[Microplastic-Induced Alterations to Antibiotic Resistance Genes in Seawater].

The increasing and combined pollution of microplastics (MPs) and antibiotic resistance genes (ARGs) in aquatic environments is a great ecological and health concern. However, MP-induced alterations to ARGs in seawater is poorly understood, impeding risk assessment of plastics. We profiled the diversity and abundance of ARGs and mobile genetic elements (MGEs) in seawater after the addition of three different MPs (PE, PVC, and PVA) and 49-day aerated incubation.A total of 20, 35, 42, and 64 ARGs were detected in BLK, PE, PVC, and PVA, with 2, 4, 2, and 3 MGEs, respectively. The absolute abundance of ARGs in the seawater aerated with MPs ranged from 4.01×106 copies ·L-1 to 1.05×108 copies ·L-1. Additionally, the variety and richness of ARGs and MGEs in PVA were significantly higher than in the original seawater, or the seawater aerated with the other two MPs. This indicates that PVA, which is water soluble, could induce more diverse and abundant ARGs in seawater. Significant correlations among ARGs, MGEs, and 16S rRNA genes were observed, implying that the occurrence of MGEs in seawater may accelerate the transmission of ARGs through horizontal gene transfer, and bacterial microorganisms could directly affect the propagation and dissemination of ARGs.

Profiling of Antibiotic Resistance Genes in Different Croplands

Soil is the primary interface of Earth's critical zone and plays an important role in food security and sustaining environmental balance. Antibiotic resistance genes (ARGs) pose significant threat to human health and ecosystems, with croplands being intensively affected via planting patterns and the application of fertilizers. The transmission of ARGs in croplands remains largely unknown. Using high throughput quantitative Polymerase Chain Reaction (HT-qPCR) techniques, we investigated the occurrence and diversity of ARGs and their association with heavy metals in different croplands in China. A total of 187 ARGs were identified, ranging from 89 to 159 in agricultural soils. The abundance of ARGs ranged from 6.47×109 to 1.41×1010 copies·g-1 with multidrug resistance genes being the most abundant. Heavy metals including As, Co, Cr, Mo, Ni, and Pb were correlated with ARGs using the R package 'evnfit'. Furthermore, redundancy analysis (RDA) revealed that the heavy metals explained 59.3% of the variability of ARGs in the different croplands, indicating that heavy metals might exert an important influence on the composition and transmission of ARGs. Croplands soils act as a vital reservoir and reaction media for ARGs. Different crop cultivation coupled with selection pressure of heavy metals from fertilizers could have potential impacts on the prevalence, diversity, and distribution of ARGs.

Distinct patterns of soil bacterial and fungal community assemblages in subtropical forest ecosystems under warming

Climate change globally affects soil microbial community assembly across ecosystems. However, little is known about the impact of warming on the structure of soil microbial communities or underlying mechanisms that shape microbial community composition in subtropical forest ecosystems. To address this gap, we utilized natural variation in temperature via an altitudinal gradient to simulate ecosystem warming. After 6 years, microbial co-occurrence network complexity increased with warming, and changes in their taxonomic composition were asynchronous, likely due to contrasting community assembly processes. We found that while stochastic processes were drivers of bacterial community composition, warming led to a shift from stochastic to deterministic drivers in dry season. Structural equation modelling highlighted that soil temperature and water content positively influenced soil microbial communities during dry season and negatively during wet season. These results facilitate our understanding of the response of soil microbial communities to climate warming and may improve predictions of ecosystem function of soil microbes in subtropical forests.

Acidity of Soil and Water Decreases in Acid-Sensitive Forests of Tropical China

Acid deposition in China has been declining since the 2000s. While this may help mitigate acidification in forest soils and water, little is known about the recovery of soils and water from previous severe acidification in tropical China. Here, we assessed the chemistry of mineral soils, water, and acid gases (SO2 and NOx) from three successional forest types in tropical China from 2000 to 2022. Our results showed that soil pH increased synchronously from 3.9 (2000-2015) to 4.2 (2016-2022) across all three forest types, with exchangeable acid initially decreasing and thereafter stabilizing. Surface and ground water pH also gradually increased throughout the monitoring period. Soil pH recovery was stronger in the primary than in the planted forest. However, soil pH recovery lagged behind the increase in rainfall pH by approximately a decade. The recovery of soil pH was likely related to the positive effects of the dissolution of Al/Fe-hydroxysulfate mineral and subsequent sulfur desorption on soil acid-neutralizing capacity, increased soil organic matter, and climate warming, but was likely moderated by increased exchangeable aluminum and potentially proton-producing hydroxysulfate mineral dissolution that caused the lagged soil pH recovery. Surface and ground water pH recovery was attributed to increased water acid-neutralizing capacity. Our study reports the potential for the recovery of acidified soil and water following decreased acid deposition and provides new insights into the functional recovery of acid-sensitive forests.

Prevalence of Antibiotic Resistome in Ready-to-Eat Salad

Ready-to-eat salad harbors microorganisms that may carry various antibiotic resistance genes (ARGs). However, few studies have focused on the prevalence of ARGs on salad, thus underestimating the risk of ARGs transferring from salad to consumers. In this small-scale study, high-throughput quantitative PCR was used to explore the presence, prevalence and abundance of ARGs associated with serving salad sourced from two restaurant types, fast-food chain and independent casual dining. A total of 156 unique ARGs and 9 mobile genetic elements (MGEs) were detected on the salad items assessed. The abundance of ARGs and MGEs were significantly higher in independent casual dining than fast-food chain restaurants. Absolute copies of ARGs in salad were 1.34×107 to 2.71×108 and 1.90×108 to 4.87 ×108 copies per g salad in fast-food and casual dining restaurants, respectively. Proteobacteria, Bacteroidetes, Actinobacteria and Firmicutes were the dominant bacterial phyla detected from salad samples. Pseudomonas, Acinetobacter, Exiguobacterium, Weissella, Enterobacter, Leuconostoc, Pantoea, Serratia, Erwinia and Ewingella were the ten most dominant bacterial genera found in salad samples. A significant positive correlation between ARGs and MGEs was detected. These results integrate knowledge about the ARGs in ready-to-eat salad and highlight the potential impact of ARGs transfer to consumers.

Changes in Soil Infiltration and Water Flow Paths: Insights from Subtropical Forest Succession Sequence

Maintaining and improving the water conservation capacity of forest ecosystems is an important step in building regional ecological protection barrier. At present, there is a lack of knowledge about the patterns of soil water flow in different stages of forest succession. Therefore, this study quantified the change tendency of soil infiltration and water flow paths in subtropical different forest types, including pine forests (PF), mixed pine and broadleaf forests (MF) and monsoon evergreen broadleaf forests (BF), and analysed the effect of soil physical properties on hydraulic properties. Results showed that (1) both BF and MF were characterized by a smaller bulk density and the larger porosity than PF. (2) The saturated hydraulic conductivity of BF and MF was 2.1 times and 3.1 times higher than that of PF, respectively, showing that there were clear differences in the soil infiltrability resistance of the different forest types. The observed soil water exchange amount also showed that both BF and MF had higher water holding capacity than PF. (3) However, the pattern of soil water flow was found to be obvious spatial variability, restricted by the joint influence of soil porosity, root systems and rocks Dye coverage, depth, preferential fraction and other indices showed obvious differences among the three forest types, and did not evolve along succession sequence PF, MF and BF. The above results suggest that soil infiltration improve with the sequence of forest succession, but that soil water flow patterns may be independent of forest succession due to the complex conditions of the soil. This study provided effective data information of forest soil hydrological antecedents.

[Dynamics of Antibiotic Resistance Genes During the Municipal Solid Waste Leachate Treatment].

Antibiotic resistance genes (ARGs) are considered to be emerging environmental contaminants. ARGs are frequently detected in municipal solid waste landfill and its leachate. In this study, high-throughput quantitative PCR techniques were used to investigate the dynamics of ARGs during the treatment process of landfill leachate. The results showed that ARGs in leachate were diverse and abundant, and leachate treatment could effectively reduce their abundance; the absolute abundance of ARGs was reduced to 9.2×1010 copies·L-1, which was five orders of magnitude less than the abundance of the influent. However, the range and abundance of antibiotic resistance genes in the leachate effluents were still high compared to the natural environment. Therefore, the secondary contamination of ARGs might be caused by leachate being directly discharged into municipal sewage system. In this regard, ecological and environmental risk is an inconvenient truth. Furthermore, strong correlations indicate that heavy metals and mobile genetic elements (MGEs) in landfill leachate might influence the transfer of ARGs.
