HLA-mismatched allogeneic adoptive immune therapy in patients with severely immunosuppressed AIDS: a multicenter, open-label, controlled, phase 2a study

Recurrent opportunistic infections (OIs) in patients with severely immunosuppressed AIDS remain an unresolved medical challenge despite advancements in antiretroviral therapy (ART). To address this gap, we developed an HLA-mismatched allogeneic adoptive immune therapy (AAIT) specifically targeting this patient population. The safety and efficacy of this novel therapeutic approach were preliminarily confirmed in our phase 1 trial. Subsequently, a multicenter, open-label, controlled, phase 2a trial was conducted to evaluate the efficacy of AAIT in combination with ART compared with the conventional ART-only regimen. CRF07_BC is associated with slow HIV disease progression in Chinese patients

HIV subtypes convey important epidemiological information and possibly influence the rate of disease progression. In this study, HIV disease progression in patients infected with CRF01_AE, CRF07_BC, and subtype B was compared in the largest HIV molecular epidemiology study ever done in China. A national data set of HIV pol sequences was assembled by pooling sequences from public databases and the Beijing HIV laboratory network. Logistic regression was used to assess factors associated with the risk of AIDS at diagnosis ([AIDSAD], defined as a CD4 count < 200 cells/µL) in patients with HIV subtype B, CRF01_AE, and CRF07_BC. Of the 20,663 sequences, 9,156 (44.3%) were CRF01_AE. CRF07_BC was responsible for 28.3% of infections, followed by B (13.9%). In multivariable analysis, the risk of AIDSAD differed significantly according to HIV subtype (OR for CRF07_BC vs. B: 0.46, 95% CI 0.39─0.53), age (OR for ≥ 65 years vs. < 18 years: 4.3 95% CI 1.81─11.8), and transmission risk groups (OR for men who have sex with men vs. heterosexuals: 0.67 95% CI 0.6─0.75). These findings suggest that HIV diversity in China is constantly evolving and gaining in complexity. CRF07_BC is less pathogenic than subtype B, while CRF01_AE is as pathogenic as B.

福州市闽江下游桔园洲防洪堤由于堤型选用反滤砂堤,即用无粘性土砂作为堤身建筑材料,无任何防渗防水处理,其工程实例国内十分少见,以砂筑堤,它具有就地取材,填筑简便,造价较低的优势,但砂毕竟是一种松散的建材,不仅自身的抗渗、抗冲能力很低,也可能存在地段液化问题.因此,如何控制填筑质量和怎样做好防冲防渗,历来都是一个颇具争议的问题.根据桔园洲这条全省最长的砂堤工程实践,采用新型反滤砂堤施工技术,经多年的现场监控,堤身也未发现严重的变形渗漏.因而,笔者认为填筑质量是好的,该方法是可行的,可为类似工程提供借鉴.

Evaporated Perovskite Thick Junctions for X-Ray Detection

X-ray detection is widely utilized in our daily life, such as in medical diagnosis, security checking, and environmental monitoring. However, most of the commercial X-ray detectors are based on inorganic semiconductors, e.g., Si, CdTe, and Ge, which require complex and costly fabrication processes. Metal halide perovskites have recently emerged as a set of promising candidates for ionizing radiation detection, owing to the high attenuation coefficient, long carrier lifetime, and excellent charge transport properties. Perovskite single crystals have been successfully implemented in X-ray detection, but the fragile single crystals limit the device fabrication and the integration with a read-out circuit. In addition, it is hard to reach inch-size single crystals for real application. Flexible devices based on perovskite films or composite films have also been reported, but either the thickness or charge transport properties are limited by the solution processes. In this work, we introduced thermal co-evaporation to deposit highly efficient formamidinium lead iodide perovskite films. Considering the trade-off between X-ray absorption and charge transport, we optimized the active layer thickness and achieved large-area and flexible X-ray detectors with state-of-the-art device performance, including extremely low dark current and noise, fast response, and high sensitivity of 142.1 μC Gyair–1 cm–2.

Efficient Algorithm for Constructing Order K Voronoi Diagrams in Road Networks

The order k Voronoi diagram (OkVD) is an effective geometric construction to partition the geographical space into a set of Voronoi regions such that all locations within a Voronoi region share the same k nearest points of interest (POIs). Despite the broad applications of OkVD in various geographical analysis, few efficient algorithms have been proposed to construct OkVD in real road networks. This study proposes a novel algorithm consisting of two stages. In the first stage, a new one-to-all k shortest path finding procedure is proposed to efficiently determine the shortest paths to k nearest POIs for each node. In the second stage, a new recursive procedure is introduced to effectively divide boundary links within different Voronoi regions using the hierarchical tessellation property of the OkVD. To demonstrate the applicability of the proposed OkVD construction algorithm, a case study of place-based accessibility evaluation is carried out. Computational experiments are also conducted on five real road networks with different sizes, and results show that the proposed OkVD algorithm performed significantly better than state-of-the-art algorithms.
