Development and identification of transgenic rapeseed with a novel gene for glyphosate resistance

Glyphosate is the most widely used broad-spectrum herbicide in the world. However, at present there is no glyphosate-tolerant rapeseed variety with independent intellectual property rights in China. In the study, a novel glyphosate-resistant genes I. variabilis EPSPS was transferred to the Brassica napus pure line J9707 via the Agrobacterium tumefaciens-mediated hypocotyl method, and 126 T₀-positive transgenic plants with 97.0% positive rate were generated. The T-DNA insertion with a single copy (44.8%) is dominant. The insertion locations of T-DNA in the lines of EPS-2, EPS-6, and EPS-7 were identified by inverse PCR method. The stability of the T-DNA insertion in these lines were further confirmed by insertion-specific PCR in their T₀ to T₃ plants. The gene expression analysis revealed that the I. variabilis EPSPS gene and its protein was stably expressed in different generations of transgenic lines in RNA and protein levels. Treatments with different doses of glyphosate indicated that the lines of EPS-1, EPS-2, EPS-5, EPS-6, and EPS-7 could tolerate four times of the recommended dose of glyphosate in production. Thus, the novel glyphosate-tolerant rapeseed lines generated in the present study will lay the foundation for the herbicide- tolerance rapeseed breeding in China.

Driver Attribute Filling for Genes in Interaction Network via Modularity Subspace-Based Concept Learning from Small Samples

The aberrations of a gene can influence it and the functions of its neighbour genes in gene interaction network, leading to the development of carcinogenesis of normal cells. In consideration of gene interaction network as a complex network, previous studies have made efforts on the driver attribute filling of genes via network properties of nodes and network propagation of mutations. However, there are still obstacles from problems of small size of cancer samples and the existence of drivers without property of network neighbours, limiting the discovery of cancer driver genes. To address these obstacles, we propose an efficient modularity subspace based concept learning model. Our model can overcome the curse of dimensionality due to small samples via dimension reduction in the task of attribute concept learning and explore the features of genes through modularity subspace beyond the network neighbours. The evaluation analysis also demonstrates the superiority of our model in the task of driver attribute filling on two gene interaction networks. Generally, our model shows a promising prospect in the application of interaction network analysis of tumorigenesis.

Fuzzy Active Contour Model With Markov Random Field for Change Detection

The traditional active contour models are sensitive to the speckle noise in the synthetic aperture radar (SAR) images. In this paper, the Markov random field (MRF) theory is incorporated into the fuzzy active contour model to detect the changes of multitemporal SAR images. In the proposed method, neighboring information is considered to modify the pointwise prior probability for exploiting the mutual and spatial information. In addition, we incorporate MRF into the fuzzy active contour model and get the resulting MRF-based energy function. Finally, we drive the associated first variation of the energy function to compute the fuzzy membership. Due to the introduction of MRF, the proposed MRF-based fuzzy active contour model is robust to the speckle noise in the SAR images and can achieve accurate change detection results. Experiments on four SAR image datasets demonstrate that the proposed MRF-based fuzzy active contour model is able to accurately segment the difference image and has better performance in comparison with other change detection techniques.

Federated Learning for Supply Chain Demand Forecasting

With the country’s policy support and the rapid development of Internet technology, the domestic consumption level has been escalating and the consumption structure has changed. The traditional retail industry cannot integrate all the relevant data due to data security and privacy protection concerns so that it is unable to adjust sales strategies in an accurate and timely manner. New retail has sounded the clarion call for the retail revolution. The supply chain demand forecasting is an important problem for the supply chain management. In this research, we propose a new retail supply chain commodity demand forecasting framework based on vertical federal learning, which solves the problems of data security and privacy faced by new retail theoretically and empirically. In experiments, we use datasets from different platforms (such as social platforms, e-commerce platforms, and retailers) in the same region for federated learning. The experiment results demonstrate the superiority of the proposed algorithm.

Experimental study on performance evaluation of fluorescence diffuse optical tomography system

Fluorescence diffuse optical tomography (FDOT) is an emerging optical imaging tool for in-vivo observation of organisms and small-animal. This modality retrieves the distribution of the concentration of the fluorescent biomarkers noninvasively and quantitatively, which is related to the interior cellular and molecular events. After the FDOT system is established, its performance parameters should be comprehensively and systematically evaluated. This paper conducts an experimental study on the parameters such as spatial resolution and sensitivity of our FDOT system. The fluorescent reagent used in the experiment was indocyanine green, and the phantom material was polyformaldehyde.

Scrub typhus is an acute infectious disease caused by Orientia tsutsugamushi that is transmitted primarily through the bite of infected chigger mite larvae. The disease prevalence is closely associated with environments characterized by high moisture levels and abundant vegetation. This study documents the first confirmed case of scrub typhus in Ningxia Hui Autonomous Region, China. The patient presented with characteristic clinical manifestations, including abnormal biochemical indicators and positive serum-specific IgM antibodies against scrub typhus. Epidemiological evidence suggests local acquisition of the disease. Enhanced surveillance and preventive measures for scrub typhus are essential in this region, particularly for individuals residing in or visiting areas where the disease may be endemic.

The Complex Genomic Diversity of <i>Yersinia pestis</i> on the Long-Term Plague Foci in Qinghai-Tibet Plateau

Yersinia pestis , the bacterial causative agent of plague, remains an important threat to human health. Here we conducted genomic population and phylogenetic analyses using the nucleotide polymorphisms (SNPs) of 626 global whole-genome sequencing data to reconstruct the evolutionary history and correlation between the genetic groups and plague foci. The phylogeographic diversity of Y. pestis in different kinds plague foci in China showed host niches adaptation. Most natural plague foci strains are region-and focus-specific, with one main cluster except for that of the Marmota himalayana plague focus of the Qinghai-Tibet plateau (MHPF). The strains from MHPF scattered into 10 lineages, with several isolates scattered in multiple populations over ancestral branch. The average phylogenetic diversity among isolates was greater in MHPF than other plague foci. It has the largest pan-genome and widest SNP distances with most accessory genes enriched in mobilome functions (prophages, transposons).
