidy of hepatocytes in the cirrhotis liver rose as compared with norm by 20.6 and 9.3%, respectively. Activities of alanine and aspartate aminotransferases in blood of rats of experimental group exceeded normal ones 2.0 and 1.4 times, respectively. Concentration of total bilirubin in blood of the cirrhotic animals increased 1.7 times, while concentration of total protein decreased by 22%. Concentration of diene conjugates in the liver of rats of experimental group increased 2.1 times as compared with normal one, while the level of malonic dialdehyde - by 34%. Activities of superoxide dismutase and catalase in the cirrhotic liver were lower than in the normal liver were lower than in the normal liver by 16 and 23 %, respectively. Morphometry of the hepatocyte mitochondrial apparatus has shown that in spite of an increase in the voluminous density of mitochondria in hepatocytes of the cirrhotic liver (by 28 %), concentration of internal mitochondrial membranes in the cells was reduced almost 1.5 times, while the total length of internal membrane in a single mitochondrion was reduced about twice as compared with norm. Thus, despite compensation of the partial loss of hepatocytes because of their polyploidization and hypertrophy, the specific synthetic activity of cells in the case of cirrhosis is decreased due to deterioration of the antioxidant system and electron transport chain of the mitochondrial apparatus.

[Morphological types of activated microglia in the hippocampus observed following transient total brain ischemia].

The aim of the study was to evaluate, by using immunohistochemistry (demonstration of Iba-1 protein), the morphological features of the rat hippocampal microglia and to describe its cellular types during the response to a single total brain ischemia of 12 min duration. The results obtained suggest the existence of several morphologic types of microgliocytes in the intact hippocampus, while some more types appear in response to ischemic lesion of neurons. These changes in general population of the hippocampal microgliocytes is related to their functional activation. During the post-ischemic period (3, 7 and 14 days following the challenge), there appeared multiple micro-outgrowths of the microglial cell processes, which suggests the enhancement of their interaction with surrounding cellular elements and may be the manifestation of the process of the elimination of neuropil lesioned synaptic structures.

MICROGLIA OF THE HUMAN SUBSTANTIA NIGRA

The structure and distribution of microglial cells was studied in the human substantia nigra (the main dopaminergic center of the brain). Microgliocytes were revealed using immunocytochemistry for the protein Iba1, the specific microglial marker. The found microgliocytes in the substantia nigra belong to the ramnified microglia, which is the most common in the adult brain. Predominant distribution of microgliocytes was revealed in the reticular part of the substantia nigra in comparison with the compact part. Atypical concentration of the protein Iba1 was found in the nucleus of the microglial cells and a variety of the intranuclear immunocytochemical reaction is described.

[Three-dimensional visualization of the brain ependyma and tanycytes].

The aim of this investigation was to develop an integrated approach to spatial reconstruction of the cells lining the ventricles of the brain using confocal laser microscopy and immunocytochemical reaction to vimentin. The work was performed on paraffin sections of rat brain of different thickness (5 and 10 microm). To visualize the immunocytochemical reaction the fluorescent dyes in the visible range were selected: SYTOX Green selectively staining the nucleus and indocarbocyanin (Cy-3) conjugated with streptavidin. As a result of testing of various processing conditions, the protocol which allows to receive an intensive staining of the structures was developed. The set of fluorochromes proposed in confocal laser microscopy allows to separate easily the channels, to study the structures independently, if needed, and does not require the use of an expensive ultraviolet laser.
