Identification of novel sweet protein for nutritional applications

The prevalence of obesity and diabetes has increased exponentially in recent years around the globe, especially in India.Sweet proteins have the potential to substitute the sugars, by acting as natural, good and low calorie sweeteners.They also do not trigger a demand for insulin in diabetic patients unlike sucrose.In humans, the sweet taste perception is mainly due to taste-specific G protein-coupled heterodimeric receptors T1R2-T1R3.These receptors recognize diverse natural and synthetic sweeteners such as monelin, brazzein, thaumatin, curculin, mabinlin, miraculin and pentadin.Structural modeling of new sweetener proteins will be a great leap in further advancement of knowledge and their utility as sweeteners.We have explored the fingerprints of sweetness by studying the aminoacid composition and structure properties of the above proteins.The structural analysis of monellin revealed that the individual A or B chains of monellin are not contributing to its sweetness.However, the native conformation and ionic interaction between AspB7 of monellin with active site of T1R2-T1R3 receptor, along with hydrogen bonding stability of IleB6 and IleB8 are responsible for the sweet taste.Based on structural similarity search, we found a new hypothetical protein from Shewanella loihica, which has the presence of Asp32 with adjacent isoleucine residues.Further, we examined the lead protein by two-step docking for the study of interaction of functionally conserved residues with receptors.The identified protein showed similar ionic and hydrophobic interactions with monelin.This gives a promising opportunity to explore this protein for potential health application in the low calorie sweetener industry viz., soft drinks, snacks, food, chocolate industries etc.

Harnessing Machine Learning for Uterine Fibroid Detection Using Advanced Classification Techniques

In the 40 to 50 age range, uterine fibroids are prevalent in over 70% of women. The proposed system employs a dataset comprising 1985 ultrasound images of uterus for fibroid detection. Utilizing advanced classification techniques like ResNet-152 and MobileNet V2, and an ensemble of Logistic Regression and MobileNet V2, the proposed system attained remarkable precision at 87.23% and an impressive accuracy of 91.67%. This outperformed the previous HIFU-Net Segmentation method's precision of 84.48% on MRI scans, highlighting the potential of advanced techniques in medical diagnostics.

Reproducible and stable surgical hypertension using a smaller renal artery clip model in Wistar rats

Animal models of hypertension are crucial for studying the pathophysiology of diseases and developing new treatments. This study introduces a reproducible technique for inducing surgical renovascular hypertension in rats using a smaller renal artery clip. The surgical groups were compared to the control group and standard drug group to assess the effectiveness of the surgical model and its stability for 21 days. The results revealed a progressive increase in systolic and diastolic blood pressure in the surgical group. Moreover, the heart rate and body weight in the renal hypertension group demonstrated a sustained elevation at the end of 21 days. These findings confirm the efficacy of this surgical model in inducing hypertension. Effects of fermentatively recovered fish waste lipids on the growth and composition of broiler meat

1. The aim of the present study was to evaluate the effects of feeding fermentatively recovered fish oil (FFO) from fish processing waste (FPW), on the performance and carcass composition of broilers. A total of 60 one-d-old VenCobb broiler chicks randomly assigned to 5 treatment groups were studied. 2. The birds were randomly assigned to treatment groups and fed with a normal commercial diet (control, T1), a diet with 2% groundnut oil (positive control, T2), a diet with 1% FFO (T3), a diet with 1.5% FFO (T4) and a diet with 2% FFO (T5). Performance and growth parameters (feed intake and body weight) and fatty acid composition of serum, liver and meat were determined. 3. The performance characteristics of broiler meat did not differ among treatments. Feeding FFO reduced total cholesterol concentration in serum, meat and liver of the FFO-fed groups (T3 to T5) as compared to both the controls (T1 and T2), but there was no significant difference in triglyceride concentration between treatments. Increased concentrations of EPA and DHA in serum, liver and meat of FFO-fed groups, as compared to both controls, were observed as the FFO concentration increased. 4. The study clearly demonstrates the value of oil recovered from FPW in addition to addressing the environmental issues related to disposal of such biological waste.

Phytochemical and Functional Characterization of Different Parts of the Garcinia xanthochymus Fruit

The fruit of Garcinia xanthochymus is consumed traditionally and is known to possess health-promoting effects. However, studies involving the characterization of phytochemicals of different parts of the fruit, and their biological activity were limited and hence warranted a comprehensive study. The proximate analyses reveal that fruit peel was rich in crude fiber. The levels of essential minerals, fatty acids, amino acids, carotenoids, organic acids, and polyphenols were significantly higher in the peel, followed by the rind, seed, and pulp. The in vitro antioxidant assays revealed that the polyphenolic extract of the peel possesses a high antioxidant effect compared to the extracts from other parts of theG. xanthochymus fruit. Furthermore, the in vitro assays reveal the antidiabetic potential of the methanol extract. This is the first comprehensive report involving the characterization and biological properties of different parts of the G. xanthochymus fruit. Hence, our study implicates the potential use of this fruit for the development of functional foods for diabetes.

Effect of Silymarin and Quercetin in a Miniaturized Scaffold in Wistar Rats against Non-alcoholic Fatty Liver Disease

Silymarin and quercetin (SQ) are known antioxidants with substantial free radical scavenging activities. The efficacy of SQ activity is restricted due to poor absorption and availability. This study aims to increase the hepatoprotective activity of SQ by a newer delivery technique. We have optimized a technique, miniaturized scaffold (MS), for the delivery of active compounds of SQ. SQ molecules were embedded in MS and characterized by morphology, particle size, miniaturization efficiency, and functional group. Further, the hepatoprotective effects of MSQ were investigated through in vitro and in vivo methods. Hepatotoxicity was induced in rats by carbon tetrachloride (CCl4), and subsequently, hepatotoxic rats were treated with the miniaturized scaffold of SQ (MSQ) for 8 weeks. The body weight were significantly high in groups fed with MSQ. A substantial decrease in triglyceride, total cholesterol, low-density lipoprotein, alanine aminotransferase, and aspartate aminotransferase activities were observed in rats treated with MSQ. Similarly, rats treated with MSQ exhibited lower lipid accumulation in the hepatocytes. The experiments clearly demonstrated the efficacy of MSQ as a superior hepatoprotective agent against non-alcoholic fatty liver disease simulated through toxicity induced by CCl4.
