Identification of Genes Causing Hybrid Necrosis in Bread Wheat

Seedling mortality due to hybrid necrosis was observed in the cross PBW 443/HUW 533. Detailed information was generated by developing BC1, BC2 and F2 generations and evaluating them in the field along with parents and F1,s to examine the genetics of necrosis. The results revealed that this particular combination has varying degree of hybrid necrosis in F1, BC1 and F2 generations but the BC2 generation showed high degree of necrosis as all the plants died before the crown root initiation (CRI) stage. The present study suggests that some seeds may be obtained by crossing even the carrier of Ne1 and Ne2 genes through bridge crosses i.e. involving a third parent which carry inhibitor gene or a gene that modifies the complementary action of Ne1 and Ne2 genes.

Significance of yield sustainability to develop climate smart wheat (Triticum aestivum) in India

To ensure food security in India, not only the wheat (Triticum aestivum L.) productivity but yield sustainability is also crucial especially when the production environments are quite diverse. An experiment was conducted to examined multi-environment wheat yield trial data of popular wheat cultivars in two agro-climatically diverse regions i.e. north- western plains zone (NWPZ) and central zone (CZ) under timely-sown (TS) and late-sown (LS) conditions with an aim to differentiate yield and yield sustainability at the level of 4 production environment, 11 prominent locations and 7 crop years. Wheat productivity matched in both zones; yet the level of yield-sustainability was much less in warmer climate of CZ. Yield sustainability was poorest in late-sown wheat of CZ and the drop in sustainability index (SI) was realized in locations, years and genotypes. For grain yield; locations and location-year interaction mattered most in each production environments but the years were crucial only in NWPZ-TS. Results showed that yield sustainability of test sites can not be adjudged by its productivity alone. Location status was associated with the yield sustainability only in NWPZ. Substantial drop in sustainability could be noticed in some crop seasons of CZ but deviations in NWPZ were smaller. In test sites, the causative factors associated with yield-sustainability varied under different production environments. Genotype-year interaction was effective in each situation except CZ-LS where only the crop year variations were supreme. Variations in locations and genotypes mattered most in NWPZ-TS whereas the crop year deviations were impactful in NWPZ-LS and CZ-TS environments. Based on the results; prospect of improvisation and breeding strategy have been suggested to select suitable production sites and the genotypes.

An innovative approach for determining composite wheat quality index to identify quality enriched genotypes - insights and implications

Abstract Ranking test entries or test sites on a quality basis is very difficult in wheat as value addition is perceived by several grain properties and end-products. Here, a novel approach has been developed and tested by deriving wheat quality index based on principal component analysis of 13 physico-chemical grain parameters and 3 end products of 45 wheat varieties. Depending upon the observed index range (0.15 to 0.71), the cultivars were assorted into 3 distinct classes i . e . elite, moderate and poor. The top group ascertained high quality standards of grain suited for bread and chapati whereas bottom group assured better cookies quality. This technique was also tested to differentiate quality enriched test sites within a zone or demarcate the most suited production environments to harness good quality wheat. The index will have an implication on farmers (premium price for varietal segregation), industry (product specific quality cultivars), and consumers (superior quality products).

Elucidating the Population Structure and Genetic Diversity of Indian Puccinia striiformis f. sp. tritici Pathotypes Based on Microsatellite Markers

In India, systematic wheat yellow rust survey and pathotype (race) analysis work began in 1930. However, information on population structure and genetic diversity of yellow rust pathogen has not been available. To address this, we conducted studies on population structure and genetic diversity of Puccinia striiformis f. sp. tritici (Pst) pathotypes using 38 simple sequence repeat primer-pairs. Bayesian assignment and discriminant analysis of principal components indicated the presence of two distinct Pst subpopulations (Pop1 and Pop2) along with 37.9% admixed pathotypes. The unweighted pair-group method with arithmetic mean also categorized these pathotypes into two major clusters. Principal coordinates analysis explained 20.06 and 12.50% variance in horizontal and vertical coordinates, respectively. Index of association (IA) and the standardized index of association ([Formula: see text]) values showed that Pst subpopulations reproduced asexually (clonally). In total, 102 alleles were detected, with the expected heterozygosity (Hexp) per locus ranging from 0.13 to 0.73, with a mean of 0.47. The average polymorphic information content value of 0.40 indicated high genetic diversity among pathotypes. Analysis of molecular variance revealed 12% of the total variance between subpopulations, 11% among the pathotypes of each subpopulation, and 77% within pathotypes. A significant moderate level of genetic differentiation (FST = 0.122, P < 0.001) and gene flow (Nm = 1.80) were observed between subpopulations. The Pst virulence phenotypes showed a weak positive correlation (R2 = 0.027, P < 0.02) with molecular genotypes.

Multi-environment analysis of nutritional and grain quality traits in relation to grain yield under drought and terminal heat stress in wheat (Triticum aestivum L.)

Abstract Heat and drought stress on wheat genotypes are examined in the study, emphasizing the need to improve grain yield, nutrition, and quality. Sixteen modern wheat cultivars were tested in nine environments, including normal, heat, and drought, in three peninsular Indian locations. Genotype performance in yield, nutrition, and quality parameters is assessed using multi-environment trials like AMMI and GGE Biplot. We discovered intriguing stress dynamics in Zn and Fe. Under heat stress, zinc yield increases but decreases under drought stress, while iron does the opposite. Selecting zinc, starch, and kernel weight under stress can boost yield. Protein content and yield are inversely related, making it difficult for breeders to optimize both traits. G x E interactions and stability indices have found genotypes like G12 (MP 1358) and G5 (HI 1544) with high grain yield and iron content, making them crop improvement sources. G12 (MP 1358) has high protein content. Also notable are zinc-rich G11 (MACS 4058) and G15 (WH 730). Genotype G12 (MP 1358) improved crop yield, nutritional quality, and stability "win-win". This study shows the complexity of nutrient metabolism under different stress conditions and suggests targeted breeding to improve wheat grain quality and micronutrient profiles without yield loss.

Identification of physiological traits at seedling stage associated with salt tolerance in wheat variety KH 65 using RILs

Salinity is a major stress affecting production and productivity of wheat across the world. Understanding physiological traits associated with salt tolerance can help in breeding for improving wheat under salt stress. In the present investigation, physiological traits in 3rd leaf at seedling stage and grain yield at maturity were studied in a population of RILs derived from a cross between salt tolerant (Kharchia 65) and susceptible (HD 2009) cultivars under control (pH8.2) and sodic condition (pH 9.2) in microplots. Though HD 2009 had higher yield under control conditions, it exhibited a higher reduction in yield (44.7%) under sodic stress as compared to KH 65 (9.8%). There was a significantly lower accumulation of Na+ content and higher accumulation of K+, proline and chlorophyll content in the 3rd leaf of KH 65 as compared to HD 2009 under sodic stress. Na+ content exhibited significant negative correlation (P<0.01) while K+/Na+ ratio, proline content, and chlorophyll content showed significant positive correlation (P<0.01) with grain yield (GY) under sodic condition. First and second principal component analysis (PCA) explained the total variation of66.43% (PCI 50.18 % and PCII 16.25%) among different traits under sodic conditions. Na+ content made an independent group with strong negative correlation with grain yield and K+/Na+ ratio while proline content and thousand grain weight (TGW) were grouped together along with GY. The study demonstrated that low Na+ concentration and high K+/Na+ ratio, proline and chlorophyll content at the seedlingÂ stage are important physiological traits contributing towards yield under sodic stress. Identification and development of novel salt-responsive candidate gene based SSRs (cg-SSRs) and MIR gene based SSRs (mir-SSRs) in bread wheat (Triticum aestivum)

Abstract Salt stress adversely affects the global wheat production and productivity. To improve salinity tolerance of crops, identification of robust molecular markers is highly imperative for development of salt-tolerant cultivars to mimic yield losses under saline conditions. In this study, we mined 171 salt-responsive genes (including 10 miRNAs) from bread wheat genome using the sequence information of functionally validated salt-responsive rice genes. Salt-stress, tissue and developmental stage-specific expression analysis of RNA-seq datasets revealed the constitutive as well as the inductive response of salt-responsive genes in different tissues of wheat. Fifty-four genotypes were phenotyped for salt stress tolerance. The stress tolerance index of the genotypes ranged from 0.30 to 3.18. In order to understand the genetic diversity, candidate gene based SSRs (cg-SSRs) and MIR gene based SSRs (miR-SSRs) were mined from 171 members of salt-responsive genes of wheat and validated among the contrasting panels of 54 tolerant as well as susceptible wheat genotypes. Among 53 SSR markers screened, 10 cg-SSRs and 8 miR-SSRs were found to be polymorphic. Polymorphic information content between the wheat genotypes ranged from 0.07 to 0.67, indicating the extant of wide genetic variation among the salt tolerant and susceptible genotypes at the DNA level. The genetic diversity analysis based on the allelic data grouped the wheat genotypes into three separate clusters of which single group encompassing most of the salt susceptible genotypes and two of them containing salt tolerance and moderately salt tolerance wheat genotypes were in congruence with penotypic data. Our study showed that both salt-responsive genes and miRNAs based SSRs were more diverse and can be effectively used for diversity analysis. This study reports the first extensive survey on genome-wide analysis, identification, development and validation of salt-responsive cg-SSRs and miR-SSRs in wheat. The information generated in the present study on genetic divergence among genotypes having a differential response to salt will help in the selection of suitable lines as parents for developing salt tolerant cultivars in wheat.
